Dietary isoleucine affects muscle fatty acid and amino acid profiles through regulating lipid metabolism and autophagy in hybrid catfish(Pelteobagrus vachelli ♀×Leiocassis longirostris ♂)

The present study explored the impacts of Ile on muscle fatty acid and amino acid profiles,lipid metabolism,and autophagy in hybrid *** isonitrogenous(387.8 g/kg protein)semi-purified diets were formulated to contain 5.0(control),7.5,10.0,12.5,15.0,17.5,and 20.0 g Ile/kg diet *** fish(initial weight of 33.11±0.09 g)were randomly assigned to 7 groups for a 56-day *** group has 3 replicates with 30 fish per replicate,fed at 08:00 and 18:00 each *** showed that muscle protein and lipid,C14:0,C18:0,C22:0,C14:1,C18:1n-9,polyunsaturated fatty acid(PUFA),Arg,Ile,Ala,Cys,Gly,Tyr,essential amino acid(EAA),and total amino acid(TAA)contents and flavor amino acid(FAA)/TAA in muscle had positive linear and/or quadratic responses to dietary Ile levels(P<0.05).Fatty acid synthase(FAS),stearoyl-Co A desaturase(SCD),acetyl-Co A carboxylase(ACC),and lipoprotein lipase(LPL)activities had positive linear and/or quadratic responses,but carnitine palmitoyl transferase 1(CPT1)activity had a negative response with increasing dietary Ile levels(P<0.05).The m RNA expressions of FAS,SCD,ACC,LPL,fatty acid binding protein 4(FABP4),FATP1,sterol response element-binding protein 1c(SREBP-1c),sequestosome 1(SQSTM1),and adenosine 5’-monophosphateactivated protein kinase(AMPK)had positive linear and/or quadratic responses to dietary Ile levels(P<0.05).The m RNA expressions of hormone-sensitive lipase(HSL),CPT1,peroxisome proliferatoractivated receptor a(PPARa),PPARγ,uncoordinated 51-like kinase 1(ULK1),beclin1(Becn1),autophagy-related protein 9a(Atg9a),Atg4b,Atg7,autophagy marker light chain 3 B(LC3B),and SQSTM1in muscle had negative linear and/or quadratic responses to dietary Ile levels(P<0.05).The p-AMPK and ULK1 protein levels,and p-AMPK/AMPK were decreased by 12.5 g Ile/kg in the diet(P<0.05).Finally,SQSTM1 protein level had the opposite effect(P<0.05).The above results indicate that dietary Ile improves fish muscle fatty acid and amino acid profiles potent

Effects of dietary forage to concentrate ratio and wildrye length on nutrient intake, digestibility, plasma metabolites, ruminal fermentation and fecal microflora of male Chinese Holstein calves

Twenty-eight male, weaned Chinese Holstein calves（（156.8±33.4） kg） were used to investigate the effects of dietary forage to concentrate ratio（F：C） and forage length on nutrient digestibility, plasma metabolites, ruminal fermentation, and fecal microflora. Animals were randomly allocated to four treatments in a 2×2 factorial arrangement： whole-length forage（WL） with low F：C（50：50）; WL with high F：C（65：35）; short-length forage（SL） with high F：C（65：35）; and SL with low F：C（50：50）. Chinese wildrye was used as the only forage source in this trial. The grass in the SL treatments was chopped using a chaff cutter to achieve small particle size（-50% particles 〉19 mm）. Dry matter intake（DMI） and organic matter（OM） intake was increased by increasing both F：C（P〈0.01） and forage length（FL）（P〈0.05）, while acid detergent fiber（ADF） and neutral detergent fiber（NDF） intakes were only increased by increasing the F：C（P〈0.01）. The digestibility of NDF was increased as the FL increased（P〈0.01）, and it was also affected by interaction between F：C and FL（P〈0.05）. Cholesterol（CHO）（P〈0.01）, leptin（LP）（P〈0.05）, and growth hormone（GH）（P〈0.01） concentrations in plasma were increased as dietary F：C increased. A significant increase in plasma triglyceride（TG）（P〈0.01）, insulin（INS）（P〈0.05）, and GH（P〈0.01） levels was observed with decreasing dietary FL. Ruminal p H values of calves fed with low F：C diets were significantly lower than those in high F：C treatment（P〈0.05）. Increasing the F：C enhanced ruminal acetic acid（P〈0.05） and acetic acid/propionic acid（P〈0.01）. Fecal Lactobacillus content was significantly higher, while Escherichia coli and Salmonella contents were significantly lower in WL and high F：C groups（P〈0.05）. Asymmetrically modified Janus graphene oxide(JGO)has attracted great attention due to its unique physical chemistry properties and wide *** modification degree of Janus nanosheets inevitably affects their interfacial activity,which is essential for their performances in enhanced oil recovery(EOR).In this study,the interfacial properties of Janus graphene oxide(JGO)with various modification degrees at liquid-liquid and liquid-solid interfaces were systematically evaluated via the measurements of interfacial tension(IFT),dilatational modulus,contact angle,and EOR efficiency was further assessed by core flooding *** is found that JGO-5 with higher modification degree exhibits the greater ability to reduce IFT(15.16 mN/m)and dilatational modulus(26 mN/m).Furthermore,JGO can construct interfacial and climbing film with the assistance of hydrodynamic power to effectively detach the oil from the rock surface and greatly enhance oil *** modified JGO-2 can highly improve recovery of residual crude oil(11.53%),which is regarded as the promising EOR agent in practical *** present study firstly focuses on the effects of modification degrees on the JGO interfacial properties and proposes diverse EOR mechanisms for JGO with different modification degrees.

CETP polymorphism confer genetic contribution to centenarians of Hainan,south of China

Objective:In this paper,we will discuss if the CETP polymorphism contributes to the centenarians in Hainan ***:We tested the Taq IB and I405 V polymorphisms of CETP gene among 276 centenarians and 301 matched healthy individuals by AS-PCR and analyzed the data with SPSS software package(Version 19.0).Results:Our data indicated that allele B1 and V have the significant differences between centenarians and healthy control groups with P<*** analysis implied that genotypes B1B1(P<0.001,OR=0.148,95% CI=0.095-0.230) and VV(P<0.001 and OR=0.353,95% CI=0.237-0.525) were significantly different between centenarians and matched *** combination of B and V,such as B1B1-II(P<0.001,OR=0.128,95% CI=0.049-0.329),B1B1-IV(P<0.001,OR=0.115,95% CI=0.056-0.237),B1B2-VV(P<0.05,OR=0.534,95% CI=0.310-0.920),and B2B2-VV(P<0.001,OR=0.198,95% CI=0.086-0.453) have significant differences between centenarians and matched healthy individuals from ***:Our results implied that allele B1B1 and VV,as well as the combination B1B1-II,B1B1-IV,B1B2-VV and B2B2-VV may contribute to the longevity in centenarians of Hainan,south of China.

Identification and functional prediction of long intergenic noncoding RNAs in fetal porcine longissimus dorsi muscle

Pigs are globally farmed animals which provide protein for human consumption in the form of skeletal muscle. To better understand the function of long intergenic noncoding RNAs(linc RNAs) in porcine skeletal muscle growth and development, we collected RNA-seq data from porcine longissimus dorsi muscle(LDM) during embryonic development. We identified a total of 739 linc RNA transcripts, which were distributed on all chromosomes except the chromosome Y, and analyzed their molecular characteristics. Compared to protein-coding genes, linc RNAs showed shorter transcripts, longer exons, fewer exons and higher tissue specificity. In addition, the abundance of linc RNAs in five embryonic development stages were analyzed and 45 differentially expressed linc RNAs were screened, three of which were highly expressed in LDM during porcine embryonic development. Finally, we predicted the potential target genes and functions of the linc RNAs, and identified 1 537 cis-target genes and 8 571 trans-target genes. Furthermore, we identified two key candidate linc RNAs involved in muscle development, XLOC_024652 and XLOC_001832, for post-trial validation. Our results provide a genome-wide resource of linc RNAs which are potentially involved in porcine embryonic skeletal muscle development and lay a foundation for the further study of their functions.

Modified Method for Estimating Organic Carbon Density in Discontinuous Karst Soil Using Ground-Penetrating Radar and Geostatistics

The conventional method which assumes the soil distribution is continuous was unsuitable for estimating soil organic carbon density(SOCD) in Karst areas because of its discontinuous soil distribution. The accurate estimation of SOCD in Karst areas is essential for carbon sequestration assessment in China. In this study, a modified method,which considers the vertical proportion of soil area in the profile when calculating the SOCD, was developed to estimate the SOCD in a typical Karst peak-cluster depression area in southwest China. In the modified method, ground-penetrating radar(GPR) technology was used to detect the distribution and thickness of soil. The accuracy of the method was confirmed through comparison with the data obtained using a validation method, in which the soil thickness was measured by excavation. In comparison with the conventional method and average-soil-depth method,the SOCD estimated using the GPR method showed the minimum relative error with respect to that obtained using the validation method. At a regional scale, the average SOCDs at depths of 0-20 cm and 0-100 cm, which were interpolated by ordinary kriging,were 1.49(ranging from 0.03-5.65) and 2.26(0.09-11.60) kgm-2based on GPR method in our study area(covering 393.6 hm2), respectively. Therefore, the modified method can be applied on the accurate estimation of SOCD in discontinuous soil areas such as Karst regions.

Bi-directional Selection in Upland Rice Leads to Its Adaptive Differentiation from Lowland Rice in Drought Resistance and Productivity

Drought resistance is required in rice breeding to address the challenge of frequent droughts . However, the evolutionary mechanism of rice drought resistance is not fully understood. We investigated the genetic differentiation between upland and lowland rice domesticated in agro-ecosystems with contrasting water-soil conditions using genome-wide SNPs. We estimated morphological differences among upland and lowland rice in drought resistance and productivity through common garden experiments. Upland rice had better drought resistance but poorer productivity. The negative correlations between traits of drought resistance and productivity are attributed to the underlying genetic trade-offs through tight linkages (e.g., DCA1 and OsCesA7) or pleiotropic effects (e.g., LAX1). The genetic trade-offs are comm on and greatly shape the evolutio n of drought resista nee in upland rice . In genomic regions associated with both productivity and drought resistance, signs of balancing selection were detected in upland rice, while signs of directional selection were detected in lowland rice, potentially contributing to their adaptive differentiation. Signs of balancing selection in upland rice resulted from bi-directional selection during its domestication in drought-prone upland *** genome-wide association analysis, we ide ntified several valuable quantitative trait loci associated with drought resista nee, for which highly differentiated genes should be considered candidates. Bi-directional selection breaking tight linkages by accumulating recombination events would be applicable in breeding water-saving and droughtresistance rice.
