Efficient in vivo delivery of anti-inflammatory proteins to modulate the microenvironment of an injured spinal cord and promote neuroprotection and functional recovery is a great ***-modified messenger RNA(mRNA)has become a promising new modality that can be utilized for the safe and efficient delivery of therapeutic ***,we used lipid nanoparticle(LNP)-encapsulated human interleukin-10(hIL-10)-encoding nucleoside-modified mRNA to induce neuroprotection and functional recovery following rat spinal cord contusion *** administration of hIL-10 mRNA-LNP to rats led to a remarkable reduction of the microglia/macrophage reaction in the injured spinal segment and induced significant functional recovery compared to ***,hIL-10 mRNA treatment induced increased expression in tissue inhibitor of matrix metalloproteinase 1 and ciliary neurotrophic factor levels in the affected spinal segment indicating a time-delayed secondary effect of IL-105 d after *** results suggest that treatment with nucleoside-modified mRNAs encoding neuroprotective factors is an effective strategy for spinal cord injury repair.

Culture Optimization for Mass Production of <i>Rhizobium</i>Using Bioreactor Made of Readily Available Materials and Agitated by Air Flow

Large quantities of Rhizobium leguminosarum by conventional technique are a time-consuming and labor-intensive method of production. The selection of different types of nutrient media for the production of large amounts of rhizobium at low cost, determination of environmental regulators of culture medium and the effect of different levels of air flow into nutrition media have been investigated. Rhizobium cultured in modified air lift bioreactor as improved conventional system using Yeast Extract Mannitol Broth by assessing aeration and agitation in their submerged growth. For selection of suitable nutrient media for Rhizobium before mass production, YMA (Yeast Mannitol Agar), YsA (Yeast sucrose Agar), YMA + bromthymol blue (BTB), YMA + Congo red (CR) medium was tested. To optimize the growing conditions of different temperatures, pH levels and salt strengths were investigated. Three air flow speeds at the rate of 0.01, 0.05 and 0.1 VVM for oxygen supply were ensured. A thick off-white bacterial colony was observed on the YMA plate among different culture media. Room temperature (25˚span>C - 30˚span>C), neutral pH (7.0), low salinity (1%) was observed to be suitable among the environmental conditions for Rhizobium mass production. The maximum density of Rhizobium was observed using air supply of 0.1 VVM level in bioreactor system. The result seemed that the air supply and agitation was an important growth factor in submerged cultivation. This study provides useful information on large scale production of Rhizobium of interest employing low cost effective modified air lift bioreactor within less time.

Influence of Concentrate in the Diet on the Metabolic, Biochemical, and Endocrine Profile of Buffalo Heifers

The objective of this study was to evaluate the profile of the main serum biochemical variables in buffalo heifers receiving diets with different concentrations of concentrate. Twenty buffalo heifers of the Murrah breed, aged six to eight months, were randomly assigned to four groups, with five replicates each, to be adapted to levels of 20%, 40%, 60%, and 80% of concentrate in the diet, composed of sugarcane (bulk) and concentrated feed based on wheat, corn, and soybean meal. The experimental diets were constituted in the proportion “bulk: concentrate”, based on dry matter, of “80:20” (Treatment 1), “60:40” (Treatment 2), “40:60” (Treatment 3), and “20:80” (Treatment 4). Biomarkers were evaluated for energy, protein, mineral, and endocrine profiles, as well as enzymatic activity. A positive linear effect was observed for Final Body Weight. In the energy profile, there was a positive linear effect for total cholesterol and HDL cholesterol and a quadratic effect for plasma glucose. In the protein profile, a quadratic effect was observed for the urea: creatinine ratio and a positive linear effect for total protein and albumin. For the enzymatic activity, only the variable gamma-glutamyltransferase presented a quadratic effect. In the mineral profile, a quadratic effect was observed for calcium levels and a positive linear effect for phosphorus levels and negative for chlorine levels. In the endocrine profile, no statistical differences were observed. Diet with until 80% of concentrate does not negatively influence biochemical parameters that represent the profile of energy, protein, mineral and thyroid hormones for growing buffalo heifers and favors the performance of the animals.
