Amyotrophic lateral sclerosis (AlS) is a fatal motor neuron disease typically leading to death within 5 years from symptom onset. AlS familial forms are associated with mutations in several genes, including Superoxide Dismutase 1 (SOD1) and Fused in Sarcoma (FUS). Although genes linked to AlS participate in disparate biological processes, AlS genetic variants largely trigger shared pathogenic events such as oxidative stress, protein aggregation and defects in RNA processing. Moreover, AlS patients show systemic hypermetabolism that leads to increased energy expenditure at rest and thus weight loss during the disease course.1 AlS hypermetabolic phenotype and weight loss have been extensively characterized in mice bearing the G93A substitution in SOD1 protein (SOD1-G93A), which exhibit skeletal-muscle metabolic reprogramming before disease onset.

SARS-CoV-2 virus is responsible for the current worldwide coronavirus disease 2019 (COVID-19) pandemic, infecting millions of people and causing hundreds of thousands of deaths. Understanding the antibody response to SARS-CoV-2 is crucial for the development of vaccines, therapeutics and public health interventions. However, lack of consistency in methods used to monitor antibody response to SARS-CoV-2 leaves some uncertainty in our fine understanding of the human antibody response mounted following SARS-CoV-2 infection. We developed a peptide-based enzyme-linked immunosorbent assay (ElISA) by selecting 7 synthetic peptides from the spike, membrane, and nucleocapsid protein sequences of SARS-CoV-2, which effectively detects the antibody response mounted by all COVID-19 convalescent tested. Strikingly, the assay shows a profound difference in antibody response among individual subjects, which may have a significant impact on disease severity. Together, our results define an efficient and specific serological assay to consistently measure the antibody response following SARS-CoV-2 infection, as well as help the design of vaccine and therapeuticals for prevention and treatment of COVID-19.

Eco-toxicity investigation of polymer materials was considered extremely necessary for their potential menace,which was widely use as mulching materials in *** this study,polyethylene(PE),polystyrene(PS)and synthetic biomaterials-Ecoflex and cellulose were applying into soil cultivated with two potential indicator plants species:oat(A v ena sati v a)and red radish(Raphanus sativum).Variety of chemical,biochemical parameters and enzyme activity in soil were proved as effective approach to evaluate polymers phytotoxicity in plant-soil *** F-value of biomass,pH,heavy metal and electoral conductivity of Raphanus behaved significant different from *** analysis results indicated biodegradation was fast in PE than PS,besides,heavy metals were dramatically decrease in the end implied the plant absorption may help decrease heavy metal *** increase value at T2 of Dehydrogenase activity(0.84 higher than average value for Avena&0.91 higher for Raphanus),Metabolic Index(3.12 higher than average value for Avena&3.81 higher for Raphanus)means during soil enzyme activity was promoted by biodegradation for its heterotrophic organisms’energy transportation was *** analysis was carried on Biplot PC1(24.2%of the total variance),PC2(23.2%of the total variance),versus PC3(22.8%of the total variance),which indicated phosphatase activity and metabolic index was significant correlated,and high correlation of ammonium and protease ***,the effects were more evident in Raphanus treatments than in Avena,suggesting the higher sensitivity of Raphanus to polymers treatment,which indicate biodegradation of polymers in Raphanus treatment has produced intermediate phytotoxic compounds.

Programmed death-ligand-1(PD-l1)plays a crucial role in the suppression of the antitumor immune response upon interaction with programmed cell death protein-1(PD-1)on cytotoxic T ***-l1 is constitutively expressed on antigen-presenting cells or tumor ***,PD-l1 has been detected by immunohistochemistry in a variety of tumors,including malignant pleural mesothelioma(MPM),in which it has emerged as a predictive biomarker for PD-1/PD-l1 immune checkpoint blockers(ICBs).1,2 PD-l1 expression on tumor cells can be upregulated by several factors including the interferon gamma(IFN-γ)cytokine produced by tumor-infiltrating lymphocytes(TIls).3 In MPM cells,it has been reported that IFN-γupregulates PD-l1 mRNA expression due to the activation of the interferon regulatory factor 1(IRF1)transcription factor,4 but no data have been shown regarding cell surface PD-l1 that is functionally relevant for its contact with PD-1-positive cells.

BACKGROUND Polyomavirus-associated nephropathy is a leading cause of kidney allograft failure. Therapeutic options are limited and prompt reduction of the net state of immunosuppression represents the mainstay of treatment. More recent application of aggressive screening and management protocols for BK-virus infection after renal transplantation has shown encouraging results. Nevertheless,long-term outcome for patients with BK-viremia and nephropathy remains obscure. Risk factors for BK-virus infection are also *** To investigate incidence, risk factors, and outcome of BK-virus infection after kidney *** This single-centre observational study with a median follow up of 57(31-80) mo comprises 629 consecutive adult patients who underwent kidney transplantation between 2007 and 2013. Data were prospectively recorded and annually reviewed until 2016. Recipients were periodically screened for BK-virus by plasmaquantitative polymerized chain reaction. Patients with BK viral load ≥ 1000 copies/ml were diagnosed BK-viremia and underwent histological assessment to rule out nephropathy. In case of BK-viremia, immunosuppression was minimized according to a prespecified protocol. The following outcomes were evaluated: patient survival, overall graft survival, graft failure considering death as a competing risk, 30-d-event-censored graft failure, response to treatment,rejection, renal function, urologic complications, opportunistic infections, newonset diabetes after transplantation, and malignancies. We used a multivariable model to analyse risk factors for BK-viremia and *** BK-viremia was detected in 9.5% recipients. Initial viral load was high(≥ 10000 copies/ml) in 66.7% and low(l) in 33.3% of these ***-associated nephropathy was diagnosed in 6.5% of the study population. Background:The prognosis of colorectal cancer depends on the number of positive lymph nodes(lNþ)and the total number of lymph nodes resected(rlN).This represents the lymph-node ratio(lNR).The aim of our study is to assess how the length of the resected specimen(Rl)influences the prognostic values of the ***:We conducted a retrospective study of all the patients operated on for colorectal cancer from 2000 to 2015 at our *** details were *** total number of rlN,the number of lNþ,and the lNR were calculated and measured against the *** receiver-operating characteristic(ROC)curve of patients with lNþwas ***:Of the 670 patients included in our study,337 were men(50.3%)and the mean age was 69.2 *** correlation with prognosis of the lNR is greater than that of the lNR adjusted to Rl(lNR/Rl),both in subjects with positive nodes(n=312)and in all cases(n=670).The lNR presents a higher prognostic value than lNR/Rl and Rl in patients with lNþexcept for metastatic recurrence,for which the predictive value appears slightly higher for lNR/*** statistical significance of the maximal divergence in Kaplan–Meier survival plots was demonstrated for the lNR(P=0.043),not for lNR/Rl(P=0.373)and Rl alone(P=0.314).Conclusion:An increase in Rl causes an increase in the number of harvested lymph nodes without affecting the number of lNþ,thus representing a confounding factor that could alter the prognostic value of the *** larger-scale studies are needed to confirm these findings.

Porous biomaterials which provide a structural and biological support for cells have immense potential in tissue engineering and cell-based therapies for tissue *** biomaterials that can host endothelial cells represent promising tools for the vascularization of engineered ***-dimensional collagen scaffolds possessing controlled architecture and mechanical stiffness are obtained through freeze–drying of collagen suspensions,followed by chemical cross-linking which maintains their ***,cross-linking scaffolds renders their biological activity suboptimal for many cell types,including human umbilical vein endothelial cells(HUVECs),by inhibiting cell–collagen ***,we have improved crucial HUVEC interactions with such cross-linked collagen biomaterials by covalently coupling combinations of triple-helical peptides(THPs).These are ligands for collagen-binding cell-surface receptors(integrins or discoidin domain receptors)or secreted proteins(SPARC and von Willebrand factor).THPs enhanced HUVEC adhesion,spreading and proliferation on 2D collagen *** grafted to 3D-cross-linked collagen scaffolds promoted cell survival over seven *** study demonstrates that THP-functionalized collagen scaffolds are promising candidates for hosting endothelial cells with potential for the production of vascularized engineered tissues in regenerative medicine applications.

The incidence of paediatric inflammatory bowel disease(PIBD) has dramatically increased in the last 20 years. Although first reported in mid 1970s',diagnostic laparoscopy has started to be routinely adopted in paediatric surgical practice since late 1990s'. Minimally invasive surgery was first limited to diagnostic purposes. After 2002 it was also applied to the radical treatment of PIBD,either Crohn's disease(CD) or Ulcerative colitis. During the last decade minimally invasive approaches to PIBD have gained popularity and have recently became the "gold standard" for the treatment of such invalidating and troublesome chronic diseases. The authors describe and track the historical evolution of minimally invasive surgery for PIBD and address all available opportunities,including most recent advancements such as robotic surgery,single port approaches and minimally invasive treatment of perianal fistulising CD. A systematic review of all series of PIBD treated with minimally invasive approaches published so far is provided in order to determine the incidence and type of patients' complications reported up to present days. The authors also describe their experience with minimally invasive surgery for PIBD and will report the results of 104 laparoscopic procedures performed in a series of 61 patients between January 2006 and December 2014.
