Combining Phytate/Ca^(2+) Fractionation with Trichloroacetic Acid/Acetone Precipitation Improved Separation of Low-Abundant Proteins of Wheat (Triticum aestivum L.) Leaf for Proteomic Analysis

Proteomic assessment of low-abundance leaf proteins is hindered by the large quantity of ribulose-1,5-bisphosphate carboxylase/oxygenase （Rubisco） present within plant leaf tissues. In the present study, total proteins were extracted from wheat （Triticum aestivum L.） leaves by a conventional trichloroacetic acid （TCA）/acetone method and a protocol first developed in this work. Phytate/Ca2＋ fractionation and TCA/acetone precipitation were combined to design an improved TCA/acetone method. The extracted proteins were analysed by two-dimensional gel electrophoresis （2-DE）. The resulting 2-DE images were compared to reveal major differences. The results showed that large quantities of Rubisco were deleted from wheat leaf proteins prepared by the improved method. As many as （758±4） protein spots were detected from 2-DE images of protein extracts obtained by the improved method, 130 more than those detected by the TCA/acetone method. Further analysis indicated that more protein spots could be detected at regions of pI 4.00-4.99 and 6.50-7.00 in the improved method-based 2-DE images. Our findings indicated that the improved method is an efficient protein preparation protocol for separating low-abundance proteins in wheat leaf tissues by 2-DE analysis. The proposed protocol is simple, fast, inexpensive and also applicable to protein preparations of other plants.

Ubiquitin E2 enzyme variant （UEV） proteins are similar to ubiquitin-conjugating enzyme （E2） in both sequence and structure, but the lack of a catalytic cysteine residue renders them incapable of forming a thiolester linkage with ubiquitin. While the functional roles of several UEVs have been defined in yeast and animal systems, Arabidopsis COP10, a photomorphogenesis repressor, is the only UEV characterized in plants. Phylogenetic analysis revealed that the eight Arabidopsis UEV genes belong to three subfamilies. The expresslon of those genes is supported by either the presence of ESTs or RT-PCR analysis. We also characterized the other members of the COP10 subfamily, UEV2. Semi-quantitative RT-PCR analysis indicated that the UEV2 transcripts can be detected in most organs of Arabidopsis. Analysis of UEV2：：GUS transgenic lines also showed its ubiquitous expression in nearly all the developmental stages of Arabidopsis. Transient expression analysis indicated that the sGFP-UEV2 fusion protein can localize to both the cytoplasm and nucleus. A T-DNA insertion mutant, uev2-1, which abolished the transcription of UEV2, displays no visible phenotype. Further, the cop10-4 uev2-1 double mutant exhibits the same phenotype as the cop10-4 mutant in darkness. UEV2 is therefore not functionally redundant with COP10.

Influence of social capital on the livelihood strategies of farmers under China's rural revitalization strategy in poor mountain areas: A case study of the Liangshan Yi autonomous prefecture

Social capital in the form of social resources or social networks is one of the most important livelihood capital of farmers, which can increase the labor productivity of poor households and increase income. It is important to explore the reasons underlying the livelihood strategy choices of farmers from the perspective of social capital under China’s rural revitalization strategy. In this study, the Liangshan Yi Autonomous Prefecture, a povertystricken mountainous area in southwestern China, was selected as the case study area, and multivariable linear regression models were constructed to analyze the influence of social capital on livelihood *** results are as follows:(1) Individual social capital had a positive effect on non-agricultural livelihood strategies. On average, with a one-unit increase in individual social capital, the ratio of farmers’ nonagricultural income to total productive income(Income_Rto) increased by 0.002% and 0.062%,respectively. Collective social capital, with the Peasant Economic Cooperation Organization(PECO) as the carrier, had a negative effect on the non-agricultural livelihood strategies of farmers;on average, with a oneunit increase in PECO, Income_Rto decreased by approximately 0.053%. However, this effect was only significant in the river valley area.(2) The income differences among the different livelihood strategy types were explained by the livelihood strategy choices of farmers. As non-agricultural work can bring more benefits, the labor force exhibited one-way migration from villages to cities, resulting in a lack of the subject of rural revitalization. It is necessary to implement effective measures to highlight the role of PECO in increasing agricultural income for farmers. Finally,based on the above conclusions,policy recommendations with respect to livelihood transformation of farmers and rural sustainable development are discussed.

Environmentally-friendly magnetic metallic absorbers with high-performing antioxidant property,thermal stability,and anticorrosion capability have attracted great attention in real-world applications.A surface modification technology of magnetic metallic absorbers with dense and inert materials has been an effective strategy to solve the aforesaid ***,fluorinefree core–shell carbonyl iron-organic silicon absorbers(CI@SiO_(2)/1,1,1,3,3,3-hexamethyl disilazane(HMDS))were fabricated via a facile one-pot synthesis using tetraethyl orthosilicate(TEOS)and HMDS as the precursor of protective layer(SiO_(2)/HMDS),and CI@SiO_(2)/HMDS hybrid reveals its long-term corrosion resistance and excellent microwave absorption performance with a minimum reflection loss value of−44.3 dB and an effective absorption bandwidth of 5.3 GHz at a thin thickness of 2.0 mm after immersion in 5.0 wt.%NaCl acidic solutions for 2,160 ***,CI@SiO_(2)/HMDS hybrid can still achieve the maximum radar cross-sectional(RCS)reduction values about 16.5 dB·m^(2) at the detectionθof 0°.The exceptional microwave absorption performance and structural stability are largely due to the extraordinary wave-transparent property and shielding ability against corrosive medium of SiO_(2)/HMDS hydrophobic protective layer with a contact angle of 132.5°.The research paves the way for the large-scale and batch production of high-performance magnetic metallic absorbers and increases their survivability and reliability in the harsh environments.

Hydrogen fuel cells are among the promising energy sources worldwide,which could accomplish cyclic production of energy and avoid the emission of green-house or contaminative ***,sulfur compounds(SCs)even at trace level(nmol/mol)are usually involved in cell construction and further H_(2)production,which would cause degradation of the catalysts and shorten the lifetime of the fuel ***,the highly reactive SCs could cause varied species and concentrations of them in complex matrices,so online rather than offline analysis of SCs in H_(2)would be *** this context,we developed a new system combining online cryogenic preconcentration of nine SCs and subsequent determination by GC-SCD(sulfur chemiluminescent detector),with the correlation coefficients of the calibration curves higher than 0.999,calculated limits of detection no higher than 0.050 nmol/mol,analytical time around 30 min per sample,and satisfactory precision and accuracy(RSD<5%and SD<15%).The analytical performance was much better than or at least comparable to the previously reported and the developed system was successfully applied for real sample analysis.

Prediction for the Multi-band Afterglows of FRB 200428 and its Implication

The physical mechanism of fast radio bursts(FRBs) is still unknown. On 2020 April 28, a special radio burst, FRB200428, was detected and believed to be associated with the Galactic magnetar SGR 1935+2154. It confirms that at least some of the FRBs were generated by magnetars, although the radiation mechanism continues to be *** this end, we study in detail the multiband afterglows of FRB 200428 described by the synchrotron fireball shock model. We find the prediction for the optical and radio afterglows of FRB 200428 is consistent with the observations when considering the post-FRB optical and radio upper limits from the literature. We also show that the follow up detection of the afterglows from fast radio bursts like—FRB 200428 is possible at the radio band,though challenging. Based on our model, one can obtain information about the energy of the fireball, the radiation zone, and the nature of the surrounding medium. That may shed light on the physical mechanism of FRBs.

Beta gallium oxide(β-Ga_(2)O_(3)) has attracted significant attention for applications in power electronics due to its ultrawide bandgap of ~ 4.8 eV and the large critical electric field of 8 MV/cm. These properties yield a high Baliga's figures of merit(BFOM) of more than 3000. Though β-Ga_(2)O_(3) possesses superior material properties, the lack of p-type doping is the main obstacle that hinders the development of β-Ga_(2)O_(3)-based power devices for commercial use. Constructing heterojunctions by employing other p-type materials has been proven to be a feasible solution to this issue. Nickel oxide(NiO) is the most promising candidate due to its wide band gap of 3.6–4.0 eV. So far, remarkable progress has been made in NiO/β-Ga_(2)O_(3) heterojunction power devices. This review aims to summarize recent advances in the construction, characterization, and device performance of the NiO/β-Ga_(2)O_(3) heterojunction power devices. The crystallinity, band structure, and carrier transport property of the sputtered NiO/β-Ga_(2)O_(3) heterojunctions are discussed. Various device architectures, including the NiO/β-Ga_(2)O_(3) heterojunction pn diodes(HJDs), junction barrier Schottky(JBS) diodes, and junction field effect transistors(JFET), as well as the edge terminations and super-junctions based on the NiO/β-Ga_(2)O_(3) heterojunction, are described.
