Interplay between post-translational cyclooxygenase-2 modifications and the metabolic and proteomic profile in a colorectal cancer cohort

BACKGROUND Colorectal cancer(CRC) is the second most common cause of cancer death worldwide. It is broadly described that cyclooxygenase-2(COX-2) is mainly overexpressed in CRC but less is known regarding post-translational modifications of this enzyme that may regulate its activity, intracellular localization and stability. Since metabolic and proteomic profile analysis is essential for cancer prognosis and diagnosis, our hypothesis is that the analysis of correlations between these specific parameters and COX-2 state in tumors of a high number of CRC patients could be useful for the understanding of the basis of this cancer in humans.AIM To analyze COX-2 regulation in colorectal cancer and to perform a detailed analysis of their metabolic and proteomic profile.METHODS Biopsies from both healthy and pathological colorectal tissues were taken under informed consent from patients during standard colonoscopy procedure in the University Hospital of Bellvitge(Barcelona, Spain) and Germans Trias i Pujol University Hospital(Campus Can Ruti)(Barcelona, Spain). Western blot analysis was used to determine COX-2 levels. Deglycosylation assays were performed in both cells and tumor samples incubating each sample with peptide N-glycosidase F(PNGase F). Prostaglandin E2(PGE2) levels were determined using a specific ELISA. 1 H high resolution magic angle spinning(HRMAS) analysis was performed using a Bruker AVIII 500 MHz spectrometer and proteomic analysis was performed in a nano-liquid chromatography-tandem mass spectrometer(nano LC-MS/MS) using a QExactive HF orbitrap MS.RESULTS Our data show that COX-2 has a differential expression profile in tumor tissue of CRC patients vs the adjacent non-tumor area, which correspond to a glycosylated and less active state of the protein. This fact was associated to a lesser PGE2 production in tumors. These results were corroborated in vitro performing deglycosylation assays in HT29 cell line where COX-2 protein profile was modified after PNGase F incubation, showing higher PGE2 levels. M

The recovery and utilization of carbon dioxide(CO_(2))is the key to achieve the targets of peak carbon dioxide emissions and carbon neutrality.The Na-CO_(2)battery made with cheap alkali metal sodium and greenhouse gas CO_(2)is an effective strategy to consume CO_(2)and store clean renewable energy.However,the liquid electrolyte volatilization in the open battery system and inevitable dendrite growth restrict the application of Na-CO_(2)batteries.In this work,magnesium-doped Na_(3)Zr_(2)Si_(2)PO_(12)(NZSP)was studied as a solid electrolyte for solid-state Na-CO_(2)batteries.The ionic conductivity of Na_(3.2)Zr_(1.9)Mg_(0.1)Si_(2)PO_(12)reaches 1.16 mS cm^(−1)at room temperature by replacing Zr ions in Na_(3.2)Zr_(1.9)Mg_(0.1)Si_(2)PO_(12)with Mg ions,and the structural changes are analyzed by neutron powder diffraction.The composite electrolyte consisting of highly conductive Na_(3.2)Zr_(1.9)Mg_(0.1)Si_(2)PO_(12)and high processability poly(vinylidene fluoride-co-hexafluoropropylene)(PVDF-HFP)is utilized for the first time to assemble a solid-state Na-CO_(2)battery.The cell shows a full discharge capacity of 7720 mAh g^(−1)at 200 mA g^(−1).The middle gap voltage is lower than 2 V after 120 cycles at 200 mA g^(−1)and at a cut-off capacity of 500 mAh g^(−1).This work demonstrates a promising strategy to design high-performance solid-state Na-CO_(2)batteries.

Robotic surgery for rectal cancer: Current immediate clinical and oncological outcomes

Laparoscopic rectal surgery continues to be a challenging operation associated to a steep learning curve. Robotic surgical systems have dramatically changed minimally invasive surgery. Three-dimensional, magnified and stable view, articulated instruments, and reduction of physiologic tremors leading to superior dexterity and ergonomics. Therefore, robotic platforms could potentially address limitations of laparoscopic rectal surgery. It was aimed at reviewing current literature on short-term clinical and oncological(pathological) outcomes after robotic rectal cancer surgery in comparison with laparoscopic surgery. A systematic review was performed for the period 2002 to 2014. A total of 1776 patients with rectal cancer underwent minimally invasive robotic treatment in 32 studies. After robotic and laparoscopic approach to oncologic rectal surgery, respectively, mean operating time varied from 192-385 min, and from 158-297 min; mean estimated blood loss was between 33 and 283 mL, and between 127 and 300 mL; mean length of stay varied from 4-10 d; and from 6-15 d. Conversion after robotic rectal surgery varied from 0% to 9.4%, and from 0 to 22% after laparoscopy. There was no difference between robotic(0%-41.3%) and laparoscopic(5.5%-29.3%) surgery regarding morbidity and anastomotic complications(respectively, 0%-13.5%, and 0%-11.1%). Regarding immediate oncologic outcomes, respectively among robotic and laparoscopic cases, positive circumferential margins varied from 0% to 7.5%, and from 0% to 8.8%; the mean number of retrieved lymph nodes was between 10 and 20, and between 11 and 21; and the mean distal resection margin was from 0.8 to 4.7 cm, and from 1.9 to 4.5 cm. Robotic rectal cancer surgery is being undertaken by experienced surgeons. However, the quality of the assembled evidence does not support definite conclusions about most studies variables. Doped and undoped TiCh nanoparticles were prepared by Stober method and thermally treated at 600 ℃.The effect of Nd^（3＋） ion on the structure and micro structure of anatase-phase TiCh nanocrystals was studied by Rietveld refinement method using X-ray powder diffraction data.Bond lengths,bond angles,and edges distances were analyzed.The phase formation was confirmed by high-resolution transmission electron microscopy.The adjustment of Ti-0 bond length induced by the addition of Nd^（3＋） ions,reduced the octahedral distortion and altered the octahedral array in the anatase-phase TiCh nanocrystal.The changes of structure and microstructure were mainly observed for TiCh nanoparticles doped with 0.1 at.%of Nd^（3＋） ions and attributed to the cationic substitution of Ti^（4＋） ions which promoted changes in the density of states and gap band of TiCh.The dopant insertion resulted in a better structural stability of the nanocrystals that enhanced their charge transference and photocatalytic efficiency.

This proposal aims to assess the market introduction of advanced technologies for the production of 2nd generation solid biofuels, specifically technologies for the production of briquettes and pellets from agro-industrial wastes. The development of this project will evaluate the socio-environmental and techno-economical feasibility and use of 2nd generation solid biofuels in the CMR (Campinas Metropolitan Region). The successful introduction of second generation briquettes and pellets to market depends, mainly, on two aspects: logistics in supply chains which generate waste, and the efficiency of production technologies. The study of logistics (supply chain) is based on survey data of the main productive supply chains, analysis, and modeling to optimize the facility location in the network for each case. The evaluation of the efficiency of production technology is provided by testing specially designed waste compacting devices, and comparing these results with the resulting power consumption during the production, in demonstration-scale, of a round of briquettes. The costs and consumption during the demonstration-scale production of briquettes are used for validation and correction of an optimization model. This project was approved in late 2012 with a period of two years for its implementation. Later in 2013, it was decided also to extend its implementation to the Metropolitan Region of Manaus, Amazon. Due to its recent beginning, the results shown here are only preliminary.

BACKGROUND The association between digoxin and mortality is an unclear issue.In older patients with atrial fibrillation(AF),where use of digoxin is frequent,the evidence of its safety is scarce.Our aim is to assess the safety of digoxin in nonagenarian patients with AF.METHODS We evaluated data from 795 nonagenarian patients with non-valvular AF from the Spanish Multicenter Registry.We analyzed the relationship between digoxin and all-cause mortality with the Cox proportional-hazards model.RESULTS Follow-up was 27.7±18.3 months.Mean age was 92.5±3.8 years,and 71%of nonagenarian patients were female.Digoxin was not associated with increased risk of mortality[adjusted hazard ratio(aHR)=1.16,95%CI:0.96−1.41,P=0.130].However,we found a significant increase in mortality in the subgroup with estimated glomerular filtration rate(eGFR)<30 mL/min per 1.73 m^(2)(aHR=2.01,95%CI:1.13−3.57,P=0.018),but not in the other subgroups of eGFR(30−59 mL/min per 1.73 m^(2) and≥60 mL/min per 1.73 m^(2)).When exploring the risk of mortality according to sex,male subgroup was associated with an in-crease in mortality(aHR=1.48,95%CI:1.02−2.14,P=0.041).This was not observed in females subgroup(aHR=1.03,95%CI:0.81−1.29,P=0.829).Based on the presence or absence of heart failure,we did not find significant differences(aHR=1.20,95%CI:0.87−1.65,P=0.268 vs.aHR=1.15,95%CI:0.90−1.47,P=0.273,respectively).CONCLUSIONS In our large registry of nonagenarian patients with AF,we did not find an association between digoxin and mortality in the total sample.However,in the subgroup analyses,we found an increase in mortality with the use of digoxin in men and in patients with an eGFR<30 mL/min per 1.73 m^(2).

Radical Vaginal Hysterectomy and Trachelectomy in Early-Stage Cervical Cancer

Objective: To communicate a minimally invasive technique for surgical handling of early-stage cervical cancer and its results. Methods: 110 patients with cervical cancer in stages IA2 and IB1, all of them operated in a period of 5 years in both hospitals, are presented. Laparoscopic systemic pelvic lymphadenectomy with radical vaginal hysterectomy or radical vaginal trachelectomy was performed to patients, with the exception of those patients who had compromised nodes detected in contemporary biopsy. Results: Between April 2008 and May 2013, 110 patients were submitted to this technique. 15 patients had their surgery aborted: 13 presented positive nodes for carcinoma in contemporary biopsy and 2 had extensive cervical compromise when performing radical vaginal hysterectomy or trachelectomy. Analysis of the remaining 95 cases shows an average age of 43.9 years (26 - 61), all of them had given birth before, 23 (21%) of them through C-section. BMI averaged 30.5 and 31 (28.2%) had cone surgery performed previously. Average duration of surgery was of 220 minutes. Postoperative hospitalization averaged 3.1 days. Bleeding volume was estimated at 125 cc and one patient required blood transfusion. En 25 patients’ uterine annexes were kept and all of them were suspended by means of laparoscopy. On average, 25.4 pelvic nodes were obtained. Complications 13.6% with eight patients suffered bladder injury, two had rectovaginal fistula, 3 patients requires surgical repair of ureteral obstruction, two patients present thromboembolic disease. The disease-free and overall survivals are consistent with reports in the literature. Conclusion: We believe that handling patients with this technique is possible and has the advantages of vaginal and laparoscopic surgery with minimal complications.
