Endogenous stress and exogenous toxicants(chemicals and UV light) alter genetic information either directly or indirectly through the production of reactive oxygen species(ros), thereby driving genomic instability in cells and promoting tumorigenesis. All living cells try to faithfully preserve and transmit their genomic information from one generation to the next using DNA repair mechanisms to repair oxidative DNA damage to prevent cancer or premature aging. Oxidative DNA damage comprises a mixture of DNA lesions including base damage, DNA single strand breaks(ssBs), and DNA double strand breaks(DsBs). This review summarizes some of the studies on DNA damage response at a defined genome locus that are performed by students from the Tsinghua University school of Medicine and the school of Medicine of Central south University(Xiangya Hospital) at the University of Pittsburgh school of Medicine. A summary of their work highlights the continuous contribution of the students to a particular research program and exemplifies the achievements of this China-U.s. collaborative training program.

Corynebacterium pseudotuberculosis is an infectious agent that occurs in small ruminants causing caseous lymphadenitis, and more rarely in humans causing lymphadenitis and pneumonia. The breeding small ruminants have great economic importance in Brazil. Rural farm workers and veterinary students who acquired this disease suffered from weakening symptoms for weeks, and the identification of the etiological agent was time-consuming and complex. Due to the low prevalence of case records, there is probably no available commercial diagnostic kit for C. pseudotuberculosis infection in humans. This study aimed to describe human seroreactivity to secreted antigens from C. pseudotuberculosis. Reactivity of serum from farm workers (n = 14), individuals who work with the bacillus at laboratory (n = 8) or individuals without contact (n = 25) was tested with secreted proteins from PAT10 strain of C. pseudotuberculosis by Western blotting. samples of all (100%) farm workers showed reactivity to 31 kDa, 71 kDa and 164 kDa proteins, while laboratory workers showed 87.5%, 62.5 % and 37.5%, and no-contact 20%, 0% and 16%, respectively. All sera recognized the 275 kDa protein. Our data suggest that C. pseudotuberculosis secreted proteins are antigenic in humans and the recognition profiles allowed the identification of individuals with and without prior contact with this bacillus. This is the first paper which describes human reactivity to C. pseudotuberculosis in serum samples of workers in Brazil.

Age-related macular degeneration（AMD） is a leading cause of blindness and is becoming a global crisis since affected people will increase to 288 million by ***,age,diabetes,gender,obesity,hypertension,race,hyperopia,iris-color,smoking,sun-light and pyroptosis have varying roles in AMD,but oxidative stress-induced inflammation remains a significant driver of *** is a unique organ as it contains a remarkable oxygengradient that generates reactive oxygen species（ros） which upregulates inflammatory *** becomes a source of functional and morphological impairments in retinal pigment epithelium（RPE）,endothelial cells and retinal ganglion *** demonstrated that hydrogen sulfide（H2s） acts as a signaling molecule and that it may treat ***,we propose a novel hypothesis that H2s may restore homeostasis in the eyes thereby reducing damage caused by oxidative injury and *** H_2s has been shown to be a powerful antioxidant because of its free-radicals＇ inhibition properties in addition to its beneficial effects in age-relatedconditions,therefore,patients may benefit from H2s salubrious effects not only by minimizing their oxidant and inflammatory injuries to retina but also by lowering retinal glutamate excitotoxicity.

Objective Toxic metal ions have been implicated in the generation of reactive oxygen species (ros) and nitric oxide (NO). Metallothionines (MT) and plant flavonoids have been reported in the intervention against oxidative damage. We investigated the effect of zinc induced MT and green tea polyphenol (GTP) in reducing the oxidative responses induced by nickel and platinum. Methods Zinc (10 mg/kg b. wt, sc) was administered to rats twice at a gap of 24hrs and GTP (10 mg/100 mL in drinking water) was fed ad libitum for 8 days. Nickel chloride (150 umol/***, ip) and cisplatin (50 mmol/kg ***, sc) was administered to rats 24 h after Zn or GTP pre-treatment. Animals of all the groups were sacrificed 16 hrs after treatment and biochemical markers for toxicity were monitored. Results Zinc or GTP pre-treatment caused significant protection against nickel or cisplatin enhanced mortality in rats, and reduction in lipid peroxidation and NO. Conclusion it is proposed that inhibition of ros and NO by GTP and zinc may prove useful as a selective pharmacological agent in the amelioration of metal toxicity.

Difructose dianhydride Ⅳ(DFA-Ⅳ) is produced from levan, which is a natural polysaccharide that belongs to the fructan family, through the activity of levan fructotransferase(LF) derived from microorganisms. Recently, DFA-Ⅳ has been expected to have diverse applications in the food and medical industry. Here, we examined the potential application of DFA-Ⅳ for in vitro fertilization(ⅣF) in pigs. in the assessment of acrosomal integrity during incubation, intact acrosomal or viable spermatozoa were highly sustained in 0.1% or 0.25% DFA-Ⅳ(69.8%-70.8%,Pive oxygen species(ros) levels during sperm incubation decreased following the addition of DFA-Ⅳ, and 0.1 %-0.5% DFA-Ⅳ in particular significantly decreased ros production relative to that seen with no addition or 0.75% DFA-Ⅳ. Total fertilization(mono + polyspermic oocyte) rate was significantly higher in the addition of0.1% DFA-Ⅳ(94.2%) than with other concentrations(71.8%-86.7%, Psing reduced ⅣF times and lower sperm numbers, we found that addition of 0.1%-0.5% DFA-Ⅳ significantly increased the fertilization rate(Pilized oocytes treated with 0.1% DFA-Ⅳ exhibited higher embryonic development and blastocyst formation than those treated with other concentrations(Psequently, the addition of DFA-Ⅳ during ⅣF improved fertilization and embryonic development, suggesting the possible use of novel sugars for enhancement of assisted reproductive technology(ART) in mammals.

Although several artificial nanotherapeutics have been approved for practical treatment of metastatic breast cancer,their inefficient therapeutic outcomes,serious adverse effects,and high cost of mass production remain crucial ***,we developed an alternative strategy to specifically trigger apoptosis of breast tumors and inhibit their lung metastasis by using natural nanovehicles from tea flowers(TFENs).These nanovehicles had desirable particle sizes(131 nm),exosome-like morphology,and negative zeta ***,TFENs were found to contain large amounts of polyphenols,flavonoids,functional proteins,and *** experiments revealed that TFENs showed strong cytotoxicities against cancer cells due to the stimulation of reactive oxygen species(ros)*** increased intracellular ros amounts could not only trigger mitochondrial damage,but also arrest cell cycle,resulting in the in vitro anti-proliferation,anti-migration,and anti-invasion activities against breast cancer *** mice investigations demonstrated that TFENs after intravenous(i.v.)injection or oral administration could accumulate in breast tumors and lung metastatic sites,inhibit the growth and metastasis of breast cancer,and modulate gut *** study brings new insights to the green production of natural exosome-like nanoplatform for the inhibition of breast cancer and its lung metastasis via *** oral routes.
