落叶松幼苗在其人工林内难以更新。为探讨红皮云杉和青海云杉幼苗是否具有在落叶松人工林内成功定植并替代落叶松更新的潜力,采用红皮云杉和青海云杉幼苗在不同落叶松人工林(样地Ⅰ:1 300株·hm^(-2)的平地,样地Ⅱ,660株·hm^(-2)的平地;样地Ⅲ,330株·hm^(-2)的平地;样地Ⅳ,660株·hm^(-2)的坡地)下进行人工更新。结果表明:1)样地Ⅰ和样地Ⅱ更适宜红皮云杉和青海云杉幼苗的存活,且同一样地内,红皮云杉幼苗的成活率均高于青海云杉的;2)同一样地内,红皮云杉幼苗的地上生物量均高于青海云杉的,并且在样地Ⅰ和样地Ⅱ红皮云杉的地上生物量显著高于青海云杉的;3)在不同落叶松人工林样地,红皮云杉和青海云杉幼苗的1级根均有较好的外生菌根侵染;4)红皮云杉幼苗的1级根直径受环境影响较大,而青海云杉幼苗的1级根皮层厚度受环境影响较大。同一样地内,2种云杉幼苗的1级根解剖结构除在样地Ⅳ表现出明显的差异外,在其它样地基本无显著差异。红皮云杉和青海云杉幼苗均适宜在落叶松人工林内存活,但相对高密度的林分更适宜云杉幼苗的生长,且红皮云杉比青海云杉更具生长优势。

Gastro-protective effect of Ziziphus abyssinica root extracts in ethanol-induced acute ulcer in Wistar rats

Objective: To evaluate the gastro-protective effect of the aqueous, methanol and hexane root extracts of Ziziphusabyssinica on ethanol induced gastric ulcer in Wistar rats as models. Methods: Seventy rats were divided into seven groups of ten rats each: control group, ulcer control group, standard control group (cimetidine 100 mg/kg body weight), aqueous, methanol, ethyl acetate and hexane extract groups. They were administered the extracts at 300 and 600 mg/kg for two weeks before ulcer was induced, and the protective effect of all extracts determined. Results:Histological changes in gastric tissue were evaluated. Pretreatment withZiziphus abyssinica extracts showed significant (P < 0.05) gastro-protective effect with aqueous extract having the highest gastro-protective effect (95% and 93% at 300 and 600 mg/kg extract respectively). Methanol extract had 66.6% and 65.5% protection at 300 and 600 mg/kg respectively. The percentage protection in ethyl acetate extract treated group was 51% and 45% respectively. The group treated with hexane had 23.8% and 28.6% protection at 300 and 600 mg/kg, respectively. Histological study showed that pretreatment with aqueous extract resulted in the preservation of the functional cyto-architecture of the entire mucosa with little pathological changes, compared to other extracts. Conclusions:The results of this study indicated that aqueous extract is effective against induced gastric ulcer.

Use of Inhaled PGE1 to Improve Diastolic Dysfunction, LVEDP, Pulmonary Hypertension and Hypoxia in ARDS—A Randomised Clinical Trial

Introduction: We wished to see the effects of inhaled PGE1 on diastolic dysfunction, left ventricular end diastolic pressure (LVEDP), pulmonary hypertension and hypoxia in ARDS patients. Methods: This is a randomized, prospective, clinical trial conducted in the main adult intensive care unit of a tertiary care University hospital. A total of 67 patients were recruited. Inclusion criteria included all adult patients with a P/F ratio 35 mmHg on Pulmonary artery catheter or suspected on clinical grounds. A transthoracic echo was performed to record the diastolic function, LVEDP and Pa pressures. Subsequently patients were randomized by a block computerized randomization to either cases (n = 34) or controls (n = 33). Cases received nebulised PGE1 over 30 minutes in the ICU and normal saline was administered to controls blindly. Following this the echo and arterial blood gases were repeated. Our primary outcomes were an improvement in diastolic function and P/F ratio of greater than 20% and a decrease in pulmonary pressure and LVEDP of >20%. Results: At baseline, mean diastolic dysfunction was grade II, with a mean LVEDP of >15 and the PaO2/FiO2 ratio was 148.38 ± 60.05 with a mean pulmonary artery pressure of 81.35 ± 16.91. Inhaled PGE1 was followed by an improvement in diastolic dysfunction (grade I, p = 0.001) with a resulting improvement in LVEDP (12 +/? 2, p = 0.001) as well as Pa pressures (97.09 ± 30.06, p = 0.04) and a non significant improvement in PaO2/FiO2 ratio (161.45 ± 77.52, p = 0.21). There were no side effects observed in any patients. Conclusion: Our study shows that there is a significant improvement in diastolic dysfunction, LVEDP and Pa pressures after administration of nebulised PGE1, and an improvement although non-significant in hypoxia in ARDS patients. The trial was registered with *** (NCT00314548) and funded by the Pakistan medical research council.

Extracellular sulfatase-2(Sulf-2)influences receptor-ligand binding and subsequent signaling by chemokines and growth factors,yet Sulf-2 remains unexplored in inflammatory cytokine signaling in the context of rheumatoid arthritis(RA).In the present study,we characterized Sulf-2 expression in RA and investigated its potential role in TNF-α-induced synovial inflammation using primary human RA synovial fibroblasts(RASFs).Sulf-2 expression was significantly higher in serum and synovial tissues from patients with RA and in synovium and serum from hTNFtg *** sequencing analysis of TNF-α-stimulated RASFs showed that Sulf-2 siRNA modulated~2500 genes compared to scrambled *** Pathway Analysis of RNA sequencing data identified Sulf-2 as a primary target in fibroblasts and macrophages in *** blot,ELISA,and qRT‒PCR analyses confirmed that Sulf-2 knockdown reduced the TNF-α-induced expression of ICAM1,VCAM1,CAD11,PDPN,CCL5,CX3CL1,CXCL10,and *** studies identified the protein kinase C-delta(PKCδ)and c-Jun N-terminal kinase(JNK)pathways as key in the TNF-α-mediated induction of proteins related to cellular adhesion and *** of Sulf-2 abrogated TNF-α-induced RASF ***-2 knockdown with siRNA and inhibition by OKN-007 suppressed the TNF-α-induced phosphorylation of PKCδand JNK,thereby suppressing the nuclear translocation and DNA binding activity of the transcription factors AP-1 and NF-κBp65 in human ***,Sulf-2 expression positively correlated with the expression of TNF receptor 1,and coimmunoprecipitation assays demonstrated the binding of these two proteins,suggesting they exhibit crosstalk in TNF-α*** study identified a novel role of Sulf-2 in TNF-αsignaling and the activation of RA synoviocytes,providing the rationale for evaluating the therapeutic targeting of Sulf-2 in preclinical models of RA.

Citrus canker is a perilous disease caused by Xanthomonas axonopodis pv. citri (Xac) which is hardly manageable. Till now, chemicals used against Xac either proved less effective against the disease or hazardous for environment. In this study, we investigated how phytohormones such as Salicylic acid (SA, 3 mM), Jasmonic acid (JA, 0.3 mM), 3-indolacetonitrile (IAN, 0.6 mM), Nicotinic acid (NA, 5 mM), Folic acid (FA, 0.5 mM) influenced canker disease. Disease severity and lesion diameter were significantly mitigated by exogenous application of these phytohormones compared with water (control) accompanied by the increase of latent period. However, these phytohormones did not show any in vitro antimicrobial properties against Xac. Concomitant with boosted resistance, phytohormones treated leaves showed the higher level of phenyl ammonia lyase (PAL) activity. Besides, total phenolic compounds (TPC) and potassium contents were also higher in phytohormone treated plants compared with control. Phytohormones showed the significant and better performance on disease mitigation than control among which, SA showed the highest performance while JA and INA showed the moderate performance. However, NA and FA showed the lowest performance. This result recommended that application of some phytohormones had elicited some biochemical and physiological events which induced the resistance against Xac.

To evaluate the impact of different zinc application methods on yield and yield components of various wheat cultivars, a field experiment was carried out at Student’s Farm, Department of Agronomy, University of Agriculture Faisalabad, during winter 2014-2015. The experiment was laid out in RCBD (Randomized Complete Block Design) with factorial arrangement. In this experiment, all the treatments were replicated three times, using the net plot size of 1.8 m × 5.0 m. The experiment comprised of two factors. Factor A consists of two varieties: Faisalabad-2008 and Punjab-2011. While, Factor B consists of different methods of zinc application, control, zinc application in soil before planting 23 kg·ha-1, zinc foliar application, 4% ZnSO4 solution at two stages (tillering and stem elongation stage). The data regarding different parameters were collected and analyzed from the crop using standard procedures. Regarding the impact of zinc application method maximum plant height at maturity (103.6 cm), total number of tillers (564.67 m-2), spike length (10.83 cm), number of spikelets spike-1 (19.50), number of grains spike-1 (50.36), 1000-grain weight (34.16 g), biological yield (11.93 t·ha-1), grain yield (6.00 t·ha-1) and harvest index (39.25%) were recorded in treatment where zinc was applied both in the soil before planting and by foliar application on later growth stages. Similarly, grain zinc contents (33.11 mg·kg-1), grain protein contents (10.1%) and grain carbohydrate contents (64.23%) were also observed in the treatment where zinc was applied both in the soil before planting and by foliar application on later growth stages, which is better than all other treatments. In case of wheat varieties, Faisalabad-2008 performed better than Punjab-2011, with maximum method maximum total number of tillers (460.67 m-2), spike length (9.70 cm), number of grains spike-1 (48.80), 1000-grain weight (33.81 g), biological yield (10.32 t·ha-1), grain yield (5.0 t·ha-1) and harvest index
