Systemic treatment strategies for triple-negative breast cancer

Triple-negative breast cancer(TNBC) is defined by the lack of immunohistochemical expression of the estrogen and progesterone receptors and human epidermal growth factor receptor 2(EGFR2). Most TNBC has a basal-like molecular phenotype by gene expression profiling and shares clinical and pathological features with hereditary BRCA1 related breast cancers. This review evaluates the activity of available chemotherapy and targeted agents in TNBC. A systematic review of PubM ed and conference databases was carried out to identify randomised clinical trials reporting outcomes in women with TNBC treated with chemotherapy and targeted agents. Our review identified TNBC studies of chemotherapy and targeted agents with different mechanisms of action, including induction of synthetic lethality and inhibition of angiogenesis, growth and survival pathways. TNBC is sensitive to taxanes and anthracyclins. Platinum agents are effective in TNBC patients with BRCA1 mutation, either alone or in combination with poly adenosine diphosphate polymerase 1 inhibitors. Combinations of ixabepilone and capecitabine have added to progression-free survival(PFS) without survival benefit in metastatic TNBC. Antiangiogenic agents, tyrosine kinase inhibitors and EGFR inhibitorsin combination with chemotherapy produced only modest gains in PFS and had little impact on survival. TNBC subgroups respond differentially to specific targeted agents. In future, the treatment needs to be tailored for a specific patient, depending on the molecular characteristics of their malignancy. TNBC being a chemosensitive entity, combination with targeted agents have not produced substantial improvements in outcomes. Appropriate patient selection with rationale combinations of targeted agents is needed for success.

Biochar is considered as a good metal sequester and ameliorates the metal toxicity and uptake in ***,its effec-tiveness over other organic amendments is not well *** aim of this study is to compare the biochar amendments alleviating the levels of Pb and Cd in Withania somnifera *** with other organic *** manure,vermicom-post,and biochar were applied in the field spiked with highly available Pb and *** bioavailability and accumulation,plant growth and plant physiology,antioxidant enzymes and metabolite content of *** along with soil properties were evaluated in all *** indicates that the application of organic manures significantly alleviated the metal levels(33-72%)in the plant in comparison with *** herbage yield was significantly higher(33-69%)under different organic manures as compared to the metal *** metabolite contents and antioxidant enzymes were higher in metal treatments in comparison with *** manures were not only able to restrain the Pb and Cd in soil but also enhanced the soil microbial *** indicate that biochar amendments were more promising than farmyard manure and vermicompost due to the presence of more stable carbon in biochar and more alleviation in metal *** addition,net profit in the cultivation of *** was higher for biochar amendments(50%higher)compared to *** study recommended that biochar could be a better option for commercial and safer production of ***.

The paper proposes a model for a micro-grid architecture incorporating the role of aggregators and renewable sources on the prosumer side, working together to optimize configurations and operations. The final model takes the form of a mixed-integer linear programming model. This model is solved using the CPLEX solver via GAMS by having a consistent data set.

Ocimum sanctum extract preserves neuronal echotexture and controls seizure in lithium-pilocarpine induced status epilepticus rats

Objective:To investigate the effect of Ocimum sanctum hydroalcoholic extract(OSHE)on seizure control and neuronal injury in rats with lithium-pilocarpine-induced status epilepticus(SE).Methods:SE was induced by administering lithium chloride followed by pilocarpine 24 h *** was administered either alone or in combination with valproate(VPA)3 days before SE induction until 14 days post-SE *** parameters were recorded on day 1(0-3 h),day 1-3 and day 4-14 *** day 14 post-SE,neurobehavioural tests(elevated plus maze and passive avoidance)were done followed by total antioxidant capacity,neuron-specific enolase,immunohistochemistry,and electron microscopic assessment in the hippocampus and cortex ***:OSHE+VPA provided more significant seizure protection(75%)than VPA(62.5%),OSHE(62.5%),or SE control(12.5%)(overall P=0.003).The latency to stage-3/4 seizures was increased and the number of stage-3/4 seizures was reduced in all treatment groups compared to the SE control group(P=0.002 and<0.001,respectively).The OSHE+VPA group also had better memory retention than other treatment groups(P<0.001)in the passive avoidance *** antioxidant capacity level was significantly higher and neuron-specific enolase was lower in the OSHE and OSHE+VPA groups compared to the SE control *** microscopic study showed significant myelin sheath damage(67.5%,P<0.05)and axonal degeneration(51.8%,P<0.001)in the hippocampus of the SE control group,which were alleviated by OSHE or OSHE+VPA *** immunohistochemical analysis,the OSHE,OSHE+VPA,and VPA groups had a significantly higher number of viable neurons and less neuronal loss compared to the SE control in the hippocampus(P<0.001).Conclusions:OSHE either alone or in combination with VPA shows better seizure control by preservation of neuronal echotexture and reducing oxidative stress in the hippocampus.

Background In the Indo-Gangetic Plain,rice-wheat is the most extensively practiced crop *** escalating issue of crop residue burning,particularly rice straw,and the necessity to lower the exorbitant expenses associated with fertilizer inputs stand out as significant challenges for farmers in the region.A well-suited integrated nutrient management(INM)strategy that focuses on recycling crop residues can serve as a solution to address these *** a strategy not only mitigates air pollution resulting from residue burning but also helps combat water pollution due to nitrate losses from *** experiments were used to evaluate the suitability of eight INM-modules that included various combinations of inorganic fertilizer rates(50%,100%,150%of recommended dose),crop residues(wheat and rice stubble retention at 30 cm standing stubble equivalent to 1/3 the straw yield),rice straw compost(RSC),farmyard manure(FYM),and green manuring(GM),compared to 100%recommended dose of fertilizers(F)and no fertilizer *** There was a considerable improvement in nitrogen mineralization,grain yields,and nitrogen use efficiency under GM+RSC-F50 and GM+*** INM modules would permit a 50%reduction in the use of chemical *** was a little yield penalty with in situ rice residue incorporation at 100%F;however,this could be overcome with 150%F fertilizer *** situ retention of wheat straw with a full application of fertilizer resulted in steadily rising crop yields over *** in the redox potential,soil pH,and soil organic carbon best accounted for the observed trajectories in nitrogen use *** The most promising INM modules for adoption by farmers in the Indo-Gangetic Plain to judiciously use crop residues and curtail chemical fertilizer inputs are green manuring with Sesbania aculeata+rice straw compost at 5 t ha^(−1)+only 50%of recommended dose of fertilizers(GM+RSC-F50),and green manuring with

Unravelling Effects of Temperature and Soil Moisture Stress Response on Development of Dry Root Rot [<i>Rhizoctonia bataticola</i>(Taub.)] Butler in Chickpea

Erratic rainfalls and rise in temperature have become more frequent under the changing scenario of climate particularly in semiarid tropics. As a consequence of it, a drastic shift of chickpea diseases have been recorded throughout the major chickpea growing regions in India and elsewhere. Dry root rot (DRR) caused by Rhizoctonia bataticola (Taub.) Butler [Pycnidial stage: Macrophomina phaseolina (Tassi) Goid] was found as a potentially emerging constraint to chickpea production than wilt (Fusarium oxysporum f. sp. ciceris). Increasing incidence of DRR indicate strong influence of climate change variables such as temperature and moisture on the development of disease. The present study therefore was conducted to quantify the role of temperature and soil moisture associated with infection, colonization and development of DRR under controlled environment. The DRR incidence was significantly affected by high temperature and soil moisture deficit. Out of five temperature regimes (15?C, 20?C, 25?C, 30?C and 35?C) and four moisture levels (40%, 60%, 80% and 100%), a combination of high temperature (35?C) and soil moisture content (60%) predisposes chickpea to DRR. The study clearly demonstrates that high temperature coupled with soil moisture deficit is the climate change variables predisposing chickpea to R. bataticola infection, colonization and development.

PepBind: A Comprehensive Database and Computational Tool for Analysis of Protein-peptide Interactions

Protein-peptide interactions, where one partner is a globular protein （domain） and the other is a flexible linear peptide, are key components of cellular processes predominantly in signal- ing and regulatory networks, hence are prime targets for drug design. To derive the details of the protein-peptide interaction mechanism is often a cumbersome task, though it can be made easier with the availability of specific databases and tools. The Peptide Binding Protein Database （Pep- Bind） is a curated and searchable repository of the structures, sequences and experimental observa- tions of 3100 protein^eptide complexes. The web interface contains a computational tool, protein inter-chain interaction （PICI）, for computing several types of weak or strong interactions at the pro- tein peptide interaction interface and visualizing the identified interactions between residues in Jmol viewer. This initial database release focuses on providing protein-peptide interface informa- tion along with structure and sequence information for protein-peptide complexes deposited in the Protein Data Bank （PDB）. Structures in PepBind are classified based on their cellular activity. More than 40% of the structures in the database are found to be involved in different regulatory pathways and nearly 20% in the immune system. These data indicate the importance of protein- peptide complexes in the regulation of cellular processes. PepBind is freely accessible at http：// ***/.

Composition and diversity of five major forest types in moist temperate climate of the western Himalayas

The present study was undertaken in five major forest types （dominated by Quercus semecarpifolia, Quercusfloribunda, Acer acuminatum, Abiespindrow and Aesculus indica, respectively） between 2400 and 2850 m a.s.1, in a moist temperate forest of the Mandal-Chopta area in the Garhwal region of Uttarakhand, India. The aim was to assess the variation in composition and diversity in different vegetation layers, i.e., herb, shrub and tree, among these five forest types. Diversity indices, such as the Shannon-Wiener diversity index, density, total basal cover, Simpson＇s concentration of dominance index, Simpson＇s diversity index, Pielou＇s equitability, species richness, species heterogeneity and r-diversity, were calculated to understand community structures. Dominance-diversity curves were drawn to ascertain resource apportionment among various species in different forest types.
