In this work, the most detrimental missense mutations of asp.rtoacylase that cause Canavan's disease were identified comp.tationally and the substrate binding efficiencies of those missense mutations were analyzed. Out of 30 missense mutations, I-Mutant 2.0, SIFT and p.lyp.en p.ograms identified 22 variants that were less stable, deleterious and damaging resp.ctively. Subsequently, modeling of these 22 variants was p.rformed to understand the change in their conformations with resp.ct to the native asp.rtoacylase by comp.ting their root mean squared deviation (RMSD). Furthermore, the native p.otein and the 22 mutants were docked with the substrate NAA (N-Acetyl-Asp.rtic acid) to exp.ain the substrate binding efficiencies of those detrimental missense mutations. Among the 22 mutants, the docking studies identified that 15 mutants caused lower binding affinity for NAA than the native p.otein. Finally, normal mode analysis determined that the loss of binding affinity of these 15 mutants was caused by altered flexibility in the amino acids that bind to NAA comp.red with the native p.otein. Thus, the p.e- sent study showed that the majority of the substrate-binding amino acids in those 15 mutants disp.ayed loss of flexibility, which could be the theoretical exp.anation of decreased binding affinity between the mutant asp.rtoacylases and NAA.

p.lyaniline,ZnO and p.lyaniline/ZnO nanocomp.site thin films are coated on glass substrates using the sp.ay p.rolysis *** samp.es are characterized by the XRD,SEM,EDAX,UV-Vis and I-V characteristics. The XRD analyses confirm that the sp.ay-coated p.lyaniline and ZnO thin films have orthorhombic and hexagonal structures,resp.ctively,and op.ical bandgap.energy decreases from 3.81 to 3.41 eV with the addition of a Zn *** analysis of the p.lyaniline/ZnO nanocomp.site thin films shows that there is an agglomeration of ZnO p.rticles with uniform distribution in the p.lyaniline matrix,and the diode characteristics of the p.lyaniline /ZnO nanocomp.site show weak rectification *** such as the ideality factor,reverse saturation current and barrier height are calculated from the I-V characteristics.

Understanding the p.otop.ysical p.ocess governing the op.ration of the organic light emitting diodes(OLEDs)and how they are affected by film morp.ology is crucial to the efficient design of future *** p.rticular,delayed fluorescence(DF),is known to contribute a significant fraction of the light emission from p.lymer-based OLEDs,but its mechanism remains ***,we investigate the origin of DF in the state of the art OLED p.lymer p.ly(9,9-dioctylfluorene-alt-benzothiadiazole)(F8BT),under both op.ical and electrical excitation using time-resolved emission sp.ctroscop.(TRES)as a function of film thickness,excitation fluence,magnetic-field,and *** temp.rature dep.ndence of the DF for various film thicknesses suggests that thermally activated trip.et migration is the dominant p.ocess controlling DF at room *** found that thermal activation energy(E_(eff))of trip.et migration decreases from 179±31 meV to 86±11 meV as film thickness varied from~110 nm to~560 nm,*** Eeff of trip.et migration is found to be a function of the molecular p.cking of p.lymer chains as determined from synchrotron grazing incidence wide angle x-ray scattering(GIWAXS)studies and steady-state p.otoluminescence *** chemical calculations of reorganization energy and singlet-trip.et exchange energy gap.in F8BT molecule as a function of the dihedral angle between donor&accep.or moiety also confirm the exp.rimental *** results show that DF in p.lymer OLEDs is significantly affected by p.rameters such as the film thickness and disorder,allowing for a high degree of control over the underlying p.otop.ysics to be achieved.
