The efficiency and safety evaluation of hemoglobin hydrolysate as a non-heme iron fortifier

Hemoglobin hydrolysate is derived from the enzymatic degradation of hemoglobin.This work aimed to evaluate whether hemoglobin hydrolysate promotes the absorption of non-heme iron and the safety of absorbed iron in mice by analyzing the iron binding content,iron circulation,and liver homeostasis.We found that hemoglobin hydrolysate promoted the absorption of non-heme iron with high efficiency in duodenum by spontaneously binding non-heme iron during digestion,and increased hepatic iron content by up-regulating divalent metal transporter 1,zinc transporter 14,but hepatic iron content only increased at 3 weeks.Duodenal iron entered the blood through ferroportin without restriction at 3 weeks,and excessive iron entered the liver and then affected the hepatocyte membranes permeability and lipid synthesis through oxidative stress.With the prolongation of dietary intervention,the up-regulated hepcidin acted on the ferroportin to restrict excess iron from entering the blood,and then the hepatic homeostasis recovered.In addition,hemoglobin hydrolysate enhanced the hepatic antioxidant capacity.Taken together,hemoglobin hydrolysate has a strong ability to promote the absorption of non-heme iron in vivo,and the absorbed iron is relatively safe due to the regulation of hepcidin.

Inverse design of mechanical metamaterial achieving a prescribedconstitutive curve

Besides exhibiting excellent capabilities such as energy absorption,phase-transforming metamaterials offer a vast design space for achieving nonlinear constitutive relations.This is facilitated by switching between different patterns under deformation.However,the related inverse design problem is quite challenging,due to the lack of appropriate mathematical formulation and the convergence issue in the post-buckling analysis of intermediate designs.In this work,periodic unit cells are explicitly described by the moving morphable voids method and effectively analyzed by eliminating the degrees of freedom in void regions.Furthermore,by exploring the Pareto frontiers between error and cost,an inverse design formulation is proposed for unit cells.This formulation aims to achieve a prescribed constitutive curve and is validated through numerical examples and experimental results.The design approach presented here can be extended to the inverse design of other types of mechanical metamaterials with prescribed nonlinear effective properties.

Maize(Zea mays subspecies mays)is an important commercial crop across the world,and its flowering time is closely related to grain yield,plant cycle and latitude adaptation.FKF1 is an essential clock-regulated blue-light receptor with distinct functions on flowering time in plants,and its function in maize remains unclear.In this study,we identified two FKF1 homologs in the maize genome,named ZmFKF1a and ZmFKF1b,and indicated that ZmFKF1a and ZmFKF1b independently regulate reproductive transition through interacting with ZmCONZ1 and ZmGI1 to increase the transcription levels of ZmCONZ1 and ZCN8.We demonstrated that ZmFKF1b underwent artificial selection during modern breeding in China probably due to its role in geographical adaptation.Furthermore,our data suggested that ZmFKF1bHap_C7 may be an elite allele,which increases the abundance of ZmCONZ1 mRNA more efficiently and adapt to a wider range of temperature zone than that of ZmFKF1bHap_Z58 to promote maize floral transition.It extends our understanding of the genetic diversity of maize flowering.This allele is expected to be introduced into tropical maize germplasm to enrich breeding resources and may improve the adaptability of maize at different climate zones,especially at temperate region.

Background The thirteen-valent pneumococcal conjugate vaccine(PCV13)is not included in the national immu-nization program and is administered voluntarily with informed consent in China.In preparation for assessing the impact of pilot introduction in Hainan Province,we conducted a carriage study among children under 5 years of age from four locations in Hainan Province,China.Methods From March to June 2022,nasopharyngeal(NP)swabs,collected from healthy children aged younger than 59 months who lived in the 4 different locations(Haikou,Wanning,Baisha and Qiongzhong)in Hainan Province,were tested for pneumococcus using conventional culture.Pneumococcal isolates were serotyped using the Quel-lung reaction.Risk factors associated with pneumococcal colonization were assessed using univariate analysis and multivariable logistic regression adjusting for age,daycare attendance and other factors.Results Pneumococcus was isolated in 710(30.4%)of the 2333 children enrolled.Of 737 pneumococci,29 serotypes were identifed;60.9%were PCV13 serotypes;the most common vaccine serotypes were 6B(20.4%),19F(13.0%),6A(11.9%)and 23F(6.1%);and the most common nonvaccine serotypes were 23A(12.9%),34(6.1%)and nontypeable(NT)pneumococci(5.6%).Children vaccinated with PCV13 had lower carriage(17.7%vs 32.5%;P=0.0001)and fewer PCV13 serotypes(41.9%vs 62.7%;P=0.0017)compared to unimmunized children.After adjustment,NP carriage Was higher among children attending daycare(aOR=2.3,95%CI:1.7-3.2),living in rural areas(aOR=1.4,95%CI:1.1-1.8),living with siblings(aOR=1.3,95%CI:1.0-1.6)and whose mothers had completed senior high/technical sec-ondary school(aOR=1.5,95%CI:1.1-2.0).In contrast,completion of 3-4 doses of PCV13 were associated with a lower carriage rate(aOR=0.6,95%CI:0.4-0.9).Conclusions We established the baseline of pneumococcal carriage,serotype distribution and PCV13 immuni-zation rates among healthy children under 5 years of age in Hainan Province,prior to the introduction of PCV13 into the national immunization program.The high proport

This study unveils a novel and potentially economical particulate discrete element modeling (DEM) technique for large-scale simulation of in-situ direct shear tests (DST) and plate load tests， aimed at accurately determining the shear strength and bearing capacity of granular soil foundations， and exploring the key factors influencing these properties. To overcome the limitations associated with the oversimplification of particle shapes， this work introduces an innovative contact model based on the Hertz contact mechanism that incorporates rolling resistance， enhancing the model’s ability to simulate realistic particle interactions. Guided by Prandtl’s foundation bearing-capacity theory， the dimensions of the foundation model for the DST and plate load tests were meticulously calculated to reflect realistic conditions. DEM simulations showed the peak friction angle from in-situ DST to be consistently lower than laboratory DST， due to a smaller dilatation angle， while proposed loading methods for plate load tests established linear relationships between the foundation’s ultimate bearing capacity and its dimensions， also defining the capacity envelope. Crucially， this study highlights a crucial finding: in-situ direct shear tests (DST) conducted under high normal stress may lead to foundation failure， indicating that excessive normal stress should be avoided to ensure the accuracy of in-situ shear test data and its subsequent applicability in engineering design.

Background:According to traditional Chinese medicine(TCM),drugs supplementing the vital energy,Qi,can eliminate tumors by restoring host immunity.The objective of this study is to investigate the underlying immune mechanisms of anti-tumor activity associated with Qi-supplementing herbs,specifically the paired use of Huangqi and Danggui.Methods:Analysis of compatibility regularity was conducted to screen the combination of Qi-supplementing TCMs.Using the MTT assay and a transplanted tumor mice model,the anti-tumor effects of combination TCMs were investigated in vitro and in vivo.High content analysis and flow cytometry were then used to evaluate cellular immunity,followed by network pharmacology and molecular docking to dissect the significant active compounds and potential mechanisms.Finally,the anti-tumor activity and the mechanism of the active ingredients were verified by molecular experiments.Results:There is an optimal combination of Huangqi and Danggui that,administered as an aqueous extract,can activate immunity to suppress tumor and is more effective than each drug on its own in vitro and in vivo.Based on network pharmacology analysis,PIK3R1 is the core target for the anti-tumor immunity activity of combined Huangqi and Danggui.Molecular docking analysis shows 6 components of the combined Danggui and Huangqi extract(quercetin,jaranol,isorhamnetin,kaempferol,calycosin,and suchilactone)that bind to PIK3R1.Jaranol is the most important component against breast cancer.The suchilactone/jaranol combination and,especially,the suchilactone/kaempferol combination are key for immunity enhancement and the anti-tumor effects of the extract.Conclusions:The combination of Huangqi and Danggui can activate immunity to suppress breast cancer and is more effective than the individual drugs alone.

A General Authentication and Key Agreement Framework for Industrial Control System

In modern industrial control systems(ICSs), when user retrieving the data stored in field device like smart sensor, there exists two main problems: one is lack of the verification for identification of user and field device;the other is that user and field device need exchange a key to encrypt sensitive data transmitted over the network.We propose a comprehensive authentication and key agreement framework that enables all connected devices in an ICS to mutually authenticate each other and establish a peer-to-peer session key. The framework combines two types of protocols for authentication and session key agreement: The first one is an asymmetric cryptographic key agreement protocol based on transport layer security handshake protocol used for Internet access, while the second one is a newly designed lightweight symmetric cryptographic key agreement protocol specifically for field devices. This proposed lightweight protocol imposes very light computational load and merely employs simple operations like one-way hash function and exclusive-or(XOR) operation. In comparison to other lightweight protocols, our protocol requires the field device to perform fewer computational operations during the authentication phase. The simulation results obtained using Open SSL demonstrates that each authentication and key agreement process in the lightweight protocol requires only 0.005 ms. Our lightweight key agreement protocol satisfies several essential security features, including session key secrecy, identity anonymity, untraceability, integrity, forward secrecy, and mutual authentication. It is capable of resisting impersonation, man-in-the-middle, and replay attacks. We have employed the Gong-NeedhamYahalom(GNY) logic and automated validation of Internet security protocols and application tool to verify the security of our symmetric cryptographic key agreement protocol.

To improve the atomic utilization of metals and reduce the cost of industrialization,the one-step total monoatomization of macroscopic bulk metals,as opposed to nanoscale metals,is effective.In this study,we used a thermal diffusion method to directly convert commercial centimeter-scale Ni foam to porous Ni single-atom-loaded carbon nanotubes(CNTs).As expected,owing to the coating of single-atom on porous,highly conductive CNT carriers,Ni single-atom electrocatalysts(Ni-SACs)exhibit extremely high activity and selectivity in CO_(2)electroreduction(CO_(2)RR),yielding a current density of>350 mA/cm^(2),a selectivity for CO of>91%under a flow cell configuration using a 1 M potassium chloride(KCl)electrolyte.Based on the superior activity of the Ni-SACs electrocatalyst,an integrated gas-phase electrochemical zero-gap reactor was introduced to generate a significant amount of CO current for potential practical applications.The overall current can be increased to 800 mA,while maintaining CO Faradaic efficiencies(FEs)at above 90%per unit cell.Our findings and insights on the active site transformation mechanism for macroscopic bulk Ni foam conversion into single atoms can inform the design of highly active single-atom catalysts used in industrial CO_(2)RR systems.

Simultaneous determination of five plant hormones in cotton leaves using QuEChERS combined with HPLC‒MS/MS

Background Plant hormones profoundly influence cotton growth,development,and responses to various stresses.Therefore,there is a pressing need for an efficient assay to quantify these hormones in cotton.In this groundbreaking study,we have established QuEChERS-HPLC‒MS/MS method,for the simultaneous detection of multiple plant hormones in cotton leaves,allowing the analysis and quantification of five key plant hormones.Results Sample extraction and purification employed 0.1%acetic acid in methanol and C18 for optimal recovery of plant hormones.The method applied to cotton demonstrated excellent linearity across a concentration range of 0.05–1 mg・L−1,with linear regression coefficients exceeding 0.99.The limits of quantification(LOQs)were 20μg・kg−1 for GA3 and 5μg・kg−1 for the other four plant hormones.Recovery rates for the five plant hormones matrix spiked at levels of 5,10,100,and 1000μg・kg−1 were in the range of 79.07%to 98.97%,with intraday relative standard deviations(RSDs)ranging from 2.11%to 8.47%.The method was successfully employed to analyze and quantify the five analytes in cotton leaves treated with plant growth regulators.Conclusion The study demonstrates that the method is well-suited for the determination of five plant hormones in cotton.It exhibits excellent selectivity and sensitivity in detecting field samples,thus serving as a robust tool for indepth research into cotton physiology.
