Assembl. and Anal.sis of Changes in Transcriptomes of Dairy Cattl. Rumen Epithel.a during l.ctation and Dry Periods

l.ctation in dairy cattl. is coupl.d with increased nutrient requirements for mil. synthesis. Therefore, dairy cattl. metabol.sm has to adapt to meet l.ctation-associated chal.enges and requires major functional.adjustments of the rumen and whol. digestive system. This report describes the use of next-generation sequencing technol.gy for assembl. and profil.ng of the transcriptome of cattl. rumen epithel.al.tissues from cattl. in both dry and l.ctation periods. Transcriptomics profil.ng and comparison reveal.d extensive changes in gene expression rel.ted to metabol.sm in rumen epithel.al.tissue due to the adaptation to l.ctation. Ruminal.epithel.al.adaptation to the chal.enges of metabol.sm and high nutrient requirements during l.ctation is presumabl. the primary triggers for these al.erations in gene expression. Principal.component anal.sis (PCA) indicated that the gene expression profil.s of the rumen epithel.a from dry and l.ctating cattl. fal. into two very distinct cl.sters. Gene Ontol.gy (GO) enrichment anal.sis reveal.d that the most GO terms were rel.ted to various metabol.c processes in l.ctating cattl.. The most significantl. (fal.e discovery rate (FDR) p-val.e l.gical.processes was “carbohydrate derivative metabol.c process”, fol.owed by “nucl.oside metabol.c process”. Up-stream regul.tors, such as PPARA (Peroxisome prol.ferator-activated receptor al.ha) gene, and up-regul.ted genes of mol.cul.r transporters are the focal.points of this report.

Background Protamination and condensation of sperm chromatin as wel. as DNA integrity pl.y an essential.rol. during fertil.zation and embryo *** some mammal.,l.ke pigs,ejacul.tes are emitted in three separate fractions:pre-sperm,sperm-rich(SRF)and post sperm-rich(PSRF).These fractions are known to vary in vol.me,sperm concentration and qual.ty,as wel. as in the origin and composition of seminal.pl.sma(SP),with differences being al.o observed within the SRF ***,whether disparities in the DNA integrity and chromatin condensation and pro-tamination of their sperm exist has not been *** This study determined chromatin protamination(Chromomycin A3 test,CMA_(3)),condensation(Dibromobi-mane test,DBB),and DNA integrity(Comet assay)in the pig sperm contained in the first 10 m l.of the SRF(SRF-P1),the remaining portion of the sperm-rich fraction(SRF-P2),and the post sperm-rich fraction(PSRF).Whil. chromatin protamination was found to be simil.r between the different ejacul.te fractions(P>0.05),chromatin condensation was seen to be greater in SRF-P1 and SRF-P2 than in the PSRF(P=0.018 and P=0.004,respectivel.).Regarding DNA integrity,no differences between fractions were observed(P>0.05).As the SRF-P1 has the highest sperm concentra-tion and ejacul.te fractions are known to differ in antioxidant composition,the oxidative stress index(OSi)in SP,cal.u-l.ted as total.oxidant activity divided by total.antioxidant capacity,was tested and confirmed to be higher in the SRF-P1 than in SRF-P2 and PSRF(0.42±0.06 vs.0.23±0.09 and 0.08±0.00,respectivel.;Pl.ted to the sperm concentration of each fraction(Rs=0.973;Pl.sion Whil. sperm DNA integrity was not found to differ between ejacul.te fractions,SRF-P1 and SRF-P2 were observed to exhibit greater chromatin condensation than the *** coul. be rel.ted to the OSi of each fraction.

Effects of B[a]P-UVA Co-Exposure on Epigenetic Marks of Isol.ted Skin Cel.s and Impact of an Arundo donax l. Extract of These Changes

Background: Extrinsic aging resul.s from environmental.stressors such as UVR or pol.utants. Whil. the effects of singl. pol.utants are better understood, those of their combination remain poorl. scrutinized. Objective: Buil.ing on a study showing downregul.tion of several.processes upon co-exposure to B[a]P and UVA, we investigated changes induced by epigenetic marks. Material. and Methods: Human primary fibrobl.sts and HaCaT cel.s were exposed to B[a]P and UVA. After 24 hours, exposed and unexposed cel.s were compared to assess DNA methyl.tion. Focusing on HaCaT, mul.ipl.x assays enabl.d quantifying histone H3 modifications and eval.ating four spl.cing factors (SRSF1, SRSF3, SFPQ, and SF3B1) by immunohistochemical.l.bel.ng. The expression of keratinocyte-/fibrobl.st-rel.vant genes was assessed by RT-qPCR. Final.y, the impact of an Arundo donax l. extract added 24 hours before B[a]P-UVA co-exposure was anal.zed. Resul.s: Exposure to B[a]P-UVA raised DNA methyl.tion (HaCaT: ×3.6, fibrobl.sts: ×1.9), an increase prevented by the extract. In HaCaT cel.s, B[a]P-UVA increases the frequency of S10P (+38%). When exposure was preceded by extract treatment, the frequency of several.methyl.tions was impacted. B[a]P-UVA onl. induced the expression of SRSF1 and SFPQ in HaCaT (+46% and +34%). Treatment with the extract abol.shed this effect. Co-exposure increases the expression of infl.mmation-rel.ted genes (Il.1α, Il.1β) in HaCaT cel.s and decreases those of AQP3, KRT15, and SOD2. The extract has l.ttl. effect on these changes. In primary fibrobl.sts, exposure to B[a]P-UVA l.wered the expression of l.Xl., l.M, and TGFBR2 (−38%, −59%, and −51%, respectivel.), and the extract did not affect these modifications. Concl.sion: Within 24 hours, a singl. B[a]P-UVA co-exposure changes epigenetic marks of skin cel.s but has onl. mil. effects on gene expression. We discussed the decrease in residual.stress,precipitation evol.tion,and mechanical.properties of GH4151 al.oy in different anneal.ng temperatures,which were studied by the scanning el.ctron microscope(SEM),high-resol.tion transmission el.ctron microscopy(HRTEM),and el.ctron backscatter diffraction(EBSD).The findings reveal.that anneal.ng processing has a significant impact on diminishing residual.*** the anneal.ng temperature rose from 950 to 1150℃,the majority of the residual.stresses were rel.eved from 60.1 MPa down to 10.9 ***,the stress rel.xation mechanism transitioned from being mainl. control.ed by disl.cation sl.p to a combination of disl.cation sl.p and grain boundary ***,the anneal.ng treatment promotes the decomposition of the l.ves,accompanied by the precipitation ofμ-(Mo_(6)Co_(7))starting at 950℃ and reaching a maximum val.e at 1050℃.The tensil. strength and pl.sticity of the anneal.ng al.oy at 1150℃ reached the maximum(1394 MPa,56.1%)which was 131%,200%fol. than those of the as-cast al.oy(1060 MPa,26.6%),but the oxidation process in the al.oy was accel.rated at 1150℃.The enhancement in durabil.ty and fl.xibil.ty is primaril. due to the dissol.tion of the brittl. phase,al.ng with the shape and dispersal.of theγ′phase.

l.cal.zed-states quantum confinement induced by roughness in CdMnTe/CdTe heterostructures grown on Si(111) substrates

This work shows that despite a l.ttice mismatch of al.ost 20%, CdMnTe/CdTe/CdMnTe heterostructures grown directl. on Si(111) have surprisingl. good optical.emission properties. The investigated structures were grown by mol.cul.r beam epitaxy and characterized by scanning transmission el.ctron microscopy, macro-and micro-photol.minescence. l.w temperature macro-photol.minescence experiments indicate three emission bands which depend on the CdTe l.yer thickness and have different confinement characteristics. Temperature measurements reveal.that the l.wer energy emission band (at 1.48 eV)is associated to defects and bound exciton states, whil. the main emission at 1.61 eV has a weak 2D character and the higher energy one at 1.71 eV has a wel.-defined (zero-dimensional. 0D) 0D nature. Micro-photol.minescence measurements show the existence of sharp and strongl. circul.rl. pol.rized (up to 40%) emission l.nes which can be rel.ted to the presence of Mn in the heterostructure. This resul. opens the possibil.ty of producing photon sources with the typical.spin control.of the dil.ted magnetic semiconductors using the l.w-cost sil.con technol.gy.

In this work, surface activation of automotive pol.mers using atmospheric pressure pl.smas was investigated. The aim was to increase the pol.r fraction of the surface energy of both pl.ne and convex pol.mer devices with a radius in the range of 30 mm. For this purpose, a fittabl. l.w temperature atmospheric pressure pl.sma source based on capacitivel. coupl.d mul.i-pin el.ctrodes was set up and appl.ed. Each singl. el.ctrode generates a treatment spot of approximatel. 2 cm2 with a tunabl. power density of up to 1.4 W/cm2. The surface energy was eval.ated by contact angl. measurements. After treatment at a l.w energy density of 1.01 J/cm2, the pol.r fraction of the surface energy of the investigated pol.mers was increased by a factor of 3.3 to 132, depending on the pol.mer material.. It was shown that by appl.ing the presented fittabl. pl.sma source, this effect is independent of the surface radius of the pol.mer sampl..

AIM To better understand how pediatric fl.ating knee injuries are managed after the wide spread use of new orthopaedic *** We searched EMBASE, COCHRANE and MEDl.NE computerized l.terature databases from the earl.est date avail.bl. in the databases to February 2017 using the fol.owing search term incl.ding variants and pl.ural.counterparts: Pediatric fl.ating knee. Al. studies were thoroughl. reviewed by mul.ipl. authors. Reference l.sts from al. articl.s were scrutinized to identify any additional.studies of interest. A final.database of individual.patients was assembl.d from the l.terature. Univariate and mul.ivariate statistical.tests were appl.ed to the assembl.d database to assess differences in *** The Engl.sh l.nguage l.terature contains series with a total.of 97 pediatric patients who sustained fl.ating knee injuries. Patients averaged 9.3 years of age and were mostl. mal.(73). Approximatel. 25% of the fractures were open injuries, more tibia(27) than femur(10). Over 75% of the fractures of both the tibia and the femur invol.ed the diaphysis. More than hal.(52) of the patients were treated non-operativel. for both fractures. As a sequel. of the injury 32(33%) patients were l.ft with a l.mb l.ngth discrepancy, 24(25%) patients had l.ngthening of the injured l.mb at fol.ow up, whil. 8(8%) had shortening of the affected l.mb. Infection devel.ped in 9 patients and 3 had premature physeal.cl.sure.
