Objective:to assess the in-vitro antihacterial activity and anti-inflammatory activity of orally administered different extracts(Hydro-alcoholic,methanolic,ethyl acetate and hexane)of Rauvolfia tetraphylla(***)root bark in Carrageetiaii induced acute inflammation in ***:In-vitro antibacterial activity was evaluated for extracts against four Gram positive and four Gram negative bacteria by using cylinder plate ***-alcoholic extract(70%v/v ethanol)at 200,400 and 800 mg/kg doses and methanolic,ethyl acetate and hexane extracts at doses 100,200 and 400 mg/kg were tested for anti-inflammatory activity in Carrageenan induced rat paw oedema model and paw thickness was measured every one hour up to 6 ***:All extracts of *** root bark showed good zone of inhibition against tested bacterial *** Carrageenan induced inflammation model,hydro-alcoholic and methanolic extract of *** root bark at three different doses produced significant(Ption when compared to vehicle treated control group and hexane,ethyl acetate ***:In the present study extracts of *** root bark shows good in-vitro antibacterial activity and in-vivo anti-inflammatory activity in rats.

Pearl millet(Pennisetum glaucum)is one of the major millets with high nutritional *** crop exhibits exceptional resilience to drought and high ***,the processing of pearl millet poses a significant challenge due to its high lipid content,enzyme activity,and presence of ***,it becomes imperative to enhance the quality and prolong the shelf life of pearl millet flour by employing suitable *** treatment in the food industry has long been seen as promising due to its potential to reduce microbial load,inactivate enzymes,and improve nutrient *** study aims to investigate the effects of hydrothermal treatment on the quality characteristics of pearl *** independent variables of the study were soaking temperature(35,45,55℃),soaking time(2,3,4 h),and steaming time(5,10,15 min).treatment conditions had a statistically significant effect on nutrient *** antinutrients like tannins and phytates were reduced by 0.99% to 5.94% and 0.36% to 6.00%,respectively,after the *** activity decreased significantly up to 10% with the treatment *** findings of this study could potentially encourage the use of pearl millet flour in the production of various food items and promote the application of hydrothermal treatment in the field of food processing.

An advanced spectral diagnostic system was developed to measure the electron temperature(t_(e)),electron density(N_(e)),and ion temperature(t_(i))of the Huazhong University of Science and technology field-reversed configuration *** system consists of an optic fiber spectrometer with a wide spectral band and a 670 mm focal length high throughout Czerny-turner monochromator equipped with both a 3600 g mm^(-1)grating and a 2400 g mm^(-1)grating to measure the line *** these components is an electronmultiplying charge-coupled device camera to capture spectral *** relative intensity of the optical fiber spectrometer was calibrated using a standard luminance source,and the wavelength calibration of the spectrometer was accomplished using a Hg/Ar *** diagnostic setup was configured to measure electron density based on the Stark effect of H_γ(n=5→n=2;434.04 nm).Doppler broadening of an OⅢ(2s^(2)2p(^(2)P^(0))3p→2s^(2)2p(^(2)P^(0))3s;375.988 nm)emission line was measured and analyzed to obtain the ion temperature,and electron temperatures were estimated from the relative strength of H_(β)(n=4→n=2;486.14 nm)(D_(β))and H_(γ)(D_(γ))spectral lines when the electron density was obtained from Stark effect *** initial experimental results indicate that the highest electron temperature of the formation region was approximately 8 eV,the electron density of the colliding-and-merging region was approaching 10^(20)m^(-3),and the ion temperature reached about 40 eV.

Insufficient therapeutic strategies for acute kidney injury(AKI)necessitate precision therapy targeting its *** study reveals the new mechanism of the marine-derived anti-AKI agent,piericidin glycoside S14,targeting peroxiredoxin 1(PRDX1).By binding to Cys83 of PRDX1 and augmenting its peroxidase activity,S14 alleviates kidney injury efficiently in Prdx1-overexpression(Prdx1-OE)***,S14 also increases PRDX1 nuclear translocation and directly activates the Nrf2/HO-1/NQO1 pathway to inhibit ROS *** to the limited druggability of S14 with low bioavailability(2.6%)and poor renal distribution,a pH-sensitive kidney-targeting dodecanaminechitosan nanoparticle system is constructed to load S14 for precise treatment of AKI.L-Serine conjugation to chitosan imparts specificity to kidney injury molecule-1(Kim-1)-overexpressed *** developed S14-nanodrug exhibits higher therapeutic efficiency by improving the in vivo behavior of S14 *** encapsulation with micelles,the AUC_(0-t),half-life time,and renal distribution of S14 increase 2.5-,1.8-,and 3.1-fold,*** main factors contributing to the improved druggability of S14 nanodrugs include the lower metabolic elimination rate and UDPglycosyltransferase(UGt)-mediated *** summary,this study identifies a new therapeutic target for the marine-derived anti-AKI agent while enhancing its ADME properties and druggability through nanotechnology,thereby driving advancements in marine drug development for AKI.

the gastrointestinal tract is essential for food digestion,nutrient absorption,waste elimination,and microbial ***-cell transcriptome profiling of the intestinal tract has greatly enriched our understanding of cellular diversity,functional heterogeneity,and their importance in intestinal tract development and *** such profiling has been extensively conducted in humans and mice,the single-cell gene expression landscape of the pig cecum remains ***,single-cell RNA sequencing was performed on 45572 cells obtained from seven cecal samples in pigs at four different developmental stages(days(D)30,42,150,and 730).Analysis revealed 12 major cell types and 38 subtypes,as well as their distinctive genes,transcription factors,and regulons,many of which were conserved in *** increase in the relative proportions of CD8^(+)t and Granzyme A(low expression)natural killer t cells(GZMA^(low)NKt)cells and a decrease in the relative proportions of epithelial stem cells,tregs,RHEX^(+)t cells,and plasmacytoid dendritic cells(pDCs)were noted across the developmental ***,the post-weaning period exhibited an up-regulation in mitochondrial genes,COX2 and ND2,as well as genes involved in immune activation in multiple cell ***-cell crosstalk analysis indicated that IBP6^(+)fibroblasts were the main signal senders at D30,whereas IBP6^(−)fibroblasts assumed this role at the other *** cells established interactions with epithelial cells and IBP6^(+)fibroblasts in the D730 cecum through mediation of GZMA-F2RL1/F2RL2 *** study provides valuable insights into cellular heterogeneity and function in the pig cecum at different development stages.

Background Chitinase is an enzyme that hydrolyzes chitin,a major component of the exoskeleton of insects,including plant pests like *** present study aimed to investigate the expression of chemically synthesized barley ch1 and chi2 genes in cotton(Gossypium hirsutum)through Agrobacterium-mediated ***-five putative transgenic cotton plants were obtained,out of which fifteen plants successfully survived and were shifted to the *** gene-specific primers,amplification of 447 bp and 401 bp fragments confirmed the presence of the ch1 and chi2 genes in five transgenic cotton plants of the t0 *** five plants were further evalu-ated for their mRNA expression *** t0 transgenic cotton plants with the highest mRNA expression level and better yield performance in field,were selected to raise their subsequent *** the t1 cotton plants showed the highest mRNA expression levels of 3.5-fold in P10(2)for the ch1 gene and 3.7-fold in P2(1)for the chi2 *** in situ hybridization(FISH)confirmed a single copy number of ch1 and chi2(hemizygous)on chromosome ***,the efficacy of transgenes on whitefly was evaluated through an insect bioassay,where after 96 h of infestation,mortality rates of whitefly were calculated to be 78%–80%in transgenic cotton *** number of eggs on transgenic cotton plants were calculated to be 0.1%–0.12 per plant compared with the non-transgenic plants where egg number was calculated to be 0.90–1.00 per *** Based on these findings,it can be concluded that the chemically synthesized barley chitinase genes(ch1 and chi2)have the potential to be effective against insects with chitin exoskeletons,including *** transgenic cotton plants expressing these genes showed increased resistance to whiteflies,resulting in reduced egg numbers and higher mortality rates.

Objective: Despite cardiotoxicity overlap, the trastuzumab/pertuzumab and anthracycline combination remains crucial due to significant benefits. Pegylated liposomal doxorubicin(PLD), a less cardiotoxic anthracycline, was evaluated for efficacy and cardiac safety when combined with cyclophosphamide and followed by taxanes with trastuzumab/pertuzumab in human epidermal growth factor receptor-2(HER2)-positive early breast cancer(BC).Methods: In this multicenter, phase II study, patients with confirmed HER2-positive early BC received four cycles of PLD(30-35 mg/m^(2)) and cyclophosphamide(600 mg/m^(2)), followed by four cycles of taxanes(docetaxel,90-100 mg/m^(2) or nab-paclitaxel, 260 mg/m^(2)), concomitant with eight cycles of trastuzumab(8 mg/kg loading dose,then 6 mg/kg) and pertuzumab(840 mg loading dose, then 420 mg) every 3 weeks. the primary endpoint was total pathological complete response(tp CR, yp t0/is yp N0). Secondary endpoints included breast p CR(bp CR),objective response rate(ORR), disease control rate, rate of breast-conserving surgery(BCS), and safety(with a focus on cardiotoxicity).Results: Between May 27, 2020 and May 11, 2022, 78 patients were treated with surgery, 42(53.8%) of whom had BCS. After neoadjuvant therapy, 47 [60.3%, 95% confidence interval(95% CI), 48.5%-71.2%] patients achieved tp CR, and 49(62.8%) achieved bp CR. ORRs were 76.9%(95% CI, 66.0%-85.7%) and 93.6%(95% CI,85.7%-97.9%) after 4-cycle and 8-cycle neoadjuvant therapy, respectively. Nine(11.5%) patients experienced asymptomatic left ventricular ejection fraction(LVEF) reductions of ≥10% from baseline, all with a minimum value of >55%. No treatment-related abnormal cardiac function changes were observed in mean N-terminal pro-BNP(Nt-pro BNP), troponin I, or high-sensitivity ***: this dual HER2-blockade with sequential polychemotherapy showed promising activity with rapid tumor regression in HER2-positive BC.
