Development of a physiologically relevant dripping analytical method using simulated nasal mucus for nasal spray formulation analysis

Current methods for nasal spray formulations have been elementary evaluating the dripping characteristics of a formulation and have not assessed the behavior of the nasal formulation in the presence of varying types of mucus depending on the indication or diseased state. This research investigated the effects of nasal mucus on the dripping behavior of nasal formulations and focused on developing an improved in vitro analytical test method that is more physiologically relevant in characterizing nasal formulation dripping behavior. Method development was performed using simulated nasal mucus preparations for both healthy and diseased states as coatings for the dripping experiment representing a wide range of viscosity. Factors evaluated during development of this in vitro test method included amount of mucus, application of mucus, drying times, and compatibility of the mucus on a C18 Thin Layer Chromatography(TLC) substrate. The dripping behavior of nasal formulations containing a range of 1%Avicel to 3.5% Avicel was assessed by actuating the nasal spray on a perpendicular TLC plate coated with either healthy or diseased simulated nasal mucus. After actuation of the nasal spray, the dripping of the formulation on the coated TLC plate was measured after the plate was repositioned vertically. The method that was developed generated reproducible results on the dripping behavior of nasal formulations and provided critical information about the compatibility of the formulation with the nasal mucus for different diseased states, aiding in nasal spray formulation development and physical characterization of the nasal spray.

The study by Cao et al aimed to identify early second-trimester biomarkers that could predict gestational diabetes mellitus(GDM)development using advanced proteomic techniques,such as Isobaric tags for relative and absolute quantitation isobaric tags for relative and absolute quantitation and liquid chromatography-mass spectrometry liquid chromatography-mass *** analysis revealed 47 differentially expressed proteins in the GDM group,with retinol-binding protein 4 and angiopoietin-like 8 showing significantly elevated serum levels compared to *** these findings are promising,the study is limited by its small sample size(n=4 per group)and lacks essential details on the reproducibility and reliability of the protein quantification methods ***,the absence of experimental validation weakens the interpretation of the protein-protein interaction network identified through bioinformatics *** study's focus on second-trimester biomarkers raises concerns about whether this is a sufficiently early period to implement preventive interventions for *** GDM risk during the first trimester or pre-conceptional period may offer more clinical *** its limitations,the study presents valuable insights into potential GDM biomarkers,but larger,well-validated studies are needed to establish their predictive utility and generalizability.

Performance analysis of deep learning CNN models for disease detection in plants using image segmentation

Food security for the 7 billion people on earth requires minimizing crop damage by timely *** deep learningmodels forautomated detectionof diseases in plants suffer fromthe fatal flaw that once tested on independent data,their performance drops *** work investigates a potential solution to this problem by using segmented image data to train the convolutional neural network(CNN)*** compared to the F-CNN model trained using full images,S-CNN model trained using segmented imagesmore than doubles in performance to 98.6%accuracy when tested on independent data previously unseen by the models even with 10 disease *** only this,by using tomato plant and target spot disease type as an example,we show that the confidence of self-classification for S-CNN model improves significantly over F-CNN *** research work brings applicability of automated methods closer to non-experts for timely detection of diseases.
