Bioinformatic approach for understanding the heterogeneity of cholangiocytes

It is remarkable that microarray technologies have nearly reached a pinnacle. Establishment of further analysis and management of enormous data derived from microarray technology is currently the highest priority. The heterogeneous functions of cholangiocytes regulate the pathophysiology of the biliary epithelium in relation to secretory, proliferative and apoptotic activities. Distinct expression profiles of two murine cholangiocyte lines, termed small and large have been revealed by microarray analysis. The features of the two cholangiocyte cell lines, categorized partly according to gene ontology, indicate the specific physiological role of each cell line. The large cholangiocytes are characterized as “transport” and “immune/ inflammatory responses” In contrast, small cholangiocytes are associated with properties of limited physiological functional ability and proliferating/migrating potential with specific molecules like Eph receptors, comparable to mesenchymal cells. ＇Omic study will be of great help in understanding the heterogeniety of cholangiocytes.

We use a transient-grating （TG） process in a Kerr bulk medium to clean a femtosecond laser pulse. Using the technique, the temporal contrast of the generated TG signal is improved by more than two orders of magnitude in comparison with the incident pulse in a 0.5-mm-thick fused silica plate. The laser spectrum is smoothed and broadened, and the pulse duration is shortened simultaneously. We expect to extend this technique to a clean pulse with broadband spectral bandwidth at a wide spectral range because it is a phase-matched process.

The automated stopping of a spectral measurement with active learning is *** optimal stopping of the measurement is realised with a stopping criterion based on the upper bound of the posterior average of the generalisation error of the Gaussian process *** is revealed that the automated stopping criterion of the spectral measurement gives an approximated X-ray absorption spectrum with sufficient accuracy and reduced data *** proposed method is not only a proof-of-concept of the optimal stopping problem in active learning but also the key to enhancing the efficiency of spectral measurements for highthroughput experiments in the era of materials informatics.

Climate change can intensify drought in many regions of the world and lead to more frequent drought events or altered cycles of soil water ***,it is important to enhance the tolerance to drought and thus health,vigor,and success of transplantation seedlings used in the forestry by modifying fertilization and promoting ***,we sowed seeds of Japanese larch(Larix kaempferi)in 0.2-L containers with 0.5 g(low fertilization;LF)or 2 g(high fertilization;HF)of slow-release fertilizer early in the growing *** month later,we irrigated seedlings with non-sterilized ectomycorrhizal inoculum(ECM)or sterilized solution(non-ECM),and after about 2 months,plants were either kept well watered(WW;500 mL water/plant/week)or subjected to drought(DR;50 mL water per plant/week)until the end of the growing *** largely stimulated plant growth and above-and belowground biomass production,eff ects that are of practical signifi cance,but caused a small decrease in stomatal conductance(Gs 390)and transpiration rate(E 390),which in practice is insignifi *** treatment resulted in moderate inhibition of seedling growth and biomass and largely canceled out the enhancement of biomass and foliar K content by *** caused a large decrease in CO 2 assimilation,and enhanced stomatal closure and induced *** also largely depleted foliar Mg and enriched foliar *** DR caused a large decrease in foliar P content in LF,it moderately increased P in ***,DR increased foliar K in HF but not in LF,and decreased foliar P in ECM plants but not in non-ECM ***,ECM plants exhibited a large enrichment in foliar P under WW and had a lower water potential under DR when grown in *** results indicate that the drought tolerance and health and vigor of Japanese larch seedlings can be modifi ed by soil fertility and soil *** study provides a basis for new multifactorial research programs aimed at producing seedlings of su

Relaxation of electronic excited-state and vibrational dynamics in the electronic excited state in Ru II(TPP)(CO) [TPP=tetraphenylporphyrin] dissolved in N2 bubbled benzene,O2 bubbled benzene,N2 bubbled chloroform and N2 bubbled acetone were clarified using a sub-5fs laser pulse. The result showed that the transition energy from the 1Q x states,consisting of the 1Q x(1,0)(π,π*) state(existing at 18800 cm-1 ) and 1Q x(0,0)(π,π*) state(existing at 17800 cm-1 ),to the 1M state of six-coordinated ruthenium complex is lower than that of the five-coordinated ruthenium complex. The intersystem crossing 1Q x(0,0)(π,π*)→ 3(d,π*) in the five-coordinate Ru II(TPP)(CO) in the solution was found to take place in about 1 ps.

AIM:To evaluate the protective properties of novel prostone ClC-2 agonist SPI-8811 in porcine model of gastric acid injury. METHODS:Porcine gastric mucosa was mounted in Ussing chambers and injured by bathing mucosal tissues in an HCl Ringer's solution (pH = 1.5) with or without SP1-8811 (1 μmol/L), cystic fibrosis transmem-brane conductance regulator (CFTR) inhibitor (inhibitor 172, 10 μmol/L, apical) and ClC-2 inhibitor ZnCl 2 , 300 μmol/L, apical), on the apical surface of tissues. Transepithelial resistance and mucosal-to-serosal 3 H-mannitol fluxes were measured over a 90-min period. Tissues were analyzed by morph metric techniques, Immunofluorescence and by western blots. RESULTS:Compared with control tissues, acid exposure decreased transepithelial electrical resistance (TER) and increased 3 H-mannitol flux. Pretreatment of gastric mucosa with SPI-8811 was protective against acid-induced decreases in TER (TER, 50 Ω . cm 2 vs 100 Ω . cm 2 ) and abolished increases in flux ( 3 H-mannitol flux, 0.10 μmol/L . cm 2 vs 0.04 μmol/L . cm 2 ). Evidence of histological damage in the presence of acid was markedly at- tenuated by SPI-0811. Immunofluorescence and western analysis for occludin revealed enhanced localization to the region of the tight junction (TJ) after treatment with SPI-8811. Pretreatment with the ClC-2 inhibitor ZnCl 2 , but not the selective CFTR inhibitor 172, attenuated SPI-8811-mediated mucosal protection, suggesting a role for ClC-2. Prostone may serve both protective and reparative roles in injured tissues. CONCLUSION: ClC-2 agonist SPI-8811 stimulated enhancement of mucosal barrier function by protecting TJ protein occludin in porcine gastric mucosa and thus protected the gastric acid injury in porcine stomach.
