Endoscopic ultrasound elastography strain histograms in the evaluation of patients with pancreatic masses

AIM: To investigate the accuracy of the strain histogram endoscopic ultrasound(EUS)-based method for the diagnostic differentiation of patients with pancreatic masses. METHODS: In a prospective single center study, 149 patients were analyzed, 105 with pancreatic masses and 44 controls. Elastography images were recorded using commercially available ultrasound equipment in combination with EUS linear probes. Strain histograms(SHs) were calculated by machine integrated software in regions of interest and mean values of the strain histograms were expressed as Mode 1(over the mass) and Mode 2(over an adjacent part of pancreatic tissue, representing the reference area). The ratio between Mode 2 and Mode 1 was calculated later, representing a new variable, the strain histogram ratio. After the final diagnosis was established, two groups of patients were formed: a pancreatic cancer group with positive cytology achieved by fine needle aspiration puncture or histology after surgery(58 patients), and a massforming pancreatitis group with negative cytology and follow-up after 3 and 6 mo(47 patients). All statistical analyses were conducted in SPSS 14.0(SPSS Inc., Chicago, IL, United States).RESULTS: Results were obtained with software for strain histograms with reversed hue scale(0 represents the hardest tissue structure and 255 the softest). Based on the receiver operating characteristics(ROC) curve coordinates, the cut-off point for Mode 1 was set at the value of 86. values under the cut-off point indicated the presence of pancreatic malignancy. Mode 1 reached 100% sensitivity and 45% specificity with overall accuracy of 66%(95%CI: 61%-66%) in detection of pancreatic malignant tumors among the patients with pancreatic masses. The positive and negative predictive values were 54% and 100%, respectively. Traumatic brain injury remains a global health crisis that spans all demographics,yet there exist limited treatment options that may effectively curtail its lingering *** brain injury pathology entails a progression from primary injury to inflammation-mediated secondary cell *** this inflammation as a means of ameliorating the greater symptomology of traumatic brain injury has emerged as an attractive treatment *** this review,we recapitulate and evaluate the important developments relating to regulating traumatic brain injury-induced neuroinflammation,edema,and blood-brain barrier disintegration through pharmacotherapy and stem cell *** these studies of stand-alone treatments have yielded some positive results,more therapeutic outcomes have been documented from the promising area of combined drug and stem cell *** the facilitatory properties of certain pharmaceuticals with the anti-inflammatory and regenerative effects of stem cell transplants creates a synergistic effect greater than the sum of its *** burgeoning evidence in favor of combined drug and stem cell therapies warrants more elaborate preclinical studies on this topic in order to pave.the way for later clinical trials.

Efficient Solvothermal Synthesis of Defect-Rich Cu-BTC•MOF with Enhanced Electrocatalytic Activity in Alkaline Hydrogen Evolution Reaction

Porous metal–organic frameworks(MOFs) have been recently discovered to be efficient catalysts for energy applications and green technologies. Here, we report on a scalable catalytic platform using Cu–based MOFs for electrocatalytic alkaline hydrogen evolution reaction. First, the solvothermal synthesis of Cu–BTC MOFs(BTC = 1,3,5–benzenetricarboxylate) at 85 ℃ and a 1:60 ligand–to–solvent ratio allowed for minimizing the chemical consumption. Second, the obtained platform demonstrated enhanced electrochemical performance compared with commercially available Cu–based MOFs, with a potential of –230 versus –232 ev, logarithm of the current density of –3.6 versus –4.2 cm2, and electrochemical surface area of 75 versus 25 cm2per cm2of geometric area, respectively. Morphological and Raman analyses also revealed that the high concentration of defects in the obtained submicron Cu–BTC MOFs can contribute to their improved catalytic performance. Thus, our findings pave.the way to the low–cost synthesis of energy–efficient MOF–based catalysts for hydrogen production.

Rechargeable aqueous zinc ion hybrid capacitors(ZIHCs),as an up-and-comer aqueous electrochemical energy storage system,endure in their infancy because of the substandard reversibility of Zn anodes,structural deterioration of cathode materials,and narrow electrochemical stability ***,a scalable approach is described that addresses Zn-anode/electrolyte interface and cathode materials associated deficiencies and boosts the electrochemical properties of *** Zn-anode/electrolyte interface is self-regulated by alteration of the traditional Zn2+electrolyte with Na-based supporting salt without surrendering the cost,safety,and green features of the Zn-based system which further validates the excellent reversibility over 1100 h with suppressed hydrogen *** deficits of cathode materials were overcome by using a high-mass loaded,oxygen-rich,3D,multiscaled graphene-like carbon(3D MGC)*** to the multiscaled texture,high electronic conductivity,and oxygen-rich functional groups of 3D MGC,reversible redox capacitance was obtained with a traditional adsorption/desorption *** ZIHCs containing the modified electrolyte and an oxygen-rich 3D MGC cathode resulted in battery-like specific energy(203 Wh kg1 at 1.6 A g^(-1))and supercapacitor-type power capability(4.9 kW kg1 at 8 A g^(-1))with outstanding cycling durability(96.75%retention over 30000 cycles at 10 A g^(-1)).These findings pave.the way toward the utilization of highly efficient ZIHCs for practical applications.

The increasing rate of stress-related disorders, with mood disorders as major depression leading the way, and the social and well as economic consequences of these illnesses represent a growing threat to our society. A dysregulation of the body’s main stress system, the hypothalamic-pituitary-adrenal axis, is a major hallmark of depression, but so far no specific treatments tackling this mechanism are available. The co-chaperone FK506-binding protein 51（FKBP51） is a very promising target molecule for new drug therapies, as it directly regulates stress hormone activity. In humans, polymorphisms in the FKBP5 gene have been repeatedly linked to major depression and posttraumatic stress disorder. To further explore the interaction of FKBP51 with stress-related disorders, we have utilized conditional FKBP51 knockout or overexpression in combination with chronic social defeat stress, and tested the animals on different endocrine and behavioral parameters. Our results demonstrate that lowering FKBP51 levels in specific brain areas improves stress resilience. Treatment with the recently developed specific antagonist for FKBP51, SAFit2, lowered anxiety, improved stresscoping behavior and ameliorated the effects of chronic social defeat stress. Taken together, our results now pave.the way for tailored and specific treatment strategies that could be beneficial for improving stress resilience in health and disease.

The development of a new generation of nonantibody protein drug delivery systems requires site-directed conjugation strategies to produce homogeneous,reproducible and scalable *** that,the genetic addition of cysteine residues into solvent-exposed positions allows the thiol-mediated cysteine coupling of therapeutic drugs into protein-based ***,the high reactivity of unpaired cysteine residues usually reduces protein stability,consequently imposing the use of more methodologically demanding purification *** is especially relevant for disulfide-containing nanocarriers,as previously observed in ***,although many protein scaffolds and targeting ligands are also rich in disulfide bridges,the use of these methodologies over emerging non-antibody carrier proteins has been completely ***,we report the development of a simple and straightforward procedure for a onestep production and site-directed cysteine conjugation of disulfide-containing non-antibody thiolated carrier proteins(THIOCAPs).This method is validated in a fluorescent C-X-C chemokine receptor 4(CXCR4)-targeted multivalent nanocarrier containing two intramolecular disulfide bridges and one reactive cysteine residue strategically placed into a solvent-exposed position(THIO-T22-GFP-H6)for drug conjugation and in a humanized alternative intended for clinical applications(T22-HSNBT-H6).Thus,we produce very stable,homogeneous and fully functional antitumoral nanoconjugates(THIO-T22-GFP-H6-MMAE and T22-HSNBT-H6-MMAE)that selectively eliminate target cancer cells via ***,the developed methodology appears as a powerful tool for the rational engineering of emerging non-antibody,cell-targeted protein nanocarriers that contain disulfide bridges together with a solvent-exposed reactive cysteine(THIOCAP).This should pave.the way for the development of a new generation of stable,homogeneous and efficient nanomedicines.
