Lactobacillus kefiranofaciens and Lactobacillus kefiri are the key species in kefir for reversing oxytetracycline-induced intestinal damage in zebrafish

Increased accumulation of oxytetracycline (OTC) in environmental water bodies could potentially lead to its accumulation in human body， thereby damaging human intestinal tract. Dietary interventions could be helpful for recovery of intestinal morphology and function. Therefore， this study set zebrafish as model to explore the potential of kefir supplementation in the recovery of intestinal damage caused by exposure to OTC. In experiments by zebrafish， the rearing units used were glass tanks， each with volume of 5 liters. The tanks were stocked with 12 zebrafish each. For each treatment， there were eight replicate tanks. The zebrafish were treated with OTC followed by the addition of kefir to the food. The results showed positive improvements with kefir supplementation. Kefir treatment mitigated intestinal inflammation by reducing the levels of TNF-α， IL-6， and IL-1β; enhancing the activity of the antioxidant enzymes CAT， SOD， GPx and increasing the gene expression of intestinal tight junction proteins (ZO-1a and ZO-1b). These effects were beneficial for reversing reduced integrity of intestinal barrier caused by OTC. Moreover， kefir helped to reverse the disruption of gut microbiota caused by OTC and further impacted host metabolism. Specifically， Lactobacillus kefiranofaciens and Lactobacillus kefiri， which are derived from the kefir microbiota， were found to be enriched in the zebrafish intestine. This helped to inhibit the increased abundance of some Proteobacteria species induced by OTC treatment. Liver metabolomics analysis revealed that kefir improved OTC-induced disruptions in the tricarboxylic acid cycle， glycerophospholipid and amino acid metabolism. The differentially abundant metabolites identified included a total of 80 types after OTC exposure， with the abundance of 74 kinds significantly reversed following kefir treatment. 目的：探索不同类型先天性心脏病（ congenital heart defects, CHD）的环境危险因素,为深入研究CHD表型的危险因素提供方向,为预防CHD积累证据。方法：使用广东省CHD监测网2004年至2012年登记的来自于17个地市、34家医疗机构的3038例CHD活产儿及其1∶1对照儿数据,先后采用单因素分析、多因素条件Logistic回归,分析6类CHD的环境危险因素。结果：左向右分流类CHD的危险因素包括低出生体重（ OR=2.63,95%CI：2.04～3.39）或巨大儿（OR=2.21,95%CI：1.47～3.32）,早产（OR=1.95,95%CI：1.53～2.49）,多胎（OR=1.99,95%CI：1.22～3.26）,产母教育程度低、产母为工厂工人（OR=1.62,95%CI：1.32～1.98）,产次≥2（OR=1.38,95%CI：1.13～1.69）,有异常生育史（OR=2.29,95%CI：1.75～3.01）,孕早期3个月曾有发烧（OR=2.38,95%CI：1.26～4.48）,病毒感染（OR=1.80,95%CI：1.29～2.51）,使用药物（OR=1.73,95%CI：1.11～2.69）,被动吸烟（OR=1.69,95%CI：1.26～2.29）,接触化学制剂（OR=8.71,95%CI：2.33～32.58）,居住装修6个月内的居室（OR=2.56,95%CI：1.60～4.09）或居室距主要马路〈50 m（OR=1.40,95%CI：1.14～1.72）,父亲为工厂工人（OR=1.46,95%CI：1.23～1.73）。肺动脉流出道梗阻类CHD的危险因素包括活产儿低出生体重（OR=5.98,95%CI：2.88～12.44）或巨大儿（OR=6.56,95%CI：1.19～36.26）,产母教育程度低、产次≥2（OR=2.08,95%CI：1.03～4.22）,孕早期病毒感染（OR=4.30,95%CI：1.27～13.45）。左心室流出道梗阻类CHD危险因素为父亲为工厂工人（OR=6.01,95%CI：1.05～34.59）。大动脉转位（transposition of the great arteries, TGA）的危险因素包括活产儿低出生体重（OR=12.93,95%CI：1.14～146.26）,产母教育程度低、职业为工厂工人（OR=3.69,95%CI：1.53～8.91）。合并心内分流型CHD的危险因素包括产次=2（OR=3.45,95%CI：1.42～8.38）。其他类CHD的危险因素包括巨大儿（OR=4.87,95%CI：1.19～19.94）、产母既往有异常生育史（OR=2.96,95%CI：1.14～7.68）、孕早期3个月有病毒感染（OR=4.92,95%CI：1.56～15.47）、用药（OR=4.90,95%CI：1.22～19.77）、被动吸烟（OR=10.31,95%CI：1.25～85.05）。结论：不同CHD类的危险因素存在差异,深入研究CHD的相关环境危险因素及其影响机制需有针对性,预防CHD需特别注意避免影响多类CHD或临床治疗效果较差CHD类的环境危险因素暴露,并加强已暴露人群的产前检查。

Lactobacillus from fermented bamboo shoots prevents inflammation in DSS-induced colitis mice via modulating gut microbiome and serum metabolites

Fermented bamboo shoots(FBS)is a region-specific food widely consumed in Southwestern China,with Lactobacillus as the predominant fermenting ***,the probiotic potential of Lactobacillus derived from FBS reminds largely unexplored,especially for diseases with a low prevalence in areas consuming FBS,namely,inflammatory bowel *** this study,Lactiplantibacillus pentosus YQ001 and Lentilactobacillus senioris YQ005 were screening by in vitro probiotic tests to further investigate the probioticlike bioactivity in dextran sulfate sodium(DSS)-induced ulcerative colitis(UC)*** exhibited more positive probiotic effects than Lactobacillus rhamnosus GG in preventing intestinal inflammatory *** results revealed that both strains improved the abundance of deficient intestinal microbiota in UC mice,including Muribaculaceae and *** the serum metabolome,they modulated the DSS-disturbed levels of metabolites,with significant increment of cinnamic ***,they reduced the expression levels of interleukin-1β(IL-1β),interleukin-6(IL-6)inflammatory factors and increased zonula occludens-1(ZO-1),Occludin,and cathelicidin-related antimicrobial peptide(CRAMP)in the ***,these results demonstrated that Lactobacillus *** derived from FBS showed promising probiotic activity based on the gut microbiome homeostasis modulation,anti-inflammation and intestinal barrier protection in UC mice.

How to apply ex-vivo split liver transplantation safely and feasibly: A three-step approach

BACKGRouND Given the current organ shortage crisis,split liver transplantation(SLT)has emerged as a promising alternative for select end-stage liver disease *** To introduce an ex-vivo liver graft splitting approach and evaluate its safety and feasibility in *** A retrospective analysis was conducted on the liver transplantation data from cases performed at our center between April 1,2022,and May 31,*** study included 25 SLT cases and 81 whole liver transplantation(WLT)*** ex-vivo liver splitting was employed for SLT graft procurement in three *** outcomes were determined,including liver function parameters,postoperative complications,and perioperative *** comparisons for categorical variables were performed using theχ²-*** In the study,postoperative complications in the 25 SLT cases included hepatic artery thrombosis(n=1)and pulmonary infections(n=3),with no perioperative *** contrast,among the 81 patients who underwent WLT,complications included perioperative mortality(n=1),postoperative pulmonary infections(n=8),abdominal infection(n=1),hepatic artery thromboses(n=3),portal vein thrombosis(n=1),and intra-abdominal bleeding(n=5).Comparative analysis demonstrated significant differences in alanine aminotransferase(176.0 vs 73.5,P=0.000)and aspartate aminotransferase(AST)(42.0 vs 29.0,P=0.004)at 1 wk postoperatively,and in total bilirubin(11.8 vs 20.8,P=0.003)and AST(41.5 vs 26.0,P=0.014)at 2 wk ***,the overall incidence of complications was comparable between the two groups(P>0.05).CONCLUSION our findings suggest that the total ex-vivo liver graft splitting technique is a safe and feasible approach,especially under the expertise of an experienced transplant *** approach developed by our center can serve as a valuable reference for other transplantation centers.
