Dear Editor,Hepatocellular carcinoma(HCC)is the third leading cause of cancer-related deaths worldwide(Li and Satomura,2015).Early diagnosis and treatment are vital for reducing ***,biomarkers such as serum alpha fetoprotein(AFP),AFP-L2,and carcinoembryonic antigen are widely used tumor markers for managing HCC in patients(Shi et al.,2016).However,because of their low diagnostic sensitivity and specificity,novel biomarkers with high sensitivity and specificity for early HCC diagnosis are needed.

Oncolytic virotherapy is a promising strategy for the treatment of *** A virus(IAV) has shown potential as an oncolytic *** this study, we generated a recombinant PR8 influenza viral vector, called delNS1-GM-CSF, with a partial deletion in NS and the granulocyte-macrophage colony-stimulating factor(GM-CSF) coding sequence inserted into the influenza nonstructural protein 1 *** morphological characteristics of delNS1-GM-CSF were *** delNS1-GM-CSF virus replicated well in various cell lines, including MDCK, A549, SMCC7721, and HepG2 ***, selective cytotoxicity of the virus was observed in various hepatocellular carcinoma(HCC) cell lines, while no effect was demonstrated in the normal liver cell line LO2, as indicated by 3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide and crystal violet ***, using a model based on the growth of HepG2 cells as a xenograft in nude mice, we found that a reassortant delNS1-GM-CSF virus inhibited tumor growth significantly following intratumoral injection in a dose-dependent *** vivo results showed that the tumor inhibition efficacy of delNS1-GM-CSF was observed in HCC clinical *** together, these results are the first to demonstrate that IAVs may have potential as oncolytic virotherapeutic agents against HCC.

Staphylococcal enterotoxin B(SEB), which is produced by the Staphylococcus aureus bacterium, causes food poisoning and profound toxic shock ***, no approved vaccine or specific drug is available to treat SEB *** this study, we fabricated dissolving microneedles(MNs) loaded with recombinant SEB(rSEB) protein and evaluated its characteristics including dissolution profile, protein particle size, insertion depth, antigen retention time in vivo, and skin *** results showed that rSEB protein-loaded dissolving MNs made of chondroitin sulfate(2%) and trehalose(0.8%) could easily penetrate into the mouse skin in vivo within 5 *** rSEB particle size was unchanged before and after MN *** skin penetration depth of the MNs was 260μ***, the MNs also significantly extended the antigen retention time in *** addition, rSEB protein-loaded dissolving MNs triggered slight erythema at the beginning of administration, but this erythema disappeared within several *** importantly, we investigated whether rSEB protein-loaded dissolving MNs could induce robust immune responses that conferred protection against a lethal SEB *** challenge studies revealed that full-dose MN immunization induced a high level of antibody response, conferring 100% protection against a lethal SEB toxin challenge, but there was only a 60% survival rate in mice observed in the 1/2 dose MN(dose sparing)*** also determined the tissue pathological alterations of the immunized *** together, our dissolving MNs may present a promising transcutaneous immunization strategy for treating SEB intoxication.

Influenza virus (IAV)infection is a major cause of severe respiratory illness that affects almost every country in the *** infections result in respiratory illness and even acute lung injury and death,but the underlying mechanisms responsible for IAV pathogenesis have not yet been fully *** this study,the basic fibroblast growth factor 2 (FGF2)level was markedly increased in H1N1 virus-infected humans and ***2,which is predominately derived from epithelial cells,recruits and activates neutrophils via the FGFR2-PI3K-AKT-NFKB signaling ***2 depletion or knockout exacerbated influenzaassociated disease by impairing neutrophil recruitment and *** importantly,administration of the recombinant FGF2 protein significantly aUeviated the severity of IAV-induced lung injury and promoted the survival of IAV-infected *** on the results from experiments in which neutrophils were depleted and adoptively transferred,FGF2 protected mice against IAV , infection by recruiting ***,FGF2 plays a critical role in preventing IAV-induced lung injury,and FGF2 is a promising potential therapeutic target during IAV infection.

Hepatitis C virus(HCV) has a devastating impact on human health,and infections can progress into liver fibrosis,cirrhosis,and hepatocellular *** is no effective HCV *** this study,we rescued a recombinant PR8 influenza viral vector,called rgFLU-HCV,carrying the core and envelope glycoprotein(C/E1/E2) epitopes of HCV inserted into the influenza nonstructural protein 1 *** morphological characteristics of rgFLU-HCV and the expression of the C/E1/E2 epitopes of HCV were ***-HCV replicated in various cell lines,including MDCK,A549,and Huh7.5 *** importantly,in BALB/c mice immunized intranasally twice at a 21-day interval with 10,10,or 10 TCID rgFLU-HCV,the viral vector induced a robust antibody response to influenza and HCV and potent IFN-γ and IL-4 secretion in response to HCV antigens in a dose-dependent *** rgFLU-HCV virus also stimulated IFN-γ production by virus-specific peripheral blood mononuclear cells in patients with chronic HCV *** study demonstrated that rgFLU-HCV carrying HCV antigens is immunogenic in vivo and has potential for the development of a HCV vaccine.

Hepatocellular carcinoma(HCC) is a leading cause of cancer mortality worldwide. Despite progress in the diagnosis and treatment of HCC, its prognosis still remains unfavorable. In accumulating studies, long non-coding RNAs(lncRNAs) are emerging as vital players in tumorigenesis and cancer progression. However, the molecular mechanisms and clinical significance of lncRNAs in HCC remain largely elusive. In this study, we analyzed lncRNA microarray expressionprofiles of tumor samples and its matched adjacent tissue samples from 10 HCC patients and found that the expression of RP11J20.1 in HCC tissues is makedly lower than adjacent tissues. Expression of RP11J20.1 was also remarkably downregulated in 112 HCC tissues and cell lines by qRT-PCR. Low RP11J20.1 expression was correlated with alphafetoprotein(AFP) level, tumor size and microvascular invasion. Kaplan-Meier survival analysis demonstrated that low RP11J20.1 expression predicts poor prognosis of HCC patients. Gain-of-function and loss-of-function experiments showed that RP11J20.1 promotes HCC cells proliferation, cell cycle progression, and inhibits HCC cells apoptosis. Further experiments revealed that RP11J20.1 promotes HCC cells proliferation through inhibiting STAT1 genes expression. Collectively, our results demonstrated the clinical prognostic significance and roles of RP11J20.1 in HCC, and suggested that RP11J20.1 may be considered as a prognostic biomarker and therapeutic target for HCC.

Hepatocellular carcinoma(HCC) is the most common neoplasm and is a leading cause of cancer-related *** advances in the diagnosis and management of HCC,its prognosisremain *** evidence has shown that long intergenic noncoding RNAs(lincRNAs) play central roles inthe development of *** this study,we identified a long intergenic noncoding KNA referred to as lincRNA P7 in HCC and explored its clinical significance and biological functions in *** expression level of lincRNA P7 was significantly aberrantly deceased in HCC cancer tissues and cells ***-and loss-of-function experiments revealed that overexpression of lincRNA P7 significantly inhibited the proliferation of HCC-derived cancer cells,whereas lincRNA P7 knockdown promoted cell ***,lincRNA P7 blocked Erk1/2 signaling and repressed activation of the STAT1 *** nude mouse models,we show that overexpression of lincRNA P7 effectively repressed HCC xenograft tumor growth in ***,a clinical investigation demonstrated that down-regulated lincRNA P7 expression correlated with liver cirrhosis,Hepatitis B virus(HBV)infection,clinical stage of the tumor and recurrence.A Kaplan-Meier survival analysis showed that the expression of lincRNA P7 was significantly related to overall survival(P=0.003) and recurrence-free survival(P=0.031).Collectively,our findings suggested that the down-regulation of lincRNA P7 predicts poor clinical outcomes for HCC patients and might be a powerful candidate prognostic biomarker and target in HCC.
