Celastrus Orbiculatus Extract Inhibits Tumor Angiogenesis by Targeting Vascular Endothelial Growth Factor Signaling Pathway and Shows Potent Antitumor Activity in Hepatocarcinomas in Vitro and in Vivo

Objective： Ce/astrus orbicu/atus Thunb. has been used for thousands of years in China as a remedy against cancer and inflammatory diseases. This study aims to investigate whether C. orbiculatus extract （COE） could inhibit angiogenesis, which is the pivotal step in tumor growth, invasiveness, and metastasis. Methods： In this study, the extract from the stem of C. orbiculatus was used. Mouse hepatic carcinoma cells （Hepat-6） were treated with COE in different nontoxic concentrations （10, 20, 40, 80, and 160 μg/mL）. The mRNA and protein expression levels of vascular endothelial growth factor （VEGF） were detected by reverse transcription-polymerase chain reaction （RT-PCR） and Western blot, respectively; the active fractions were further tested on C57BL/6 mice and human umbilical vein endothelial cells （HUVEC） for any antiangiogenic effects. Results： COE significantly inhibited proliferation and induced apoptosis in Hepal-6 cells and inhibited VEGF expression at both mRNA and protein levels. Furthermore, this agent inhibited the formation of the capillary-like structure in primary cultured HUVEC in a dose-dependent manner. In vivo, COE significantly reduced the volume and weight of solid tumors with low adverse effects and decreased tumor angiogenesis. Conclusions： In summary, COE could be used to treat hepatic carcinoma. The mechanisms of the antitumor activity of COE may be due to its effects against tumor angiogenesis by targeting the VEGF protein.

Helicobacter pylori infection and gastropathy:A comparison between Indonesian and Japanese patients

AIM:To compare the effects of Helicobacter pylori(H pylori) infection on gastropathy between Indonesian and Japanese patients.METHODS:Biopsy specimens were obtained during upper gastrointestinal endoscopy from 167 subjects(125 Indonesians and 42 Japanese) with uninvestigated symptoms of dyspepsia.The specimens were analyzed for the presence of H pylori using urease analysis,histopathology,and cell culture.The grade and activity of gastritis was assessed using the updated Sydney system.RESULTS:The percentages of Indonesian and Japanese patients who were H pylori-positive at the antrum or body of the stomach were similar(68% and 59.5%,respectively;P=0.316).Of those who were H pylori-positive,more Japanese patients than Indonesian patients had high levels of polymorphonuclear cells(P=0.001),mononuclear cells(P=0.013),glandular atrophy(P=0.000),and intestinal metaplasia(P=0.011) in both the antrum and body of the stomach.CONCLUSION:The grade of gastritis and prevalence of mucosal atrophy and intestinal metaplasia were higher in Japanese patients.The difference between Indonesian and Japanese patients was signifi cant.

Long-term prognosis of focal segmental glomerulosclerosis treated with therapeutic low-density lipoprotein-apheresis in patients with severe kidney dysfunction and proteinuria

Background:The prognosis of focal segmental glomerulosclerosis patients with nephrotic syndrome is estimated to be 10%-20%in 5 years and 30%-50%in 10 years,leading to end-stage kidney disease.The response rate with steroid therapy is 40%-60%.Therapeutic low-density lipoprotein-apheresis(LDL-A)may be effective in patients with steroid resistance.Information regarding the long-term prognosis of patients with focal segmental glomerulosclerosis receiving this therapy is scarce.Methods:We investigated the effectiveness of treatment in 50 patients with primary focal segmental glomerulosclerosis diagnosed between 1961 and 2017 at Kanazawa University Hospital and related facilities.The patients were observed at least 12 months after biopsy or until end-stage kidney disease occurrence or death.Results:LDL-A was performed in four patients who presented with steroidresistant nephrotic syndrome(two patients had concurrent acute renal failure for which hemodialysis was performed).In comparison with 17 patients who did not receive LDL-A after 1989,the LDL-A group had higher urinary protein excretion(13.7 vs.5.2 g/day,P=0.053)and serum creatinine(4.11 vs.1.65 mg/dL)levels at onset,and a numerically higher remission rate(75.0%vs.58.7%)compared with the nonlipoprotein-apheresis group.Conclusion:Therapeutic LDL-A can be performed for critical cases and may improve the remission rate.

Gene expression of CCL8 and CXCL10 in peripheral blood leukocytes during early pregnancy in cows

Background: The aim of the present study was to evaluate CCL8 and CXCL10 expression and its regulatory mechanism in peripheral blood leukocytes(PBLs) at the time of maternal recognition in cows. Blood samples were collected on 14, 15, 16, 17 and 18 d after artificial insemination(AI). Based on the day of return of estrus, cows were divided into three groups, pregnant(n = 5), early embryonic mortality(EEM; n = 5) and late embryonic mortality(LEM; n = 5). The gene expression levels in PBLs were assessed with quantitative real-time reverse transcription PCR.Results: The expression of CCL8 and CXCL10 mRNA in PBLs gradually increased from 14 to 18 d of pregnant cows and significant differences were observed on 18 d(P < 0.05), whereas no significant changes were observed both in EEM and LEM cows. Interferon-stimulated protein 15 k Da(ISG15), myxovirus-resistance gene(MX) 1 and MX2 mRNA expression in PBLs increased from 14 to 18 d which was significant on 18 d of pregnant cows as well as in LEM cows(P < 0.05), but no changes were observed in EEM cows. To determine whether the expression of CCL8 and CXCL10 in PBLs was regulated by pregnancy-related substances or not, expression level was assessed after exposure to interferon-τ(IFNT) and CCL16. Monocytes, granulocytes and lymphocytes were obtained using density-gradient centrifugation and flow cytometry. The addition of IFNT(100 ng/mL) and CCL16(100 ng/mL) to cultured PBLs increased the expression of CCL8 and CXCL10 mRNA(P < 0.05). The expression of ISG15, MX1 and MX2 mRNA in PBLs was also stimulated by IFNT and CCL16(P < 0.05).Conclusions: The expression of CCL8 and CXCL10 genes increased in PBLs during early pregnancy. Since IFNT stimulated CCL8 and CXCL10 expression in cultured PBLs, the increase of CCL8 and CXCL10 might be pregnancy-dependent events.The expression of both CCL8 and CXCL10 in PBLs was stimulated by CCL16 as wel as IFNT, suggesting a chemokine interaction that at least includes CCL8, CXCL10 and CCL16, and may play a role in regulating maternal recognitio

Serous cystadenocarcinoma of the mesentery in a man:case report and review of literature

In February 2007,a 41-year-old Japanese male was admitted to our hospital with increasing upper abdominal pain.A contrast-enhanced computed tomography(CT)scan of the abdomen demonstrated a well-demarcated,hypodense cystic mass with a thickened wall in the mesocolon.The laboratory results were within normal limits,except for increased carcinoembryonic antigen,carbohydrate antigen 19-9,DUPAN-2 and SPAN-1.The patient was diagnosed as having a mesenteric malignant cyst,and during a laparotomy,a right hemicolectomy with mesenteric cystectomy was performed without rupture in March 2007.In the microscopic findings,there was a well-differentiated adenocarcinoma in the inner surface of the cyst and in the fibrous connective tissue of the hypertrophic cystic wall.The tumor cells were immunohistochemically reactive to cytokeratin(CK)7,CK18 and CK20.No remnant of the malignancy was detected in the resected margin of the colon,cyst,liver or peritoneum nor was an uptake detected in an 18[F]-fluorodeoxyglucose positron emission tomography/CT examination of other organs.Finally,the malignancy was concluded to be a serous cystadenocarcinoma of the mesentery.Nineteen months after the operation,the patient died from peritonitis carcinomatosa due to a small intestine rupture.This report suggests mesenteric cystadenocarcinomas originating in the ovary,oviduct and intestinal mucosa,but these were ruled out in our patient.In this report,we discuss a case of the malignant transformation of a cyst into adenocarcinoma,which to our knowledge has never been previously reported in a male patient.

通过电脑数据库,作者对一家日本大型医院病理实验室2002-2013年间的10,000例连续胃组织标本进行组织病理学分析。所有组织切片被回顾性观察,这10,000例胃组织标本中良性病变8579例,恶性病变1421例。恶性病变中胃癌1342例(94.4%),胃肠间质瘤(GIST)34例(2.4%),黏膜相关淋巴组织(MALT)淋巴瘤25例(1.8%),非霍奇金恶性淋巴瘤19例(1.3%),转移癌1例(0.1%)。在1342例胃癌中,管状腺癌755例,乳头状腺癌176例,黏液腺癌147例,印戒细胞癌145例,低/未分化腺癌114例,腺鳞癌4例,转移性肺小细胞癌1例。在手术切除标本中,早期胃癌258例,进展期胃癌521例。在34例GIST中,2例为上皮细胞型,32例为梭形细胞型;GIST大小为1-15 cm,平均5.6 cm;KIT(CD117)表达均为阳性,CD34阳性31例,desmin阳性2例,S100阳性4例。6例GIST进行了基因检测,均显示KIT和/或PDGFRA点突变。在25例MALT淋巴瘤中均可见到中心细胞样细胞和淋巴上皮病变;92%的病变可发现幽门螺杆菌感染。在19例非霍奇金淋巴瘤中,17例为弥漫性大B细胞淋巴瘤,1例为外周T细胞淋巴瘤,1例为NK细胞淋巴瘤。

研究了温轧下过渡元素对7475铝合金板材晶粒细化的影响。经350°C温轧、480°C固溶热处理后,用Zr代替Cr的7475铝合金具有稳定亚晶超细组织;平均亚晶直径接近3μm。结果表明,溶液中的Zr能稳定亚晶是因为温轧中析出了细小的Al3Zr粒子;另一方面,含Cr粒子化合物在温轧之前析出,并且在温轧过程中会变粗大。与传统冷轧7475铝合金板材相比,温轧细化亚晶板材具有独特的性能。固溶处理后的温轧板材,在厚度截面的亚晶组织具有高比例小于15°的小角度晶界;温轧T6态合金板材的强度高于传统7475铝合金板材强度10%。温轧铝合金板材最显著的特点是,在与轧制方向呈45°时高Lankford(r)值为3.5,而其平均Lankford(r)值为2.2。高Lankford(r)值有利于组织演变为β-fiber织构,特别是强{011}?211?黄铜部分。同时,温轧板材也具有高的抗应力腐蚀开裂性能(SCC)。根据Kikuchi线和透射电镜分析,温轧T6板材形成的小角度晶界导致无沉淀析出带(PFZs)难以形成,这是导致抗应力腐蚀开裂性能提高的重要因素,其原因是晶界和晶内的电化学性能存在差异。

In situ observation of the Rayleigh–Taylor instability of liquid Fe and Fe–Si alloys under extreme conditions: Implications for planetary core formation

Rayleigh–Taylor(RT)instability,which occurs when a heavy fluid overlies a light fluid in a gravitational field,is an important scenario for planetary core formation,especially beneath the planetary magma ocean.This process has been discussed based on numerical simulations and experiments using analog materials.However,experiments on the RT instability using the core-forming melt have not been performed at high pressures.In this study,we perform in situ observation of the RT instability of liquid Fe and Fe–Si(Si10 and 20 wt.%)alloys under high pressure using a high-power laser-shock technique.The observed perturbation on the Fe–Si surface grows exponentially with time,while there is no obvious growth of perturbations on the Fe in the measured time range.Therefore,the growth rate of the RT instability increases with Si content.The timescale of the initial growth of the RT instability in planetary interiors is likely to be much faster(by more than two orders of magnitude)than the 30–403106 year timescale of planetary core formation.

Superlattices with varying GaN well widths (2, 3, 6, 9 nm) and fixed AlGaN barrier (8 nm) with high Al-content (x=0.65) were grown. Streaky RHEED patterns indicated 2D growth mode for the superlattices. XRD measurements showed multiple satellite peaks corresponding to uniform periodicity of the GaN/AlGaN pairs. The AlGaN barrier XRD peak also shifted with increasing well widths, while the GaN XRD peak was nominally unchanged. Room temperature photoluminescence experiments revealed peak emissions at energies lower than the bulk GaN energy gap. The large red shift with respect to the bulk gap is attributed to significant Stark effect for wide multiple quantum wells.

Antimicrobial Effects of Gatifloxacin on Canine Periodontopathic Bacteria In Vitro

The aim of this study was to identify the potential antibacterial effects of gatifloxacin on one of canine periodontal pathogens, Porphyromonas gulae. The minimum inhibitory concentrations (MIC) of gatifloxacin and its bactericidal effects were investigated. Gatifloxacin inhibited the growth of the canine periodontopathic bacteria tested in broth. An MIC of 50 nM was found to be effective in inhibiting P. gulae. On performing adenosine triphosphate bioluminescence assay, gatifloxacin was found to exhibit bactericidal effects on the tested bacteria in a concentration-dependent manner. The safety of gatifloxacin in mammalian cells was evaluated by assessing the viability of rat bone marrow mesenchymal stem (BMMS) cells treated with gatifloxacin. Almost over 80% of BMMS cells survived after a 3-day culture when treated with 100 nM of gatifloxacin. These results indicate that locally administered gatifloxacin has the potential for being used to prevent canine periodontal infection.
