EGFR-targeted and gemcitabine-incorporated chemogene for combinatorial pancreatic cancer treatment

Pancreatic cancer stands out as a recognized intractable tumor due to its high malignancy and mortality rates,which are largely attributed to the insensitivity of current clinical chemotherapies or *** chemo and gene therapy that integrates different therapeutic targets,may increase the chemosensitivity of pancreatic cancer and synergistically enhance the antitumor ***,conventional co-delivery of gene and chemo drugs is intensively dependent on complex nanoparticle delivery systems,thus would be limited by unstable drug packaging,nonspecific biodistribution,and biosafety ***,we rationally designed an epidermal growth factor-receptor(EGFR)-targeted and gemcitabine-incorporated oligonucleotide(termed as chemogene)with anti-Bcl-2 sequence,which achieves simple and precise integration of gemcitabine into a gene regulative agent,as well as the EGFR-targeted delivery for pancreatic cancer *** solid-phase synthesis,gemcitabine,as the first-line chemodrug for pancreatic cancer,is introduced to the antisense oligonucleotide to replace all cytosine nucleosides to obtain the gemcitabine-integrated chemogene(Ge-ASO^(Bcl-2)).Thereafter,Ge-ASO^(Bcl-2)is covalently coupled with EGFR nanobody to construct the final targeted chemogene without any exogenous ***,this nanobody-conjugated chemogene exhibits remarkable tumor targeting capability and antitumor effects both in vitro and in vivo,which initiates a first step toward the application of combinatorial chemo and gene therapy for future pancreatic cancer treatment.

Study on the adsorption performance and regeneration of lignin-derived graphitic carbon for H_(2)S

In pursuit of effective adsorption materials for malodorous gases such as H_(2)S and to broaden the utilization avenues of lignin waste,this study employed the direct pyrolysis method to synthesize three types of alkali lignin graphitized carbons,namely C-800,KC-700,and *** them,KEC-700 exhibits a high specific surface area of 1672.9 m^(2)/g,significantly superior H_(2)S adsorption performance compared to other materials,an adsorption breakthrough time of up to 220 min,and a sulfur capacity of 67.1 mg/*** analysis showed that the more oxygen-containing functional groups of lignin charcoal and the larger specific surface area facilitated the adsorption of H_(2)*** reaching adsorption saturation,the degree of graphitization of lignin carbon *** H_(2)S adsorption products primarily manifest as elemental sulfur and sulfate within the pores of lignin carbon measuring less than 2 *** thermal regeneration,the charcoal effectively eliminates the elemental sulfur adsorption ***,sulfate removal proved unsatisfactory,as the adsorption efficiency of KEC-700 following two thermal regenerations was approximately 41%of that observed for fresh samples.

Lysobacter harbors a plethora of cryptic biosynthetic gene clusters(BGCs),albeit only a limited number have been analyzed to *** this study,we described the activation of a cryptic polyketide synthase(PKS)/nonribosomal peptide synthetase(NRPS)gene cluster(lsh)in Lysobacter *** 3655 through promoter engineering and heterologous expression in Streptomyces *** a result of this methodology,we were able to isolate two novel linear lipopeptides,lysohexaenetides A(1)and B(2),from the recombinant strain ***,we proposed the biosynthetic pathway for lysohexaenetides and identified LshA as another example of entirely iterative bacterial *** study highlights the potential of heterologous expression systems in uncovering cryptic biosynthetic pathways in Lysobacter genomes,particularly in the absence of genetic manipulation tools.

When an innovative product is introduced into the market, the innovator realizes that she will always face the competition of entrant imitator（s） after a period of monopoly time. Strategic customers may also anticipate that and can rationally make their purchase plan, which further complicates the situation of competition. In this paper, we develop a dynamic game model to formulate the problem and obtain the pricing equilibrium. In numerical studies, the relative position of monopoly price, the markdown size of the innovator and the relative price difference between the two firms are investigated to analyze the impacts of various factors on the pricing policies.

NO_x Formation from Several Chinese Coals during Combustion in Packed Bed

The formation characteristics of NO_x during combustion of four typical Chinese coals have been studied in a packed bed combustor. The studies included the effects of chemical composition of coals and volatile matters, and combustion conditions, i.e. preheating temperatures of combustion gas, on conversion of NO_x during combustion. In term of the combustion experiments, there is a close relation between coal composition and conversions of NO_x. Coals with a higher nitrogen content give a lower F_(NO_x)~0, and coals with a higher volatile content give a lower F_(NO_x)~0. The further devolatilization experiments showed that volatile matters in the coal have an important effect on reducing conversion of coal-nitrogen to NO_x. It was interpreted as that the release of volatile matters during combustion caused a fuel-rich circumstance in the vicinity of coal particles which may restrain the formation of NO_x. Preheating temperatures of combustion gas has significant effects on conversions of NO_x and there exist a critical preheating temperature. At such temperature, lower F_(NO_x)~0 can be reached.

Identification of candidate genes for aphid resistance in upland cotton by QTL mapping and expression analysis

Lignin is one of the main components of cell walls and is essential for resistance to insect pests in *** plants are damaged by aphid(Aphis gossypii) worldwide but resistant breeding is undeveloped due to scarce knowledge on resistance genes and the mechanism. This study reported a lignin biosynthesisrelated gene identified in the F_(2) population derived from the cross between cotton cultivars Xinluzao 61(resistant to aphid) and Xinluzao 50(susceptible to aphid). A quantitative trait locus was mapped on chromosome D04 with a logarithm of odds(LOD) score of 5.99 and phenotypic effect of 27%. RNA-seq analysis of candidate intervals showed that the expression level of GH_D04G1418 was higher in the resistant cultivar than in the susceptible cultivar. This locus is close to AtLAC4 in the phylogenetic tree and contains a conserved laccase domain. Hence, it was designated GhLAC4-3. Silencing of GhLAC4-3 in Xinluzao 61 via virus-induced gene silencing(VIGS) resulted in decreased lignin content and increased susceptibility to aphids. These results suggest that GhLAC4-3 might enhance aphid resistance by regulating lignin biosynthesis in cotton.

Because robotic milling has become an important means for machining significant large parts,obtaining the structural frequency response function(FRF)of a milling robot is an important basis for machining process ***,because of its articulated serial structure,a milling robot has an enormous number of operating postures,and its dynamics are affected by the motion *** accurately obtain the FRF in the operating state of a milling robot,this paper proposes a method based on the structural modification *** the traditional excitation method,the proposed method uses robot joint motion excitation instead of hammering excitation to realize *** address the problem of the lack of information brought by motion excitation,which leads to inaccurate FRF amplitudes,this paper derives the milling robot regularization theory based on the sensitivity of structural modification,establishes the modal regularization factor,and calibrates the FRF *** to the commonly used manual hammering experiments,the proposed method has high accuracy and reliability when the milling robot is in different *** the measurement can be performed directly and automatically in the operation state,and the problem of inaccurate amplitudes is solved,the proposed method provides a basis for optimizing the machining posture of a milling robot and improving machining efficiency.

Genomic surveillance of monkeypox virus(MPXV)is essential to explore the reason of its unusual *** phylogenomic analysis of the MPXV genome mainly focuses on the effect of amino acid ***,we explore the evolutionary variation of RNA G-quadruplex(RG4)of MPXV and find that the genome evolution of MPXV can also produce new effects through changes in the RG4 *** RG4 is located in MPXV’s only Kelch-like C9L gene,which encodes for an antagonist of the innate immune *** evolution of this virus increases the unfolding kinetic constant of C9L RG4 and promotes the C9 protein level in living ***,all reported MPXV genomes in 2022 carry the C9L-RG4-5 pattern with the highest unfolding kinetic ***,the RG4 ligand,RGB-1,can impede the unfolding of C9L-RG4-5 and thereby reduce the C9 protein *** findings carve out a new path to comprehensively understanding MPXV virology.
