A New Species of the Genus Tylototriton(Amphibia: Urodela: Salamandridae) from Eastern Himalaya

A new species of the genus Tylototriton is described from eastern Himalaya based on molecular and morphological comparisons. The new species is diagnosable from the closely-related species by having light brown colouration in dorsal region in life, flat and blunt snout, greatly separated dorsolateral bony ridges on head and straightthick tailfin. In addition to head morphology, the new species is also morphologically distinguishable from its closelyrelated species Tylototriton shanorum by having 16 dorsal warts and average smaller Snout Vent Length（SVL）.

Sponges (Phylum: Porifera) is one of the major groups in the Lakshadweep coral reefs. These sponges harbor diverse bacteria with metabolic potentiality. From biodiversity to biotechnological prospecting, scientific investigations related to sponge associated microorganisms have expanded, but remain rather limited to few geographic locations. In this study, culturable bacteria associated with two demosponges viz Dysidea granulosa, Sigmadocia fibulata and the ambient water were screened for commercially important enzymes such as amylase, protease, gelatinase, lipase, deoxyribonucleic, phosphatase and urease. Amylase and phosphatase were the predominant enzymes produced by >80% of sponge-associated bacteria compared to the ambient water. Nearly 50% of the sponge-associated bacteria expressed multiple enzymatic activities (> 4) with variation in the percentage of expression of individual enzymes. More than 65% of the culturable heterotrophic bacteria associated with sponges were Gammaproteobacteria. The order Vibrionales was the main source for multiple enzyme production. Sponge associated bacteria formed more closely related clusters than the water isolates based upon their activity pattern. High recovery of sponge-associated bacteria with multiple enzymatic activities suggest that these versatile bacteria are yet unexploited potential for bioprospecting.

Ciprofioxacin, commonly used in India as an anti-microbial for prolonged use in chronic and non-specific indications, may affect the bioavailability of the drug. The drug prescribed is commonly taken with multivitamins, calcium and milk. A simple and reliable analytical methodology obtaining a correlation with in vivo urinary excretion studies using UV and HPLC and in vitro dissolution studies （IVIVC） has shown a significant increase in elimination rate of ciprofloxacin co-administered with multivitamins, calcium and milk. Appreciable IVlVC results proved that dissolution studies could serve as an alternative to in vivo bioavailability and also support bio-waivers.

Simultaneous determination of ezetimibe and simvastatin in rat plasma by stable-isotope dilution LC-ESI-MS/MS and its application to a pharmacokinetic study

A simple, sensitive and specific liquid chromatography-tandem mass spectrometry method was developed for simultaneous quantification of ezetimibe and simvastatin in rat plasma. The deuterium isotopes： ezetimibe d4 and simvastatin d6 were used as internal standards for ezetimibe and simvastatin, respectively. MS/MS detection involved a switch of electron spray ionization mode from negative to positive at retention time 3.01 rain. Samples were extracted from plasma by liquid-liquid extraction using tertiary butyl methyl ether. Chromatographic separation was achieved with Agilent Eclipse XBD-CIs column using mobile phase that consisted of a mixture of ammonium acetate （pH4.5; 10 mM）-acetonitrile （25：75 v/v）. The method was linear and validated over the concentration range of 0.2--40.0 ng/rnL for simvastatin and 0.05-15.0 ng/mL for ezetimibe. The transitions selected were m/z 408.3→271.1 and m/z 412.0→275.10 for ezetimibe and ezetimibe d4, and m/z 419.30 → 285.20 and rrdz 425.40 →199.20 for simvastatin and simvastatin d6. Intra- and inter-batch precisions for ezetimibe were 1.6-14.8% and 2.1-13.4%; and for simvastatin 0.94-9.56% and 0.79-12%, respectively. The proposed method was sensitive, selective, precise and accurate for the quantification of ezetimibe and simvastatin simultaneously in rat plasma. The method was successfully applied to a pharmacokinetic study by oral co-administration of ezetimibe and simvastatin in SD rats.

This paper presents a humanoid robotic arms controlled by tracking the human skeleton movement in real-time using Kinect upper limbbody tracking. Using Kinect tracking algorithm, the positions of upper limb arms of the body to the wrist in 3D space can be estimated by processing depth images from the Kinect. An extraction of 3D co-ordinates of the user’s both arm in real-time then Arduino microcontroller is transferring the data between both of computer and the humanoid robotic arm. This method provides a way to send movement task to the humanoid robotic manipulator instead of sending the end position motion like gesture-based approaches and this method has been tested in detect, tracking and following the movement of human skeleton gesture. Designing complete prototype of a humanoid robotic arms with 4DOF three joints in shoulder and one elbow joint to the wrist that look like the Human arm Structure, Appearance and Action that represent human arm movement performed by the humanoid robotic arm. The error was and response time result generated is small (less than 4.6% and 105 ms).

灰盖鬼伞(Coprinopsis cinerea)是一种嗜粪模式食用菌,可在实验室条件下快速生长并结实。子实体发育从黑暗环境下形成初级菌丝结开始,随后在光照刺激下初级菌丝结紧密聚集形成二级菌丝结,然后二级菌丝结继续分化出菌柄和菌盖。在子实体形成培养条件下,前5天(P1-P5)为原基发育期,第6天担子及担孢子中发生核融合及减数分裂,伴随菌柄伸长和菌盖形成,子实体发育成熟。成熟的子实体在第7天自溶,含有孢子的液滴落地并释放出孢子。本文运用不同的组织染色技术观察原基组织切片,从而发现灰盖鬼伞子实体发育过程中不同的组织和细胞类型。前担子比其他组织更容易被乳酚蓝、迈尔士苏木精明矾、孔雀绿、曙红染色。菌盖皮最容易被孔雀绿和乳酚蓝染色。过碘酸雪夫氏染色(periodic acid Schiff,PAS)的菌盖皮和子实基层颜色比其他组织深,其中菌盖皮颜色很深,子实基层颜色稍深。所有切片还尝试了各种可能组合的双染。在PAS和苏木精明矾先后双染色下,子实基层和前胆子可以获得最佳区分。

Influence of Laser-Induced Bubble Formation on Laser Chemical Machining

Laser Chemical Machining (LCM) is a non-conventional processing method, which enables very accurate and precise ablation of metallic surfaces. Material ablation results from laser-induced thermal activation of heterogeneous chemical reactions between electrolytes and a metallic surface. However, when processing metallic surfaces with LCM, large fluctuations in ablation quality can occur due to rising bubbles. The for-mation of bubbles during laser chemical machining and their influence on the ablation quality has not been investigated. For a more detailed investigation of the bubbles, ablation experiments on Titanium and Ce-ramic under different thermal process conditions were performed. The experiments were recorded by a high-speed camera. The evaluation of the video sequences was performed using Matlab. The resulting bubbles were analyzed regarding their size and frequency. The results show that boil-ing bubbles formed on both materials during processing. Titanium also produces smaller bubbles, which can be identified as process bubbles ac-cording to their size. Furthermore, it was found that undisturbed laser chemical ablation can be achieved in the presence of a boiling process, since both boiling bubbles and process bubbles were detected during machining within the process window.

A Comparative Phytochemical and Biological Study between Different Solvent Extracts of <i>Bombax ceiba </i>Roots Available in Bangladesh

Use of different solvent systems for extraction of plant materials may cause variation in their bioactivities. The present study was conducted to evaluate the presence of different phytoconstituents and to compare in vitro bioactivities of petroleum ether, dichloromethane (DCM) and methanol extracts of Bombax ceiba (B. ceiba) roots available in Bangladesh. Preliminary phytochemical screening was conducted using specific standard procedure. Antioxidant activity of the extracts was evaluated using DPPH radical scavenging assay. Determination of total phenolic and flavonoid content was also carried out. Antibacterial and cytotoxic activities were investigated using disc diffusion method and brine shrimp lethality bioassay, respectively. All the experiments were carried out from February 2016 to September 2016. Phytochemical evaluation revealed the presence of alkaloids, terpenoids, carbohydrates, tannins, flavonoids, saponins and steroids. The methanol extract showed the highest DPPH radical scavenging activity and had the highest phenolic (187.42 ± 3.77 mg/g, GAE) and flavonoid content (74.67 ± 4 mg/g, QE) followed by the DCM and petroleum ether extracts. The extracts showed positive correlation between DPPH radical scavenging activity with the phenolic and flavonoid content. All the extracts showed mild to moderate in vitro antibacterial activity with zone of inhibition ranging from 7 mm to 13 mm. In brine shrimp lethality bioassay, the observed LC50 values for petroleum ether, DCM and methanol extracts were 70.72 μg/ml, 37.72 μg/ml and 22.58 μg/ml, respectively which revealed strong cytotoxic potential of the extracts compared to the positive control. The results indicated that B. ceiba roots could be a very potent source of natural radical scavenger and cytotoxic agent.
