Fluid Leakage in Submerged Offshore Pipeline: An Analysis of Oil Dispersion in Seawater

Leakages in oil pipelines can cause financial losses and several environmental damages, where large-scale offshore oil and gas exploration results in large releases of oil and gas into ocean waters. In the event of oil leakage, an immediate and adequate response is required to reduce environmental damage, such as containment barriers, for example, which depends on the agglomeration of oil particles, velocity and tendency to propagation. Thus, the understanding of the fluid flow behavior around of subsea pipeline at different depths is crucial. On the other hand, the knowledge of interfacial phenomena of immiscible liquids allows the process of adjective migration in submarine pipelines. Consequently, this science enables the prediction of the behavior and the geometric shape of the water-oil interface and provides a phenomenological foundation concerning the theories of perturbation, the stability criteria and mathematical modeling, as well as the flow patterns in the neighborhoods and submerged pipelines. From this perspective, this work aims to study the oil dispersion in sea water caused by leakage in a submerged pipeline. Here, a two-dimensional mathematical model based on the mass and linear momentum conservation equations and the standard k-ε turbulence model, was developed. The dynamic behavior of the oil and water phases is evaluated by pressure fields, surface velocity, volumetric fraction and velocity vectors. Simulation results show the presence of oil flux from the pipe to the marine stream and vice-versa. Further, the increase in oil velocity at the pipe inlet leads to an increase in pressure drop.

The Dog as a Risk Factor in Transmission of Visceral Leishmaniasis: A Review

Visceral leishmaniasis (VL) is a serious public health problem in brazil and worldwide. despite the euthanasia of dogs serologically positive, such action has not solved the endemic in brazil. A risk area for VL involves the presence of the vector, the occurrence of canines and the record of human cases. The factors that have favored the persistence and spread of VL in brazil are related to the predatory action of man on the environment, to the migratory movements and rural exodus, and a close coexistence of man and animals. Thus considering the epidemiological chain of VL, one fact seems clear: we do not know yet the true extent of the participation of the dog in the infectious cycle of VL. The clinical disease is an important indicator of the extent of the problem in an endemic area. The immunological events are complex and involve resistance and susceptibility to canine visceral leishmaniasis (CVL). One aspect in CVL is that many symptomatic dogs underwent xenodiagnosis not infect the vector. In such cases the pattern recognition receptors CD11b+, TLR2+, and NO present higher values for dogs with results in immunohistochemistry of skin and xenodiagnosis negative (IMH-/XENO-) than dogs with immunohistochemistry in skin and xenodiagnosis positive (IMH+/XENO+), suggesting that innate immunity modulates the competence of the dog to infect the vector. The organic response in CVL varies from individual to individual and within the same individual, with a strong evidence of being organ-specific. Only 20% of asymptomatic dogs have parasites in the skin and 15% are able to recover from clinical signs and eliminate the parasites spontaneously. In this review, we analyze the epidemiology of visceral leishmaniasis and clinical, immunological and pathological conditions that can contribute to understanding the role of dogs in transmission of visceral leishmaniasis.

Construction of Vector for Functional Analysis of the Intrinsically bent DNA in the ACE3 Replicator from Drosophila melanogaster

DNA replication is a crucial process for species survival, nevertheless it is not clear which factors define origin selection in multicellular eukaryotes. developmental gene amplification systems, such as the one described during ovarian follicles development in Drosophila melanogaster, are useful tools for studying of DNA replication process in these organisms. We previously described that the well characterized third chromosome amplified domain of D. melanogater displays three intrinsically bent DNA sites: b1, localized at an amplification control element (ACE3), b2 and b3, both localized at the preferential origin ori-β. This proposal aimed to construct a Drosophila transformation vector, which contains a short deletion at the ACE3, in order to reduce the intrinsically bent DNA site b1, and analyze the functional role of this site in the gene amplification process. Through a series of cloning steps, we obtained a big Parent vector derivative, containing a deletion at the positions 176-180 bp, inside the ACE3. The generation of a Drosophila transformation vector displays a reduced intrinsically bent DNA site in the third chromosome amplified domain, it will allow the analysis of the functional role of this curvature in developmental gene amplification, providing new insights on replication initiation in D. melanogaster and the function of intrinsically bent DNA sites.

A mutant library of the bacterium Xanthomonas citri subsp. citri strain 306 pathotype A (Xac), the causative agent of most aggressive Asiatic type A citrus canker, was screened regarding altered canker symptoms after inoculations into Citrus sinensis and Citrus limonia host leaves. Twenty-six mutants have shown phenotypic virulence changes and have respectively knocked out gene identified by sequencing. In vivo growth curves were obtained for nine mutants to quantify how the mutations could affect pathogen’s adaptability to growth inside and attack host plant infected tissue. Among identified genes in mutated strains, we could find those that until now had not been reported as being involved in Xac adaptation and/or virulence, such as predicted to encode for xylose repressor-like protein (XACΔxylR), Fe-S oxidoredutase (XACΔaslb), helicase IV (XACΔhelD), ubiquinol cytochrome c oxidoreductase iron-sulfur subunit (XACΔpetA), chromosome partitioning protein (XACΔparb) and cell division protein Ftsb (XACΔftsb), in addition to genes predicted to encode for hypothetical proteins. The new genes found in this study as being relevant to adaptation and virulence, improve the understanding of Xac fitness during citrus plant attack and canker symptoms development.

b-1 cells modulate the murine macrophage response to Leishmania major infection

AIM To investigate the modulatory effect of b-1 cells on murine peritoneal macrophages infected with Leishmania major(L. major) in *** Peritoneal macrophages obtained from bALb/c andbALb/c XID mice were infected with L. major and cultured in the presence or absence of b-1 cells obtained from wild-type bALb/c mice. Intracellular amastigotes were counted, and interleukin-10(IL-10) production was quantified in the cellular supernatants using an enzymelinked immunosorbent assay. The levels of the lipid mediator prostaglandin E2(PGE2) were determined using a PGE2 enzyme immunoassay kit(Cayman Chemical, Ann Arbor, MI), and the number of lipid bodies was quantified in the cytoplasm of infected macrophages in the presence and absence of b-1 cells. Culturing the cells with selective PGE2-neutralizing drugs inhibited PGE2 production and confirmed the role of this lipid mediator in IL-10 production. In contrast, we demonstrated that b-1 cells derived from IL-10 KO mice did not favor the intracellular growth of L. *** We report that b-1 cells promote the growth of L. major amastigotes inside peritoneal murine macrophages. We demonstrated that the modulatory effect was independent of physical contact between the cells, suggesting that soluble factor(s) were released into the cultures. We demonstrated in our co-culture system that b-1 cells trigger IL-10 production by L. major-infected macrophages. Furthermore, the increased secretion of IL-10 was attributed to the presence of the lipid mediator PGE2 in supernatants of L. major-infected macrophages. The presence of b-1 cells also favors the production of lipid bodies by infected macrophages. In contrast, we failed to obtain the same effect on parasite replication inside L. major-infected macrophages when the b-1 cells were isolated from IL-10 knockout mice. CONCLUSION Our results show that elevated levels of PGE2 and IL-10 produced by b-1 cells increase L. major growth, as indicated by the number of parasites

Hepatocellular carcinoma (HCC) is common and one of the most aggressive of all human cancers. Recent studies have indicated that miRNAs, a class of small noncoding RNAs that regulate gene expression post-transcriptionally, directly contribute to HCC by targeting many critical regulatory genes. Several miRNAs are involved in hepatitis b or hepatitis C virus replication and virus-induced changes, whereas others participate in multiple intracellular signaling pathways that modulate apoptosis, cell cycle checkpoints, and growth-factor-stimulated responses. When disturbed, these pathways appear to result in malignant transformation and ultimately HCC development. Recently, miRNAs circulating in the blood have acted as possible early diagnostic markers for HCC. These miRNA also could serve as indicators with respect to drug efficacy and be prognostic in HCC patients. Such biomarkers would assist stratification of HCC patients and help direct personalized therapy. Here, we summarize recent advances regarding the role of miRNAs in HCC development and progression. Our expectation is that these and ongoing studies will contribute to the understanding of the multiple roles of these small noncoding RNAs in liver tumorigenesis.

Acute Post-Activation Potentiation Effects of Heavy High-Pulls on Clean Performance

Aim The concept of post-activation potentiation(PAP)is still novel from a training perspective and lacks breadth relative to weightlifting *** The purpose of this study was to investigate the acute effects of a conditioning activity(clean high pulls)on the performance of a main activity(clean)after the conditioning *** Eleven volunteers who engaged in weightlifting(Male=5,Female=6)participated in two different testing ses-sions(experimental ***).baseline measurements were taken following the *** testing condition was performed and additional measurements were taken at 30 s,2 min,4 min,6 min,8 min,10 min,and 12 min into recovery.A linear position transducer was used to collect peak power,peak velocity,and displacement of the barbell during the *** 2×8 repeated measures ANOVAs were used to analyze the data along with a paired samples t test between baseline and"best"*** There was no interaction or main effect for condition(P>0.05).However,there was a significant main effect for time(Pbest"values were significantly(Pbaseline values(peak power:experimental=1354.76 ±521.24 W *** 1216.03 ±492.30 W;peak velocity:experimental=2.66 ±0.44 m/s *** 2.50 ±0.44 m/s;displacement:experimental=111.07 ± 12.09 cm *** 107.01 ± 11.32 cm).Conclusions The findings of this study are in partial agreement with previous literature where the warm-up was probably enough to elicit ***,recommendations for future studies could include utilizing a more elite population and to ensure all potential participants can squat at least 2 x their body weight.

Prevalence of the Nursing Diagnosis Lack of Adherence in People Living with AIDS

Introduction: In the 1990s, the development of highly active combination antiretroviral therapy in the treatment of AIDS was highlighted. The great benefit generated by the use of this treatment was the prolongation of survival of the people who got this disease, since it is no longer considered fatal, becoming a chronic condition. Objective: To identify the prevalence of the nursing diagnosis Lack of Adherence, its defining characteristics and related factors in people living with AIDS, to investigate the association between them and the prevalence rates. Methods: Cross-sectional study with 113 patients in a hospital in northeastern brazil. For data analysis, the test of Pearson chi-square and Fisher’s exact test were used, and also calculated the prevalence rates. Results: Lack of Adherence diagnosis was present in 69% of the sample investigated. The defining characteristics and related factors that are statistically associated with that diagnosis were: lack of adherence behavior, missing scheduled appointments, complications related to development, insufficient skills to implement the system and complex treatment system. The first two characteristics had the highest chances for the development of this diagnosis. Conclusion: The study showed that the diagnostic studied was prevalent in these patients and 3 characteristics and 2 factors showed significant association.

Effects of Ingested baccharis dracunculifolia D.C.(Asteraceae)Extract in the Liver of Prochilodus lineatus Fish

baccharis dracunculifolia, popularly known in brazil as “alecrim-do-campo”, is widely recognized for its therapeutic potential. The extract of its leaves is used for liver problems, stomach disorders and others. The objective of the present study was to perform a histochemical analysis of curimbata fish livers to evaluate the potential effects and risks of the ingestion of b. dracunculifolia. Thirty-two animals were divided into two experimental groups in duplicate: Control group (regular food) and b. dracunculifolia Treated group (food added with b. dracunculifolia). The fishes were collected on the 14th and 21st days after the treatment period of 21 days. The histological alterations were evaluated using the semiquantitative methods Mean Value of Alterations (MVA), Histopathological Alteration Index (HAI) and Image J®. HAI and MAV showed that the extract caused slight but statistically significant damages, widely distributed throughout the organ. The results showed significant hepatic alterations caused by the ingestion of b. dracunculifolia extract.

background: Primary effusion lymphoma (PEL) is a lymphoid proliferation related to Kaposi sarcoma herpesvirus 8/human herpesvirus 8 (KSHV/HHV8) that affects mainly human immunodeficiency virus (HIV) infected individuals but can also occur in other immunodeficiency settings. It is characterized by lymphomatous effusions in different serous body cavities without the presence of a detectable tumor mass. The diagnosis is challenging and the clinical outcomes are poor. Aim: The aim of this paper is to report a rare case of PEL in a man who have sex with women (MSW) with HIV-1/2 infection, history of visceral Kaposi sarcoma (KS) and the development of a seronegative arthritis previous to the lymphoproliferative disease diagnosis. PEL presented with ascites, was treated with high-dose chemotherapy and autologous stem cell transplantation, with a good clinical outcome. Case Presentation: We describe a case of a 48-year-old HIV-1/2-infected patient from a high HHV8 seroprevalent country, hospitalized following a three-month history of increased abdominal volume and general constitutional symptoms. Laboratory data revealed normocytic normochromic anemia and a high level of lactate dehydrogenase. A diagnostic paracentesis was performed with cytology compatible with high-grade b-cell lymphoma. Peritoneal fluid cytology showed large lymphoid cells expressing leucocyte-common antigen CD45 without expression of the CD20 antigen (b-lymphocytes) and positivity for HHV8 by immunocytochemical staining, compatible with the diagnosis of PEL.
