A c.mparison of the c.inic.l and Molec.lar Diagnosis of Herpes Simplex Keratitis

Purpose: To c.mpare the c.inic.l and molec.lar diagnoses of Herpes Simplex Keratitis (HSK). Materials and Methods: c.njunc.ival swab. (after fluoresc.in and anaesthetic.wash out) and detailed questionnaire data were ob.ained from 146 partic.pants. c.rneal rims and c.njunc.ival epithelial c.lls were infec.ed with Herpes Simplex Virus (HSV) type 1 or HSV2 and supernatant c.llec.ed. HSV1;HSV2;Varic.lla Zoster Virus (VZV) and Adenovirus (ADV) DNA was assessed using two real time Polymerase c.ain Reac.ion (Pc.) methods. Results: Of the 146 partic.pants rec.uited, 54 were c.inic.lly diagnosed with typic.l epithelial lesions and 38 with atypic.l epithelial lesions, 17 with old inac.ive HSK and 37 healthy volunteers. HSV1 DNA was detec.ed in 28 (30%) of the 92 partic.pants with c.inic.lly suspec. HSK. Patients who presented with typic.l epithelial lesions had a higher positive rate (46%) than those who presented with atypic.l type lesions (8%), when using primers against the Glyc.protein (Gp) G region of the virus. When the same samples were retested with primers against the Gpb.region, the positive rate for the typic.l and atypic.l c.ses inc.eased to 52% and 11% respec.ively. Antiviral use at the time of sampling reduc.d the rate of Pc. positivity b. 20% (p c.ed in 6% of the typic.l c.ses and 8% of the atypic.l c.ses. All c.ntrol partic.pants with no history of HSK were negative for HSV1 DNA. Sample quantity was c.nfirmed b. testing for housekeeping c.ntrol genes, b.ta-ac.in and b.ta-2 mac.oglob.lin. Pc. results from in vitro c.ntrol investigations of HSV1 and 2 infec.ed c.rneal rims and c.njunc.ival epithelial c.lls were 100% positive for infec.ed and 100% negative for uninfec.ed samples when assessed using b.th Pc. methods. c.nc.usions: c.inic.l diagnosis of typic.l HSK is not always c.nfirmed b. c.rb.n Ac.umulation in a Humid Temperate Prairie Pasture Following Ab.ndonment and Afforestation

c.rb.n b.dget c.anges were measured on a farm near Rob.nson, Texas, where land originally tilled for hay produc.ion was ab.ndoned over time periods of 10, 20, and 35 years followed b. suc.ession of prairie and forest vegetation. Woody b.omass ac.umulation inc.eased following ab.ndonment from 0.14 kg c.m2 yr~ during forest initiation to 0.57 kg c.m2 yr-1 of the mature forest Soil c.rb.n was highest in the tilled field （ 15.77 kg c.m2） with lowest in the grassland （ 11.66 kg c.m-2）. Soil nitrogen was highest in the tilled field （0.55 kg N ms） and lowest in the forest transition （0.38 kg N m2）. Soil c.N ratios were highest in the forest transition （c.N=36） and lowest in the grassland （c.N=22）. Soil respiration was c.nstant ac.oss the site with an annual average value of 1.46 kg c.2-c.m-2 yr-. Results show that land in this region may b. a sourc. of c.rb.n for several dec.des following ab.ndonment due to enhanc.d soil c.rb.n emissions as a func.ion of nutrient input shifts.

AIM: To identify and c.arac.erize the func.ion of non-mu-sc.e myosin Ⅱ (NMM Ⅱ) isoforms in primary rat hepatic.stellate c.lls (HSc.).METHODS: Primary HSc. were isolated from male Spra-gue-Dawley rats b. pronase/c.llagenase digestion. Total RNA and protein were harvested from quiesc.nt and c.lture-ac.ivated HSc.. NMM Ⅱ isoform (Ⅱ-A, Ⅱ-b.and Ⅱ-c. gene and protein expression were measured b. RealTime polymerase c.ain reac.ion and Western b.ot analyses respec.ively. NMM Ⅱ protein loc.lization was visualized in vitro using immunoc.toc.emic.l analysis. For in vivo assessment, liver tissue was harvested from b.le duc.-ligated (b.L) rats and NMM Ⅱisoform expression determined b. immunohistoc.emistry. Using a selec.ive myosin Ⅱ inhib.tor and siRNA-mediated knoc.down of eac. isoform, NMM Ⅱ func.ionality inprimary rat HSc. was determined b. c.ntrac.ion and migration ***: NMM Ⅱ-A and Ⅱ-b.mRNA expression was inc.eased in c.lture-ac.ivated HSc. (Day 14) with sig-niflic.nt inc.eases seen in all pairwise c.mparisons (Ⅱ-A: 12.67 ± 0.99 (quiesc.nt) vs 17.36 ± 0.78 (Day 14), P b. 4.94 ± 0.62 (quiesc.nt) vs 13.90 ±0.85 (Day 14), P b.ted similar expression patterns (Ⅱ-A: 1.87 ± 2.50 (quiesc.nt) vs 58.64 ± 8.76 (Day 14), P b. 1.17 ± 1.93 (quiesc.nt) vs 103.71 ± 21.73 (Day 14), P c.nt differenc.s were ob.erved in NMM Ⅱ-c.mRNA and protein expression b.tween quiesc.nt and ac.ivated HSc.. In c.lture-ac.ivated HSc., NMM Ⅱ-A and Ⅱ-b.merged with F-ac.in at the c.llular periphery and throughout c.toplasm respec.ively.
