Biochemical Estimation of Glucose 6 Phosphate Dehydrogenase Deficiency in Saudi Adults: Different Methods and Its Rationalization

Background: Glucose-6-phosphate dehydrogenase (G6PD) deficiency is the most common human enzymopathy affecting 400 million people, globally. G6PD deficiency is an X-linked genetic condition, which is more likely to affect males than females. Heterozygous females go undetected in a commonly used method. The aim of the study was to identify & rationalize different biochemical methods for detections of G6PD deficiency. Methods: Cross section retrospective study was conducted on 1584 (800 males, 784 females) blood samples collected from King Abdulaziz University Hospital (KAUH) and King Fahd Armed force hospital (KFAFH) in Jeddah, Western Saudi Arabia. Blood samples were screened for G6PD activity by fluorescence spot test, semi quantitative color reduction test and spectrometric quantitative evaluation. Hemoglobin (Hb) was measured on the same sample by BC-3200 Auto hematology Analyser. G6PD activity was recorded as U/g Hb. Samples identified as deficient with cutoff ≤4.6 U/gHb. Results: The prevalence of G6PD deficiency identified by fluorescence spot test was 73(4.6%) and all were deficient male. By semi quantitative method, the prevalence rate was 51(3.2%) and again all were male deficit patients. However, when quantitative spectrometric method was used, the prevalence was found in 90(5.7%), where in 73(4.6%) deficient patients were males and 17(1.1%) were females. Conclusion: Since the fluorescence spot test did not miss any G6PD deficient male, it should be restricted to males and quantitative test should be done on females. Each ethnic group should cultivate their own cutoff value for categorization of deficient patients.

Suppression of Meloidogyne javanica by antagonistic and plant growth-promoting rhizobacteria

Four rhizobacteria selected out of over 500 isolates from rhizosphere of the vegetables in China were further studied for suppression of the root-knot nematode and soil-borne fungal pathogens in laboratory and greenhouse in Belgium. They were identified as Brevibacillus brevis or Bacillus subtilis by Biolog test and partial 16s rDNA sequence comparison. They not only inhibited the radial growth of the root-infecting fungi Rhizoctonia solani SX-6, Pythium aphanidermatum ZJP-1 and Fusarium oxysporum ***. cucumerinum ZJF-2 in vitro, but also exhibited strong nematicidal activity by killing the second stage larvae of Meloidogyne javanica to varying degrees in the greenhouse. The toxic principles of bacterium B7 that showed the highest juvenile mortality were partially characterized. The active factors were heat stability and resistance to extreme pH values. B7 used either as seed dressing or soil drench significantly reduced the nematode populations in the rhizosphere and enhanced the growth of mungbean plants over the controls in the presence or absence of R. solani.

Component Analysis and Antifungal Activity of the Compounds Extracted from Four Brown Seaweeds with Different Solvents at Different Seasons

The polar and nonpolar extracts of Colpomenia sinuosa, Padina pavonia, Cystoseira barbata and Sargassum vulgare collected during spring, summer and autumn were evaluated for their antifungal activities. The phytochemicals of unexplored seaweeds were analyzed by gas chromatography and mass spectrometric （GC/MS）. The algal extracts were tested for their antifungal activities against Aspergillus niger, A. flavus, Penicillium parasiticus, Candida utilis and Fusarium solani. Phytochemicals were extracted from the four seaweeds with various solvents including methanol, ethanol, acetone, chloroform and dimethyl ether. Among the various extracts, methanolic extract showed the highest inhibition activity on all fungal species. Seasonal variation in antifungal activity was studied, while methanol extracts showed the best antifungal activity in spring. Cystoseira barbata was the most effective seaweed, having anatifungal activity throughout the whole year. The UV-Vis phytochemical procedure and GC-MS analysis of the methanol extracts from tested species indicated the existence of different constituents. In conclusions, the compounds with antifungal activity were identified as indoles, terpenes, acetogenins, phenols, and volatile halogenated hydrocarbons.

Sesamol Alleviates the Cytotoxic Effect of Cyclophosphamide on Normal Human Lung WI-38 Cells via Suppressing RAGE/NF-κB/Autophagy Signaling

Cyclophosphamide(CYL)is a chemotherapeutic medication commonly used in managing various malignancies like breast cancer or ***,CYL has been documented to induce lung *** of CYL toxicity is through oxidative stress and the release of damage-associated molecular patterns(DAMPs).Sesamol(SES)is a natural antioxidant isolated from Sesamum indicum and its effect against CYL-induced lung toxicity is not studied *** study aims to inves-tigate whether SES could prevent any deleterious effects induced by CYL on lung using normal human lung cells,WI-38 cell line,without suppressing its *** were pretreated with SES and/or CYL for 24 h,then cell viability was estimated by MTS and trypan blue *** mode of cell death was determined by AO/EB ***,caspase-3 level,oxidative stress,and inflammatory markers were evaluated by colorimetric and ELISA ***-PCR was performed to evaluate RAGE,NF-κB,and Beclin-1 ***-treated WI-38 cells developed a significantly increased cell death with enhanced oxidative and RAGE/NF-κb/Autophagy signaling,which were all attenuated after pretreatment with ***,we concluded that SES offered a protective role against CYL-induced lung injury via suppressing oxidative stress and RAGE/NF-κB/Autophagy signaling,which is a natural safe therapeutic option against CYL toxicities.

OBJECTIVE:To investigate the efficacy of Fig fruit powder and olive on hepatic,renal and splenic injury induced by 2-nitropropane(2-NP)in mice,especially if they were used in ***:A total of 40 adult BALB/c male mice weighting 25-30 g/*** were categorized into five groups(8 each).Group 1 as negative *** 2 as positive control group intraperitoneally injected with 2-NP(100 mg/kg b.w.)3 times/weekly for eight *** 3 injected with 2-NP and were orally supplemented with Fig(300 mg/kg).Group 4 injected with 2-NP and were orally supplemented with olive(100 mg/kg).Group 5 injected with 2-NP and were orally supplemented with mixture of Fig and olive(3∶1 respectively).RESULTS:Histopathological observation of liver in mice treated with 2-NP showed cellular degeneration,pyknosis,and congestion of the portal *** kidney there were disorganization of the cortical tissues,cellular necrosis and plenty of inflammatory lymphocytic *** elevations in liver function parameters(alanine aminotransferase and aspartate aminotransferase),m RNA expression levels of tumor necrosis factor-α,nicotinamide adenine dinucleotide phosphate oxidase and cyclooxygenase were detected as anti-inflammatory markers and 5-lipoxygenase,interleukin-1βand interleukin-6 as inflammatory biomarkers for liver and spleen,also significant elevations was detected in lipid peroxidation *** levels of antioxidants,glutathione,glutathione peroxidase,catalase and superoxide dismutase were significantly ***:our findings indicated that Fig fruit powder and olive protected against hepatic,renal and splenic injury induced with 2-NP in mice,especially if they were used in combination.

BACKGROUND Digital radiography has recently been used in dentistry as a substitute for conventional film radiography *** imaging has many advantages and provides new possibilities for recording and interpreting radiographic *** system uses different types of digital *** To detect the frequency,type,and reasons behind the appearance of intraoral image artifacts acquired by photostimulable phosphor plates(PSP).METHODS This retrospective descriptive study was conducted in the oral and maxillofacial radiology unit of the dental clinics of the College of Dentistry,Princess Nourah University(PNU).All intraoral digital radiographs were acquired using(Gendex Expert DC.,United States)an intraoral X-ray machine with 7-mA,65-kVP using a PSP system(Soredex DIGORA Optime imaging plate)and laser scanners(Soredex DIGORA Optime),which can house all sizes of reusable intraoral PSP sensor plates with image acquisition software(MIPACS Dental Enterprise viewer 3.2.2).A total of 50000 intraoral radiographs were retrieved from the clinical database from April 2018 to April 2020 to evaluate the reason,type,and solutions to these image *** Overall,50000 intraoral digital radiographs were acquired in a two-year-period;that is,from April 2018 to April *** these,3550(7.1%)retakes were performed due to the presence of image *** radiographs were the most common image type of intraoral retakes(80.8%).Imaging artifacts were divided into three categories:operator,plate and scanning *** of 3550 retakes,5%,1.37%,and 0.73%were related to operator,plate,and scanning errors,*** cone cut was the most common operator error(988 images),Bite marks were the most common plate error(276 images),and delayed scanning artifacts were the most common scanning errors(145 images).The calculated kappa value for interobserver reliability was 0.99,indicating almost perfect interobserver *** Our study discussed intraora

Frequency dependence of the electrical conductivity and dielectric constants of polycarbonate (Makrofol-E) film under the effects of γ-radiation

Irradiation effects of γ-radiation on the physical and electrical properties of polycarbonate (Makrofol-E( film has been studied to be able to investigate the dielectric response of irradiated polymers for a wide range of fluence and frequency. The dielectric constant (ε') The loss tangent (tanδ), dielectric loss factor (ε''), the a.c electrical conductivity (σ) and the relaxation time (τ), were measured in the frequency range from (40) Hz to (4) MHz. These samples were irradiated by means of γ-rays from 10 up to 200 KGy. The change in different properties as a function of absorbed dose was studied. Degradation of the polymers leading to amorphisation was observed by increasing the absorbed γ- dose. The induced changes in the electrical conductivity due to γ-rays irradiation of Makrofol-E provide a better method for γ -dose measurements. A semi-empirical equation was developed to use Makrofol-E as a dielectric dosimeter. Furthermore, Makrofol-E has much greater resistance to radiation damage;the attained results suggested strongly the applicability of Makrofol-E to be used in medical products applications.

Objective: To evaluate the effect of Moringa oleifera leaf ethanol extract as an adjunct treatment on lead acetate induced hepatonephrotoxicity in rabbits. Methods: Thirty-six male New Zealand White rabbits were assigned into two main groups. The first group(14 rabbits) served as normal control. The second group(22 rabbits) was administered orally with lead acetate at a dose of 40 mg/kg/day, 5 days/week for 8 weeks. At the 4 th and the 8 th week of treatment, 6 animals(3 animals at each period) of the second group were sacrificed while the remaining animals(16 rabbits) were assigned randomly into 2 subgroups(8 rabbits each): treated and non-treated. The first subgroup was orally given 1 m L phosphate-buffered saline for further 4 weeks while the second subgroup was administered orally with Moringa oleifera leaf ethanol extract at a dose of 400 mg/kg/day for the same period. Blood samples were collected to determine hematological and serum biochemical indices. Tissue specimens were collected from the liver and kidney for evaluation of the oxidant/antioxidant markers and for histopathological ***: Lead acetate exposure decreased the mean body weight gain, hematocrit, hemoglobin, mean corpuscular volume, and lymphocytes count. Moreover, it markedly increased counts of monocytes and platelets, serum enzyme activity, levels of creatinine, total cholesterol, triglycerides, and low-density lipoprotein cholesterol. Malondialdehyde level was markedly increased while the reduced glutathione content was significantly decreased in liver tissue of lead intoxicated-rabbits. Histopathological alterations were also noticed in the liver and kidney of lead intoxicated rabbits. Moringa oleifera leaf ethanol extract significantly improved hematological and serum biochemical parameters and histopathological structure of the liver and kidney. Is There Any Role for Calcium Ionophore in ICSI Cycles for Cases of Non Obstructive Azoospermia?

Objectives: Evaluate the effect of artificial oocyte activation (AOA) using calcium ionophore (A23187) on the rate of fertilization and cleavage of embryos in surgically retrieved sperm of patients with non-obstructive azoospermia undergoing intracytoplasmic sperm injection (ICSI). Study design: This study was conducted on 60 infertile couples undergoing ICSI cycles as a randomized controlled parallel group’s experimental study in a private IVF center in Egypt from January 2018 to July 2019. ICSI cycles were divided into two groups: Group A: includes 30 ICSI patients with surgically retrieved sperms of non-obstructive azoospermia treated with calcium ionophore (A23187). Group C/Control: includes 30 ICSI patients with surgically retrieved sperms of non-obstructive azoospermia non-treated with calcium ionophore (A23187). Results: There was no statistical difference between both groups regarding the fertilization rate (p = 0.853). There was no statistical difference between them regarding implantation rate (p = 0.237). The percentage of Class A embryos in the ca ionophore group was 81.7%, while it was 82.8% in the control group. There was insignificant difference between them (
