To date,sperm morphometric studies have assessed whole sperm populations without considering sperm *** aim of this study was to evaluate the relati on ship of sperm membrane and acrosomal integrity with sperm morphometry in liquid semen samples collected from *** this end,sperm morphometry was performed on cryopreserved semen samples from 16 bulls by a combination of fluorescent dyes,including Hoechst 33343,carboxyfluorescein diacetate,and propidium *** allowed discrimination of different subpopulations on the basis of sperm membrane and acrosomal integrity and analysis of the morphometries of the sperm head,nucleus,and acrosome using a specific plug-in module created on *** integrity was related to sperm morphometry as the heads of spermatozoa with a damaged acrosome were significantly smaller than those with a normal acrosome(P<0.001).In the case of spermatozoa with an intact acrosome,those with a damaged plasma membrane had a larger sperm head and acrosome than spermatozoa with an intact plasma membrane(P<0.001).No significant differences in the sperm head size were observed between sperm subpopulations without an acrosome or in the nuclear sperm morphometry of the different *** was a positive correlation between the sperm motility values of the samples and the morphometric parameters for in tact *** correlations were particularly strong for the morphometric parameters of the sperm *** conclude that there are clear differences in the sperm morphometry depending on the status of the sperm membrane and acrosome and this should be considered when performing this kind of analysis.

Adult stem cells,especially from bone marrow,have a tremendous therapeutic potential,and its application in regenerative medicine has been increasingly studied [1,2].It is well known that three-dimensional (3D)cell culture matrix promotes many biological relevant functions not observed in two-dimensional (2D) monolayer cell culture [3]and therefore,engineering 3D cell culture platforms to mimic the native in vivo biological systems represents a realistic approach for studies of bone marrow stem cells. High-throughput microfluidics constitutes a promising technology to generate hydrogel three dimensional constructs [4,5],however, its application to host adult stem cells remains still a challenge.

Previously regarded as simple fat storage particles,new evidence suggests thatlipid droplets(LDs)are dynamic and functional organelles involved in keycellular processes such as membrane biosynthesis,lipid metabolism,cellsignalling and ***,an increased LD content is one of the mostapparent features resulting from lipid metabolism reprogramming necessary tosupport the basic functions of cancer *** have been associated to differentcellular processes involved in cancer progression and aggressiveness,such astumorigenicity,invasion and metastasis,as well as ***,all of these processes are controlled by a subpopulation of highly aggressivetumoral cells named cancer stem cells(CSCs),suggesting that LDs may befundamental elements for stemness in *** the key role of CSCs onchemoresistance and disease relapse,main factors of therapy failure,the design ofnovel therapeutic approaches targeting these cells may be the only chance forlong-term survival in cancer *** this sense,their biology and functionalproperties render LDs excellent candidates for target discovery and design ofcombined therapeutic *** this review,we summarise the currentknowledge identifying LDs and CSCs as main contributors to cancer aggressiveness,metastasis and chemoresistance.

The addition of Au3. to spherical amine-capped CdSe@ZnS nanoparticles in toluene at room temperature and under darkness can lead to ternary CdSe@ZnS/Au nanohybrids. We demonstrate that this happens only when the nanoparticles possess a relatively thin ZnS shell, thus showing that thickness plays a key role in gold deposition on the CdSe@ZnS nanoparticle surface. Our hypothesis is that the amine ligand acts as the reductant of Au3＋ ions into Au＋ ions, whose affinity for sulfur would keep them at the CdSe@ZnS surface. This interaction stabilizes the Au＋ ion, making it less prone to reduction than a non-coordinated Au＋ ion. In CdSe@ZnS with a thin shell, Au＋ ions at the surface of, or most probably within, the ZnS shell cause the transfer of Cd2＋ ions into the solution. Subsequently, the core Se2- anion, which is a better reductant than the shell S2- reduces Au＋ ions to Au（0）, and large gold nanoparticles （AuNPs） are quickly deposited on the CdSe@ZnS surface in room temperature process, leading to ternary CdSe@ZnS/Au nanohybrids. In solution, these ternary nanohybrids progressively transform into quaternary CdSe@ZnS/Au2S/Au nanohybrids due to the reaction of the shell S2- anion with the remaining Au＋ at the CdSe@ZnS surface, thus leading to the growth of Au2S nanoparticles on the CdSe@ZnS surface while Zn concomitantly leaches from the nanohybrid into the solution. Photoirradiation of the heterostructures with visible light enhances their emission efficiency. Comparatively, irradiation of the precursors, i.e., CdSe@ZnS nanoparticles, causes a drastic decrease in their emission accompanied by a blue shift of their emission maximum. The optical properties of these nanohybrids were analyzed by absorption and fluorescence （steady-state and time-resolved） spectroscopy, and the composition of the samples and the chemical states were determined by energy dispersive X-ray spectroscopy （EDX） and X-ray photoelectron spectroscopy （XPS）, respectively. Non-alcoholic fatty liver disease(NAFLD)is currently considered the most common cause of liver *** prevalence is increasing in parallel with the obesity and type 2 diabetes mellitus(DM2)epidemics in developed *** recent studies have suggested that NAFLD may be the hepatic manifestation of a systemic inflammatory metabolic disease that also affects other organs,such as intestine,lungs,skin and vascular *** appears that local and systemic proinflammatory/anti-inflammatory cytokine imbalance,together with insulin resistance and changes in the intestinal microbiota,are pathogenic mechanisms shared by NAFLD and other *** is more common in patients with extrahepatic diseases such as inflammatory bowel disease(IBD),obstructive syndrome apnea(OSA)and psoriasis than in the general ***,there is evidence that this association has a negative impact on the severity of liver *** risk characteristics for NAFLD have been identified in populations with IBD(***,obesity,DM2,previous bowel surgery,IBD evolution time,methotrexate treatment),OSA(***,DM2,OSA severity,increased transaminases)and psoriasis(***,metabolic factors,severe psoriasis,arthropathy,elevated transaminases,methotrexate treatment).These specific phenotypes might be used by gastroenterologists,pneumologists and dermatologists to create screening algorithms for *** algorithms should include non-invasive markers of fibrosis used in NAFLD to select subjects for referral to the ***,controlled studies in NAFLD patients with extrahepatic comorbidities are required to demonstrate a causal relationship and also that appropriate multidisciplinary management improves these patients’prognosis and survival.

Samples of a cobalt-based alloy that underwent a surface treatment were evaluated. The samples, which were obtained by casting alloy ASTM F 75, were ground and polished on one side until a mirror finish was obtained. The samples were encapsulated in wollastonite （W） using uniaxial pressure, treated at 1 220 ~C for 1 h and subsequently tempered in water. The characterisation of the sample indicated that part of the ceramic encapsulating material was mechanically incorporated on the metallic surface by growth of the oxide layer of the alloy. After thermal treatment, a series of specimens were submerged in a solution with 5-fold simulated body fluid （5SBF） for 3, 5 and 21 days. Characterisation by scanning electron microscopy （SEM）, energy dispersive spectroscopy （EDS） and X-ray diffraction （XRD） indicated nucleation and growth of a homogenous layer of apatite, beginning on the third day when the sample was submerged.

Superoxide dismutase prevents development of adenocarcinoma in a rat model of Barrett's esophagus

AIM： To test whether antioxidant treatment could prevent the progression of Barrett＇s esophagus to adenocarcinoma. METHODS： In a rat model of gastroduodenoesophageal reflux by esophagojejunal anastomosis with gastric preservation, groups of 6-10 rats were randomized to receive treatment with superoxide dismutase （SOD） or vehicle and followed up for 4 too. Rat＇s esophagus was assessed by histological analysis, superoxide anion and peroxinitrite generation, SOD levels and DNA oxidative damage. RESULTS： All rats undergoing esophagojejunostomy developed extensive esophageal mucosal ulceration and inflammation by mo 4. The process was associated with a progressive presence of intestinal metaplasia beyond the anastomotic area （9% 1st mo and 50% 4th too）（94% at the anastomotic level） and adenocarcinoma （11% 1^ST mo and 60% 4th too）. These changes were associated with superoxide anion and peroxinitrite mucosal generation, an early and significant increase of DNA oxidative damage and a significant decrease in SOD levels （P〈0.05）. Exogenous administration of SOD decreased mucosal superoxide levels, increased mucosal SOD levels and reduced the risk of developing intestinal metaplasia beyond the anastomotic area （odds ratio = 0.326; 95%CI： 0.108-0.981; P = 0.046）, and esophageal adenocarcinoma （odds ratio = 0.243; 95%CI： 0.073-0.804; P = 0.021）. CONCLUSION： Superoxide dismutase prevents the progression of esophagitis to Barrett＇s esophagus and adenocarcinoma in this rat model of gastrointestinal reflux, supporting a role of antioxidants in the chemoprevention of esophageal adenocarcinoma.
