Hydroxysafflor yellow A induced ferroptosis of Osteosarcoma cancer cells by HIF-1α/HK2 and SLC7A11 pathway

Osteosarcoma is a very serious primary bone cancer with a high death rate and a dismal prognosis.Since there is no permanent therapy for this condition,it is necessary to develop a cure.Therefore,this investigation was carried out to assess the impacts and biological functions of hydroxysafflor yellow A(HYSA)in osteosarcoma cell lines(MG63).In this investigational study,MG63 cells were utilized.Microarray experiments,quantitative polymerase chain reaction(qPCR),immunofluorescent staining,extracellular acidification rate(ECAR),oxygen consumption rate(OCR),glucose consumption,lactate production,and ATP levels,proliferation assay,5-Ethynyl-2′-deoxyuridine(EDU)staining,and Western blot were performed.In MG63 cells,HYSA lowered cell proliferation and metastasis rates,suppressed EDU cell number,and enhanced caspase-3/9 activity levels.HYSA reduced the Warburg effect and induced ferroptosis(FPT)in MG63 cells.Inhibiting ferroptosis diminished HYSA’s anti-cancer activities in MG63 cells.The stimulation of the HIF-1α/SLC7A11 pathway decreased HYSA’s anti-cancer activities in MG63 cells.HIF-1αis one target spot for HYSA in a model of osteosarcoma cancer(OC).HYSA altered HIF-1α’s thermophoretic activity;following binding with HYSA,HIF-1α’s melting point increased from~55°C to~60°C.HYSA significantly enhanced the thermal stability of exogenous WT HIF-1αwhile not affecting Mut HIF-1α,suggesting that ARG-311,GLY-312,GLN-347,and GLN-387 may be involved in the interaction between HIF-1αand HYSA.Conclusively,our study revealed that HYSA induced FPT and reduced the Warburg effect of OC through mitochondrial damage by HIF-1α/HK2/SLC7A11 pathway.HYSA is a possible therapeutic option for OC or other cancers.

Catalytic transfer hydrogenation(CTH)is a green and efficient pathway for selective hydrogenation of unsaturated aldehydes and ketones.However,managing the abilities of solid catalysts to adsorb substrates and to convert them into desired products is a challenging task.Herein,we report the synthesis of carbon coated LaFe_(0.92)Pd_(0.08)O_(3) composites(LFPO-8@C)for CTH of benzaldehyde(BzH)into benzyl alcohol(BzOH),using isopropanol(IPA)as hydrogen source.The coating with carbon improves the ability to adsorb/transfer reactants from solution to active sites,and the doping of Pd2+at Fe3+site strengthens the ability of LaFeO_(3) to convert BzH into BzOH.A balanced point between them(i.e.,abilities to adsorb BzH and to convert BzH into BzOH)is obtained at LFPO-8@C,which exhibits a BzOH formation rate of 3.88 mmol·gcat^(-1)·h^(-1) at 180℃ for 3 h,which is 1.50 and 2.72 times faster than those of LFPO-8 and LaFeO_(3)@C.A reaction mechanism is proposed,in which the acidic sites(e.g.,Fe^(4+),oxygen vacancy)are used for the activation of C=O bond of BzH and O-H bond of IPA,and the basic sites(e.g.,lattice oxygen)for the activation ofα-H(O-H)bond of IPA.

Analysis of China's Electricity Price and Electricity Burden of Basic Industries under the Carbon Peak Target before 2030

The Chinese government is deepening reformation of electricity prices during the 14th Five Year Plan period and has set a carbon emission reduction target of reaching carbon peak before 2030.In this context,will the carbon emission target influence electricity pricing and will electricity price influence competitiveness of Chinese main industries are two questions needing to be answered.This paper compares China's electricity price level with the selected major countries in the world,and four typical industries are selected to evaluate their electricity burden respectively.Then,the correlation between residential electricity price and industrial electricity price and the influencing factors is analyzed,from the perspectives of scale,structure and technology.According to the model obtained by regression analysis,the electricity price level and corresponding residential and industrial electricity burden in 2025 and 2030 are forecasted.Index Terms-Electricity burden,industrial electricity price,regression analysis,residential electricity price.

MdbHLH162 connects the gibberellin and jasmonic acid signals to regulate anthocyanin biosynthesis in apple

Anthocyanins are secondary metabolites induced by environmental stimuli and developmental signals.The positive regulators of anthocyanin biosynthesis have been reported,whereas the anthocyanin repressors have been neglected.Although the signal transduction pathways of gibberellin(GA)and jasmonic acid(JA)and their regulation of anthocyanin biosynthesis have been investigated,the cross-talk between GA and JA and the antagonistic mechanism of regulating anthocyanin biosynthesis remain to be investigated.In this study,we identified the anthocyanin repressor MdbHLH162 in apple and revealed its molecular mechanism of regulating anthocyanin biosynthesis by integrating the GA and JA signals.MdbHLH162 exerted passive repression by interacting with MdbHLH3 and MdbHLH33,which are two recognized positive regulators of anthocyanin biosynthesis.MdbHLH162 negatively regulated anthocyanin biosynthesis by disrupting the formation of the anthocyanin-activated MdMYB1-MdbHLH3/33complexes and weakening transcriptional activation of the anthocyanin biosynthetic genes MdDFR and MdUF3GT by MdbHLH3 and MdbHLH33.The GA repressor MdRGL2a antagonized MdbHLH162-mediated inhibition of anthocyanins by sequestering MdbHLH162 from the MdbHLH162-MdbHLH3/33 complex.The JA repressors MdJAZ1 and MdJAZ2 interfered with the antagonistic regulation of MdbHLH162 by MdRGL2a by titrating the formation of the MdRGL2a-MdbHLH162 complex.Our findings reveal that MdbHLH162 integrates the GA and JA signals to negatively regulate anthocyanin biosynthesis.This study provides new information for discovering more anthocyanin biosynthesis repressors and explores the cross-talk between hormone signals.

3D robust anisotropic diffusion filtering algorithm for sparse view neutron computed tomography 3D image reconstruction

The most critical part of a neutron computed tomography(NCT) system is the image processing algorithm,which directly affects the quality and speed of the reconstructed images.Various types of noise in the system can degrade the quality of the reconstructed images.Therefore,to improve the quality of the reconstructed images of NCT systems,efficient image processing algorithms must be used.The anisotropic diffusion filtering(ADF) algorithm can not only effectively suppress the noise in the projection data,but also preserve the image edge structure information by reducing the diffusion at the image edges.Therefore,we propose the application of the ADF algorithm for NCT image reconstruction.To compare the performance of different algorithms in NCT systems,we reconstructed images using the ordered subset simultaneous algebraic reconstruction technique(OS-SART) algorithm with different regular terms as image processing algorithms.In the iterative reconstruction,we selected two image processing algorithms,the Total Variation and split Bregman solved total variation algorithms,for comparison with the performance of the ADF algorithm.Additionally,the filtered back-projection algorithm was used for comparison with an iterative algorithm.By reconstructing the projection data of the numerical and clock models,we compared and analyzed the effects of each algorithm applied in the NCT system.Based on the reconstruction results,OS-SART-ADF outperformed the other algorithms in terms of denoising,preserving the edge structure,and suppressing artifacts.For example,when the 3D Shepp–Logan was reconstructed at 25 views,the root mean square error of OS-SART-ADF was the smallest among the four iterative algorithms,at only 0.0292.The universal quality index,mean structural similarity,and correlation coefficient of the reconstructed image were the largest among all algorithms,with values of 0.9877,0.9878,and 0.9887,respectively.

Regulated polarization degree of upconversion luminescence and multiple anti-counterfeit applications

Polarization upconversion luminescence（PUCL） of lanthanide ions (Ln3+) has been widely used in single particle tracking,microfluidics detection,three-dimensional displays,and so on.However,no effective strategy has been developed for modulating PUCL.Here we report a strategy to regulate PUCL in Ho3+-doped NaYF4single nanorods based on the number of upconversion photons.By constructing a multiphoton upconversion system for Ho3+,we regulate the degree of polarization （DOP） of PUCL from 0.590 for two-photon luminescence to 0.929 for three-photon upconversion luminescence （UCL）.Furthermore,our strategy is verified from cross-relaxation between Ho3+and Yb3+,excitation wavelength,excitation power density,and local site symmetry.And this regulation strategy of PUCL has also been achieved in Tm3+,with the DOP ranging from 0.233 for two-photon luminescence to 0.925 for four-photon UCL Besides,multi-dimensional anti-counterfeiting display has been explored with PUCL.This work provides an effective strategy for regulating PUCL and also provides more opportunities for the development of polarization display optical encoding,anti-counterfeiting,and integrated optical devices.

Prediction of chemical short-range order in high-/medium-entropy alloys

Chemical short-range orders(CSROs),as the built-in sub-nanoscale entities in a high-/medium-entropy alloy(H/MEA),have aroused an ever-increasing interest.With multi-principal elements in an H/MEA to form a complex concentrated solution,a variety of sub-systems of species exist to induce the metastable ordered compounds as candidates for ultimate CSROs.The issues remain pending on the origin of CSROs as to how to judge if CSRO will form in an H/MEA and particularly,what kind of CSROs would be stably produced if there were multiple possibilities.Here,the first-principles method,along with the proposed local formation energy calculation in allusion to the atomic-scale chemical heterogeneities,is used to predict the CSRO formation based on the mechanical stability,thermodynamic formation energy,and electronic characteristics.The simulations are detailed in an equiatomic ternary VCoNi MEA with three kinds of potential compounds,i.e.,L1_(1),L1_(2),and B2,in the face-centered cubic matrix.It turns out that L1_(1)is stable but hard to grow up so as to become the final CSRO.L1_(1)is further predicted as CSROs in CrCoNi,but unable to form in FeCoNi and CrMnFeCoNi alloys.These predictions are consistent with the experimental observations.Our findings shed light on understanding the formation of CSROs.This method is applicable to other H/MEAs to design and tailor CSROs by tuning chemical species/contents and thermal processing for high performance.

Feature Matching via Topology-Aware Graph Interaction Model

Feature matching plays a key role in computer vision. However, due to the limitations of the descriptors, the putative matches are inevitably contaminated by massive outliers.This paper attempts to tackle the outlier filtering problem from two aspects. First, a robust and efficient graph interaction model,is proposed, with the assumption that matches are correlated with each other rather than independently distributed. To this end, we construct a graph based on the local relationships of matches and formulate the outlier filtering task as a binary labeling energy minimization problem, where the pairwise term encodes the interaction between matches. We further show that this formulation can be solved globally by graph cut algorithm. Our new formulation always improves the performance of previous localitybased method without noticeable deterioration in processing time,adding a few milliseconds. Second, to construct a better graph structure, a robust and geometrically meaningful topology-aware relationship is developed to capture the topology relationship between matches. The two components in sum lead to topology interaction matching(TIM), an effective and efficient method for outlier filtering. Extensive experiments on several large and diverse datasets for multiple vision tasks including general feature matching, as well as relative pose estimation, homography and fundamental matrix estimation, loop-closure detection, and multi-modal image matching, demonstrate that our TIM is more competitive than current state-of-the-art methods, in terms of generality, efficiency, and effectiveness. Elemene, derived from Curcuma wenyujin, one of the "8 famous genuine medicinal materials of Zhejiang province," exhibits remarkable antitumor activity. It has gained wide recognition in clinical practice for effectiveness on tumors. Dr. XIE Tian introduced the innovative concept of "molecular compatibility theory" by combining Chinese medicine principles, specifically the "monarch, minister, assistant, and envoy" theory, with modern biomedical technology. This groundbreaking approach, along with a systematic analysis of Chinese medicine and modern biomedical knowledge, led to the development of elemene nanoliposome formulations. These novel formulations offer numerous advantages, including low toxicity, well-defined composition, synergistic effects on multiple targets, and excellent biocompatibility. Following the principles of the "molecular compatibility theory", further exploration of cancer treatment strategies and methods based on elemene was undertaken. This comprehensive review consolidates the current understanding of elemene's potential antitumor mechanisms, recent clinical investigations, advancements in drug delivery systems, and structural modifications. The ultimate goal of this review is to establish a solid theoretical foundation for researchers, empowering them to develop more effective antitumor drugs based on the principles of "molecular compatibility theory".
