Tumor necrosis factor a accelerates Hep-2 cells proliferation by suppressing TRPP2 expression

TRPP2, a Ca2＋-permeable non-selective cation channel, has been shown to negatively regulate cell cycle, but the mechanism underlying this regulation is unknown. Tumor necrosis factor a （TNF-a） is a proinflammatory cytokine extensively involved in immune system regulation, cell proliferation and cell survival. However, the effects and mechanisms for the role of TNF-a in laryngeal cancer remain unclear. Here, we demonstrated using western blot analyses and intracellular Ca〉 concentration measurements that TNF-a treatment suppressed both TRPP2 expression and ATP-induced Ca2＋ release in a laryngeal cancer cell line （Hep-2）. Knockdown of TRPP2 by a specific siRNA significantly decreased ATP-induced Ca2＋ release and abolished the effect of TNF-a on the ATP-induced Ca2＋ release. TNF-a treatment also enhanced Hep-2 cell proliferation and growth, as determined using cell counting and flow cytometry cell cycle assays. Moreover, TNF-a treatment down-regulated phosphorylated protein kinase R-like endoplasmic reticulum kinase （p-PERK） and phosphorylated eukaryotic translation initiation factor （p-elF2c0 expression levels, without affecting PERK and elF2ct expression levels in Hep-2 cells. We concluded that suppressing TRPP2 expression and TRPP2-mediated Ca2＋ signaling may be one mechanism underlying TNF〈t-enhanced Hep-2 cell proliferation. These results offer new insights into the mechanisms of TNF-a-mediated laryngeal cancer cell proliferation, and provide evidences showing a potential role of TNF-a in the development of laryngeal cancer.

Elastomers are essential for stretchable electronics, which have become more and more important in bio-integrated devices. To ensure high compliance with the application environment, elastomers are expected to resist, and even self-repair, mechanical damage, while being friendly to the human ***, inspired by peptidoglycan, we designed the first room-temperature autonomous self-healing biodegradable and biocompatible elastomers, poly(sebacoyl 1,6-hexamethylenedicarbamate diglyceride)(PSe HCD) elastomers. The unique structure including alternating ester-urethane moieties and bionic hybrid crosslinking endowed PSeHCD elastomers superior properties including ultrafast self-healing,tunable biomimetic mechanical properties, facile reprocessability, as well as good biocompatibility and biodegradability. The potential of the PSeHCD elastomers was demonstrated as a super-fast self-healing stretchable conductor(21 s) and motion sensor(2 min). This work provides a new design and synthetic principle of elastomers for applications in bio-integrated electronics.

A topological polymer network with Cu(Ⅱ)-coordinated reversible imidazole-urea locked unit constructs an ultra-strong self-healing elastomer

It is exceedingly desired, but difficult to construct self-healing materials with both excellent mechanical properties and healing efficiency, which are usually realized by using mutually exclusive methods. Here, we reconcile this contradiction by utilizing copper-bis-(imidazole-2-yl)-methane-urea(Cu-BIMU) locked units based on novel designed dynamic imidazole-urea bonds with coupled multiple noncovalent bonds(coordination bonds, π-π stacking bonds, and hydrogen bonds). The coordination of Cu(II) greatly reduces the electron-cloud density of imidazole, which lowers the free energy barrier of imidazole-urea bonds and promotes their reversible dissociation, as demonstrated by the density functional theory and small-molecule model reaction. The topological design of Cu-BIMU polyurethane(Cu-BIMU-PU), which concentrates multiple crosslinking-in-one locked unit to avoid the formation of excessive crosslinking sites to ensure high chain mobility, facilitates self-healing. Accumulative extensive intermolecular interactions endowed excellent mechanical properties to the resulting Cu-BIMU-PU elastomer with a tensile strength of 65.3 MPa, among the highest ever-reported value. This work provides a novel molecular design principle for fabricating high-performance dynamic polymers.

Development and application of KASP marker for high throughput detection of the seedless trait in grapevine

Molecular marker-assisted selection(MAS) can significantly accelerate and improve the efficiency of the breeding process in seedless grape cultivars. In this study, we developed the KASP_VviAGL11 and VviAGL11_410 markers based on a single nucleotide polymorphism(SNP) site(Chr18: 26889437(A/C)) of the VviAGL11 gene, and compared them with previously reported SSR markers p3_VvAGL11 and 5U_VviAGL11 by testing 101 cultivars and 81 F1 hybrid progenies. The results showed that both of the proposed markers obtained 100% accuracy rates in detecting allele A, which was closely associated with the seedless trait in grapes, while p3_VvAGL11 and 5U_VviAGL11 had lower accuracy rates due to their tendency to produce false positives. After careful evaluation of the technical advantages and disadvantages associated with these markers, we concluded that KASP_VviAGL11 was superior in terms of simplicity,cost-effectiveness, efficiency, and accuracy. Thus, we optimized the process of molecular MAS for seedless grapes,focusing on the KASP_VviAGL11 marker as a central component, to provide key technical support for the development of new seedless grape cultivars.

Orthodontically induced tooth root resorption(OIRR)is a serious complication during orthodontic *** cementum repair is the fundamental approach for the treatment of *** hormone(PTH)might be a potential therapeutic agent for OIRR,but its effects still lack direct evidence,and the underlying mechanisms remain *** study aims to explore the potential involvement of long noncoding RNAs(lncRNAs)in mediating the anabolic effects of intermittent PTH and contributing to cementum repair,as identifying lncRNA-disease associations can provide valuable insights for disease diagnosis and ***,we showed that intermittent PTH regulates cell proliferation and mineralization in immortalized murine cementoblast OCCM-30 via the regulation of the Wnt *** vivo,daily administration of PTH is sufficient to accelerate root regeneration by locally inhibiting Wnt/β-catenin *** RNA microarray analysis,lncRNA LITTIP(LGR6 intergenic transcript under intermittent PTH)is identified as a key regulator of cementogenesis under intermittent *** isolation by RNA purification(ChIRP)and RNA immunoprecipitation(RIP)assays revealed that LITTIP binds to mRNA of leucine-rich repeatcontaining G-protein coupled receptor 6(LGR6)and heterogeneous nuclear ribonucleoprotein K(HnRNPK)*** cotransfection experiments confirmed that LITTIP plays a structural role in the formation of the LITTIP/Lgr6/HnRNPK ***,LITTIP is able to promote the expression of LGR6 via the RNA-binding protein ***,our results indicate that the intermittent PTH administration accelerates root regeneration via inhibiting Wnt *** lncRNA LITTIP is identified to negatively regulate cementogenesis,which activates Wnt/β-catenin signaling via high expression of LGR6 promoted by HnRNPK.

Antimony selenide(Sb_(2)Se_(3))has drawn tremendous research attentions in recent years as an environment-friendly and cost-efficient photovoltaic ***,the intrinsic low carrier density and electrical conductivity limited its scope of *** this work,an effective ion doping strategy was implemented to improve the electrical and photoelectrical performances of Sb_(2)Se_(3) thin *** Sn-doped and I-doped Sb_(2)Se_(3) thin films with controllable chemical composition can be prepared by magnetron sputtering combined with post-selenization treatment based on homemade plasma sintered *** a result,the Sn-doped Sb_(2)Se_(3) thin film exhibited a great increase in carrier density by several orders of magnitude,by contrast,a less increase with one order of magnitude was achieved for the Idoped Sb_(2)Se_(3) thin ***,such cation or anion doping could simultaneously modify the conduction type of Sb_(2)Se_(3),enabling the first fabrication of a substrate structured Sb_(2)Se_(3)-based quasihomojunction thin film solar cell with configuration of Mo/Sb_(2)Se_(3)-Sn/Sb_(2)Se_(3)-I/ITO/*** obtained power conversion efficiency exceeding 2%undoubtedly demonstrated its attractive photovoltaic application potential and further investigation necessity.
