Amaranthus roxburghianus root extract in combination with piperine as a potential treatment of ulcerative colitis in mice

OBJECTIVE： The present work was undertaken to determine the effects of Amaranthus roxburghianus Nevski. （Amaranthaceae） root alone and in combination with piperine in treating ulcerative colitis （UC） in mice. METHODS： Swiss albino mice were divided into seven groups （n = 6）. Standard group received prednisolone （5 mg/kg, intraperitoneally）. Treatment groups received hydroalcoholic extract of roots of A. roxburghianus （50 and 100 mg/kg, per oral） and a combination of hydroalcoholic extract of roots of A. roxburghianus （50 and 100 mg/kg, per oral） and piperine （5 mg/kg, per oral）. Ulcer index, colitis severity, myeloperoxidase （MpO）, malondialdehyde and glutathione were estimated from blood and tissue. Column chromatography of the extract was done and purified fractions were analyzed by gas chromatography-mass spectroscopy （GC-MS）. RESULTS： Treatment with the combination of hydroalcoholic extract of A. roxburghianus and piperine showed minimal ulceration, hemorrhage, necrosis and leucocyte infiltration by histopathological observation. Acetic acid increased MpO levels in blood and colon tissue to 355 U/mL and 385 U/mg, respectively. The combination of hydroalcoholic extract of A. roxburghianus （100 mg/kg） and piperine （5 mg/kg） significantly decreased MpO in blood and tissue to 182 U/mL and 193 U/mg, respectively （p 〈 0.05）. Similarly, this combination significantly reduced malondialdehyde levels and increased glutathione levels in blood and tissue. Various phytoconstituents were detected by GC-MS. CONCLUSION： The combination of hydroalcoholic extract of A. roxburghianus and piperine is effective in the treatment of UC and the effects are comparable with the standard drug prednisolone. 4H-pyran-4-one, 2,3-dihydro-3,5-dihydroxy-6-methyl, eugenol and benzene, and 1-（1,5-dimethyl-4-hexenyl）-4-methyl are reported having analgesic, anti-inflammatory, and antioxidant properties; they may play a role in the biological activity of A. roxburghianus root.

In malaria-endemic regions,people often get exposed to various pathogens simultaneously,generating coinfection *** such scenarios,overlapping symptoms pose serious diagnostic *** delayed diagnosis may lead to an increase in disease severity and catastrophic *** coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has affected various areas globally,including malaria-endemic *** plasmodium and SARS-CoV-2 co-infection and its effect on health are yet *** present a case report of a previously healthy,middle-aged individual from the malaria-endemic area who suffered SARS-CoV-2 and plasmodium falciparum *** patient developed severe disease indications in a short time *** patient showed neurological symptoms,altered hematological as well as liver-test parameters,and subsequent death in a narrow time *** hereby discuss the various aspects of this case regarding treatment and hematological ***,we have put forward perspectives related to the mechanism behind severity and neurological symptoms in this fatal parasite-virus co-infection *** malaria-endemic regions,due to overlapping symptoms,suspected COVID-19 patients should also be monitored for diagnosis of malaria without any *** SARS-CoV-2 and plasmodium co-infection could increase the disease severity in a short time *** treatment,dexamethasone may not help in severe cases having malaria as well as COVID-19 positive status and needs further exploration.

paulownia is a genus of fast-growing and multipurpose tree species that is native to China. Due to their rapid growth and value in the timber market, many paulownia species are cultivated in several temperate zones worldwide. Economic importance of paulownia is increasing as new uses and related products are developed. It is also suitable as a lignocellulosic feedstock crop for the bioethanol industry in the Southeastern USA. A number of paulownia species are valuable sources of secondary metabolites including flavonoids with high antioxidant activities. A high demand for planting material in domestic and international markets for afforestation and bioenergy production has necessitated the development of efficient micropropagation protocols for rapid and mass propagation of paulownia. Over the past several decades, research on paulownia species has been conducted to develop micropropagation, somatic embryogenesis and genetic transformation protocols for use in agroforestry and reforestation programs. Given the economic importance and current and potential future uses of paulownia, this paper reviews the development of biotechnological approaches for plant propagation and genetic improvement, and antioxidant potential of secondary metabolites occurring in species.

Background: Rabies is a dreaded disease and an estimated 55,000 people die of rabies every year. Himachal pradesh is in the North bordering China and is predominantly rural and hilly. Villages are near forests, where wild reservoirs of rabies exist. Since health facilities are not accessible easily, we need to innovate on existing schedules of rabies vaccination keeping in view the compliance of the patients and affordability so as to give them the best possible option of treatment. In the year 2006 and 2007, we, at DDU Hospital Shimla, experienced a severe shortage of rabies vaccine and patients were running from pillar to post to fetch rabies vaccine. At the same time, we learnt that some of the patients died because either they were not able to purchase the vaccine, mostly because of its high cost, $35, or they ignored the animal bites and did not seek the treatment. Since last year, we have been experiencing non-availability of rabies immunoglobulins (RIGs) in the market and have to innovate new schedules and techniques to save lives of the patients. Methods: During shortage of rabies vaccine in 2008, we contemplated to start a low cost intra-dermal (ID) clinic so as to make rabies vaccine affordable as intramuscular (IM) vaccination cost five times more than ID vaccination. But, there were three main hurdles. One hurdle was the non-availability of rabies vaccine vials having written on them “For IM/ID use” and another hurdle was only fewer animal bite patients attending the DDU Hospital, sometimes only one or two per day, which was insufficient to open a vaccine vial and distribute among them. The third problem being faced was reluctance of the hospital doctors to prescribe ID vaccine as this was not the practice at higher teaching institutions, including medical colleges. A very rare and serious complication of shoulder dislocation is a lesion to the axillary artery in the elderly population, whose vascular structures have become less flexible. Axillary artery injury secondary to anteroinferior shoulder dislocation is much rarer, especially in the young people. proper recognition and treatment of this entity of- fers a full recovery to the patient. present report highlights the possibility of axillary artery injury with anteroinferior shoulder dislocation. A few case reports and small case se- ries of this injury have been reviewed. And recommenda- tions for management have been brought up to date, in line with current thinking.

Randomized, double-blinded, placebo-controlled trial evaluating simethicone pretreatment with bowel preparation during colonoscopy

BACKGROUND The presence of small air bubbles and foam are an impediment to a successful colonoscopy. They impair an endoscopist’s view and diminish the diagnostic accuracy of the study. This has been particularly noted to be of concern with the switch to lower volume polyethylene glycol (pEG) and bisacodyl combination preparation. AIM To evaluate the effect of oral simethicone addition to bowel preparation on intraluminal bubbles reduction during colonoscopy. METHODS Described is a prospective, randomized, multi-center, double-blinded, placebocontrolled study to evaluate the use of premixed simethicone formulation with split-regimen, low-volume pEG-bisacodyl combination bowel preparation for 168 outpatients undergoing screening, surveillance, and diagnostic colonoscopies. primary outcome includes evaluation of bubbles during colonoscopy graded using the Intraluminal Bubbles Scale. Secondary outcomes include evaluation of the Boston Bowel preparation Scale (BBpS), total number of polyps, polyp size differentiation, polyp laterality, adenoma detection, mass detection, cecal insertion time, withdrawal time, and patient-reported adverse events. RESULTS Higher Intraluminal Bubbles grades III and IV (less than 75% of the mucosa cleared of bubbles/foam requiring intervention with simethicone infused wash) were detected in the placebo group [Simethicone n = 4/84 vs placebo n = 20/84 (p = 0.007)]. BBpS total score was 7.42 [standard deviation (SD)=± 1.51] in the simethicone group and 7.28 (SD =± 1.44) in the placebo group (p = 0.542) from a total of 9. Significantly higher number of adenomas were detected in the simethicone group (p = 0.001). CONCLUSION The addition of simethicone to bowel preparation is well advised for its antifoaming properties. The results of this study suggest that addition of oral simethicone can improve bowel wall visibility.

Dynamism of Antimicrobial Activity of Actinomycetes—A Case Study from Undisturbed Microbial Niche

Escalation of drug resistant microbes (bacteria) had forced researchers to search new and improved therapeutic compounds from different possible sources, including metabolites secreted by the actinomycetes. The aim of this study was to evaluate the pattern of antimicrobial actinomycetes from physiologically distinct soil of different geographical locations. Forty five soil samples were collected from 5 districts of Gujarat including two sanctuaries as source of survey for bioactive actinomycetes. Crowded plate technique was used for isolation and Agar cylinder method was employed for the antimicrobial screening. A total of 171 actinomycetes were isolated and screened against eighteen pathogens responsible for causing diseases in plants and humans. Results indicate that 79% of the isolates were active against at least one of the eighteen tested pathogens. Some of the actinomycetes strain had shown strong antibacterial and antifungal activity which may be a good source of obtaining novel antimicrobials.
