Real-time mechanical characterization of DNA degradation under therapeutic X-rays and its theoretical modeling

The killing of tumor cells by ionizing radiation beams in cancer radiotherapy is currently based on a rather empirical understanding of the basic mechanisms and effectiveness of DNA damage by *** contrast,the mechanical behaviour of DNA encompassing sequence sensitivity and elastic transitions to plastic responses is much better understood.A novel approach is proposed here based on a micromechanical Silicon Nanotweezers *** instrument allows the detailed biomechanical characterization of a DNA bundle exposed to an ionizing radiation beam delivered here by a therapeutic linear particle accelerator(LINAC).The micromechanical device endures the harsh environment of radiation beams and still retains molecular-level detection *** this study,the first real-time observation of DNA damage by ionizing radiation is *** DNA bundle degradation is detected by the micromechanical device as a reduction of the bundle stiffness,and a theoretical model provides an interpretation of the *** first real-time observations pave the way for both fundamental and clinical studies of DNA degradation mechanisms under ionizing radiation for improved tumor treatment.

Plasmon hybridization engineering in self-organized anisotropic metasurfaces

The engineering of self-organized plasmonic metasurfaces is demonstrated using a maskless technique with defocused ion-beam sputtering and kinetically controlled deposition. The proposed reliable, cost-effective, and controllable approach enables large-area （order of square centimeter） sub-wavelength periodic patterning with close-packed gold nanostrips. A multi-level variant of the method leads to high-resolution manufacturing of vertically stacked nanostrip dimer arrays, without resorting to lithographic approaches. The design of these self-organized metasurfaces is optimized by employing plasmon hybridization methods. In particular, preliminary results on the so-called gap-plasmon configuration of the nanostrip dimers, implementing magnetic dipole resonance in the near-infrared range, are reported. This resonance offers a superior sensitivity and field enhancement, compared with the more conventional electric dipole resonance. The translational invariance of the nanostrip configuration leads to a high filling factor of the hot spots. These advanced features make the large-area metasurface based on gap-plasmon nanostrip dimers very attractive for surface-enhanced linear and nonlinear spectroscopy （e.g., surface-enhanced Raman scattering） and plasmon-enhanced photon harvesting in solar and photovoltaic cells.

We determined genetic variation and metal and polycyclic aromatic hydrocarbon concentrations in Leptodon smithii moss collected in holm oak stands at cities, outskirts and remote areas of Campania and Tuscany（Italy） to investigate if anthropogenic pressure（pollutant emissions and land use change） affects moss genetic richness. In both regions, metal and polycyclic aromatic hydrocarbon concentrations reflected the trend urban 〉 outskirts 〉 remote areas, excepting Tuscany remote site. In both regions,the moss gene diversity increased from urban to remote areas. The findings suggest the extent and the fragmentation of urban green areas, as drivers of moss genetic richness.

Indications and results of renal biopsy in children:a 36-year experience

Background This study was conducted to investigate retrospectively the indications for renal biopsy(RB)in native kidneys and to analyze pathological findings in a single tertiary pediatric hospital in Southern Italy for the last 36 *** All patients who underwent RB at our hospital from 1979 to 2014 were *** renal tissue specimens were studied under light and immunofluorescent microscopy,while electron microscopy was performed only for specific clinical *** The study group included 213 patients(female 43.2%)who underwent 225 percutaneous native kidney *** age was 10.4 years(range 0.6-24 years).The most frequent indication for RB was nephrotic syndrome(44.4%),fol-lowed by proteinuria(27.6%),asymptomatic hematuria(17.3%)and acute kidney injury(9.8%).Gross hematuria appeared after biopsy in less than 5%of the patients,but none of them needed blood *** renal tissue sample was obtained in 95.5%of the renal *** glomerulonephritis(GN)was the most common finding(61.4%),followed by secondary GN(21.4%),tubulointerstitial diseases(3.7%)and hereditary nephropathy(2.8%),while in 10.7%of the cases,normal renal tissues were *** to histopathological diagnosis,the most common causes of primary GN were IgA nephropathy(20.9%),followed by minimal change disease(18.1%)and focal segmental glomerulosclerosis(11.6%).Conclusions The epidemiology of glomerular disease in our single-center cohort is similar to that shown in other national and international ***,our study shows that percutaneous ultrasound-guided RB is a safe,reliable and effec-tive technique in children.

背景既往研究显示精神分裂症患者存在广泛的认知功能损害。神经认知功能损害和社会认知功能损害被认为是精神分裂症导致严重功能残疾的原因。尽管认知功能对精神分裂症预后起关键作用,但无论是在科研研究中,还是临床实践中,相关评估都非常不足。方法本文对科学文献进行系统综述,详细阐述在科研研究和临床实践中对精神分裂症认知功能评估的若干推荐。结果专家共识和系统综述为精神分裂症认知功能的最佳评估提供了指导。基于遴选的证据,本欧洲精神病学协会精神分裂症认识功能损害评估指引(下文简称指引)建议,在精神分裂症的全病程以及精神病风险人群中进行神经认知功能和社会认知功能2个领域的全面且系统的评估。指引不仅建议使用知情者报告,也建议使用自我报告和基于访谈的认知功能评估工具。指引还系统地回顾了首次发作精神病患者和精神病高危人群的评估现状。结论对证据的全面审查和推荐建议可能有助于推动这一领域的发展,有利于进行更好的认知功能评估,并避免与其他精神病理学维度重叠。该指导性文件的传播可促进涉及精神分裂症认知功能评估的相关指南的制定,从而提高医疗质量并获得临床痊愈。

Clinical features and natural history of cryptogenic cirrhosis compared to hepatitis C virus-related cirrhosis

To characterize natural history of cryptogenic cirrhosis (CC) and compare its clinical features and outcomes to those of hepatitis C virus (HCV)-related *** prospective cohort of 102 consecutive patients at their first diagnosis of CC were enrolled in this study. The clinical data and outcomes were compared to an age- and Child-Pugh class-matched cohort of 110 patients with HCV-related cirrhosis. Diagnosis of cirrhosis was based on compatible clinical and laboratory parameters, ultrasound/endoscopic parameters and, whenever possible, on histological grounds and transient elastography. All cases of cirrhosis without a definite etiology were enrolled in the CC group. The parameters assessed were: (1) severity of liver disease at the time of first diagnosis; (2) liver decompensation during follow-up; (3) hepatocellular carcinoma (HCC); (4) orthotopic liver transplantation; and (5) death. The independent associated factors were evaluated by multiple logistic regression analysis, and survival and its determinants by the Kaplan-Meier model, log-rank test and Cox *** the first observation, median age was 66 and 65 years and male gender was 36% and 58% for CC and HCV cirrhosis, respectively. CC showed Child-Pugh class A/B/C of 47%/31%/22%, respectively. Compared to HCV cirrhosis, CC exhibited a significantly higher prevalence of metabolic syndrome (12% vs 54%, respectively), overweight/obesity, high BMI, impaired glucose tolerance, high blood pressure, dyslipidemia, hyperuricemia, cardiovascular diseases, extrahepatic cancer, and gallstones. Over a median period of 42 mo of follow-up, liver decompensation, HCC development and death for CC and HCV-related cirrhosis were 60.8%, and 54.4%, 16.7% and 17.2%, 39.2% and 30%, respectively. The median survival was 60 mo for CC. Independent predictors of death were age and Child-Pugh class at diagnosis. CC showed an approximately twofold higher incidence of HCC in Child-Pugh class ***

BACKGROUND Several studies report the useful therapeutic results of regional hyperthermia in association with chemotherapy(CHT) and radiotherapy for the treatment of pancreatic cancer. Modulated electrohyperthermia(mEHT) is a new hyperthermia technique that induces immunogenic death or apoptosis of pancreatic cancer cells in laboratory experiments and increases tumor response rate and survival in pancreatic cancer patients, offering beneficial therapeutic effects against this severe type of *** To assess survival, tumor response and toxicity of mEHT alone or combined with CHT compared with CHT for the treatment of locally advanced or metastatic pancreatic *** This was a retrospective data collection on patients affected by locally advanced or metastatic pancreatic cancer(stage Ⅲ and IV) performed in 9 Italian centers, members of International Clinical Hyperthermia Society-Italian Network. This study included 217 patients, 128(59%) of them were treated with CHT(no-mEHT) and 89(41%) patients received mEHT alone or in association with CHT. mEHT treatments were performed applying a power of 60-150 watts for 40-90 min, simultaneously or within 72 h of administration of *** Median patients’ age was 67 years(range 31-92 years). mEHT group had a median overall survival greater than non-mEHT group(20 mo, range 1.6-24, vs 9 mo, range 0.4-56.25, P < 0.001). mEHT group showed a higher number of partial responses(45% vs 24%, P = 0.0018) and a lower number of progressions(4% vs 31%, P < 0.001) than the no-mEHT group, at the three months follow-up. Adverse events were observed as mild skin burns in 2.6% of mEHT *** mEHT seems safe and has beneficial effects on survival and tumor response of stage Ⅲ-IV pancreatic tumor treatment. Further randomized studies are warranted to confirm or not these results.
