sirNA-mediated downregulation of TC21 sensitizes esophageal cancer cells to cisplatin

AIM： To determine the functional significance of TC21 in esophageal squamous cell carcinoma （EsCC）. METHODs： TC21 sirNA transfection was carried out using Hyperfectamine to knock down TC21, and tran- scr.p.s were analyzed by reverse transcription-poly- merase chain reaction and protein by Western *** demonstrated the effect of TC21 downregulation of cell signaling in esophageal cancer cells by assess- ing the phosphorylation status of its downstream tar- gets, phosphoinositide 3-kinase （pI3K）, phosphatase and tensin homolog （pTEN）, protein kinase B （pAl〈t）, nuclear factor-KB （NF-~B） and cyclinD1 using specific antibodies. Cell survival analysis after cisplatin treat- ment was carried out by cell viability assay and cell cycle analysis using flow cytometry. rEsULTs： TC21 knockdown in human EsCC cell line TEl3 cells, showed only a marginal increase （14.2%） in cell death compared with control cells. The expres- sions of the signaling proteins pI3K and pAkt, transcrip- tion factor NF-KB, and cell cycle protein cyclin D1 were markedly decreased in response to TC21 downregula- tion, whereas the level of ppTEN, an antagonist of pI3K, was increased. In addition, we evaluated the potential of TC21 as a putative target for sensitizing EsCC cells to the chemotherapeutic agent cisplatin. Increased cell death （38.4%） was observed in cells treated with cis- platin after TC21 knockdown compared with cells which were treated with cisplatin alone （20% cell death）. CONCLUsION： results suggest that TC21 mediates its effects via the pI3K-Akt pathway, NF-KB and cyclin D1, and enhances chemoresistance in esophageal cancer cells.

In this paper a fuzzy logic （FL） based model reference adaptive system （MrAs） speed observer for high performance AC drives is pr.p.sed. The error vector computation is made based on the rotor-flux derived from the reference and the adaptive model of the induction motor. The error signal is processed in the pr.p.sed fuzzy logic controller （FLC） for speed adaptation. The drive employs an indirect vector control scheme for achieving a good closed loop speed control. For powering the drive system, a standalone photovoltaic （pV） energy source is used. To extract the maximum power from the pV source, a constant voltage controller （CVC） is also pr.p.sed. The complete drive system is modeled in MATLAB/simulink and the performance is analyzed for different operating conditions.

Comparative Multivariate Analysis of Codon and Amino Acid Usage in Three Leishmania Genomes

Multivariate analysis of codon and amino acid usage was performed for three Leishmania species, including L. donovani, L. infantum and L. major. It was revealed that all three species are under mutational bias and translational selection. Lower GC12 and higher GC3s in all three parasites suggests that the ancestral highly expressed genes （HEGs）, compared to lowly expressed genes （LEGs）, might have been rich in AT-content. This also suggests that there must have been a faster rate of evolution under GC-bias in LEGs. It was observed from the esti- mation of synonymous/non-synonymous substitutions in HEGs that the HEG dataset of L. donovani is much closer to L. major evolutionarily. This is also supported by the higher ds value as compared to ds between L. donovani and L. major, suggesting the conservation of synonymous codon positions between these two species and the role of translational selection in shaping the composition of protein-coding genes.

Ayurvedic concept of shatkriyakala:a traditional knowledge of cancer pathogenesis and therapy

The term Kriyakala refers to the recognition of the stage of a disease＇s progress, which helps to determine appropriate measure to correct the imbalance in Doshas （biological factors）. It is a compound expression, comprised of Kriya and Kala, where Kriya means the choice to treatment （medicine, food and daily-routine） used to improve the disturbance in Doshas, and Kala refers to the stage of progress of a disease. sushruta, an ancient Indian surgeon, has described the concept of Kriyakala in Vamaprashnadhyaya, an ancient Vedic sanskrit text, which seeks to explain the incidence of Vamas in terms of Doshic disturbances. Vama, in modern parlance, may be described as an inflammatory process that may lead ulceration and chronic inflammation, promoting all stages of carcinogenesis. Abnormal interactions between prakriti （genotype） and environmental factors vitiate the Doshas and impair immunity, which can lead to aberrant cell growth and cancer. Moreover, the interaction between vitiated Doshas and weak Dhatus （body tissues） manifests as cancers of a specific organ. shatkriyakala （six stages of progress of a disease）, on the other hand, provides a framework to assess the cancer and its pathogenesis in different stages. According to Ayurvedic concepts, all cancer therapies treat the affected tissues indirectly by eliminating vitiated Doshas, rejuvenating Dhatus and restoring immunity in cancer patients. The present review describes the six stages of shatkriyakala in detail, with an emphasis on research areas to validate the concept of shatkriyakala. This traditional knowledge can be utilized with modern technologies to detect predisposition for cancer or diagnose cancer in its early stages.

rice Husk Biochar Influences seedling Emergence of Junglerice(Echinochloa colona)and Herbicide Efficacy

The use of carbonized rice husk biochar improves the fertility and productivity of poor soils in rice-based cropping systems. However, biochar may also influence weed seedling emergence and the efficacy of soil-applied herbicides. Experiments were conducted in a screenhouse to evaluate the effect of biochar rates (0, 20, 40, and 80 t·ha?1) and seed burial depth (0, 1, and 2 cm) on seedling emergence of junglerice (Echinochloa colona) and the effect of biochar rates and pendimethalin (0, 500, 1000, and 1500 g·a.i.·ha?1) and pretilachlor doses (0, 300, 600, and 900 g·a.i.·ha?1) on seedling emergence and seedling biomass of junglerice. Data were analyzed using nonlinear regression. The burial depth to inhibit 50% of maximum seedling emergence was 0.76 cm when biochar was not added to soil and the depth increased with an increase in biochar rates for soil. similarly, compared with the soil with no biochar, the use of bichoar increased the pretilachlor dose to inhibit 50% of maximum emergence or biomass. The pretilachlor dose to inhibit 50% of maximum biomass of junglerice was 100, 130, 240, and 240 g·ha?1 when biochar was added at 0, 20, 40, and 80 t·ha?1. However, the efficacy of pendimethalin was not influenced by biochar rate. The results of this study suggest that rice husk biochar may increase weed seedling emergence from deeper burial depths and may decrease the efficacy of some soil-applied herbicides.

Fabrication characteristics and tensile strength of novel Al2024/siC/red mud composites processed via stir casting route

The stir casting technique was used to fabricate aluminum2024matrix hybrid composites reinforced with siC(5%,mass fraction)and red mud(5%-20%,mass fraction)*** developed composites were characterized by using scanning electron microscopy(sEM)and electron dispersive spectrum(EDs)***,Taguchi’s approach of experimental design was used to examine the tensile strength of the hybrid composites(with minimum number of experiments).It was found that the reinforcing particles were well dispersed and adequately bonded in the hybrid *** density and porosity of the hybrid composites were reduced with the increase in reinforcement *** tensile strength of the composites increased with the increase in the red mud content and the ageing *** developed model indicated that the red mud content had the highest influence on the tensile strength response followed by the ageing ***,it was found that Al2024/siC/red mud composites exhibited superior tensile strength(about34%higher)in comparison to the Al2024alloy under optimized conditions.

resTs: residual Deep interpretable architecture for plant disease detection

recently many methods have been induced for plant disease detection by the influence ofDeep Neural Networks in Computer Vision. However, the dearth of transparency in thesetypes of research makes their acquisition in the real-world scenario less approving. We propose an architecture named resTs (residual Teacher/student) that can be used as visualization and a classification technique for diagnosis of the plant disease. resTs is a tertiaryadaptation of formerly suggested Teacher/student architecture. resTs is grounded on aConvolutional Neural Network (CNN) structure that comprises two classifiers (resTeacherand resstudent) and a decoder. This architecture trains both the classifiers in a reciprocalmode and the conveyed representation between resTeacher and resstudent is used as aproxy to envision the dominant areas in the image for categorization. The experimentshave shown that the pr.p.sed structure resTs (F1 score: 0.991) has surpassed the Teacher/student architecture (F1 score: 0.972) and can yield finer visualizations of symptomsof the disease. Novel resTs architecture incorporates the residual connections in all theconstituents and it executes batch normalization after each convolution operation whichis dissimilar to the formerly pr.p.sed Teacher/student architecture for plant disease diagnosis. residual connections in resTs help in preserving the gradients and circumvent theproblem of vanishing or exploding gradients. In addition, batch normalization after eachconvolution operation aids in swift convergence and increased reliability. All test resultsare attained on the plantVillage dataset comprising 54 306 images of 14 cr.p.species.
