Psoriasis is a chronic inflammatory skin condition that has a fairly wide range of clinical *** psoriasis,which is the most common manifestation of psoriasis,is located on one end of the spectrum,dominated by adaptive immune responses,whereas the rarer pustular psoriasis lies on the opposite end,dominated by innate and autoinflammatory immune *** recent years,genetic studies have identified six genetic variants that predispose to pustular psoriasis,and these have highlighted the role of IL-36 cytokines as central to pustular psoriasis *** this review,we discuss the presentation and clinical subtypes of pustular psoriasis,contribution of genetic predisposing variants,critical role of the IL-36 family of cytokines in disease pathophysiology,and treatment perspectives for pustular *** further outline the application of appropriate mouse models for the study of pustular psoriasis and address the outstanding questions and issues related to our understanding of the mechanisms involved in pustular psoriasis.

Most human pathogens originate from non-human hosts and certain pathogens persist in animal *** transmission of such pathogens to humans may lead to self-sustaining chains of *** pathogens represent the highest risk for future *** their prevention,the transmission over the species barrier—although rare—should,by all means,be *** the current COVID-19 pandemic,surprisingly though,most of the current research concentrates on the control by drugs and vaccines,while comparatively little scientific inquiry focuses on future *** in 2012,the World Bank recommended to engage in a systemic One Health approach for zoonoses control,considering integrated surveillance-response and control of human and animal diseases for primarily economic *** examples,like integrated West Nile virus surveillance in mosquitos,wild birds,horses and humans in Italy show evidence of financial savings from a closer cooperation of human and animal health *** a zoonotic origin can be ascertained for the COVID-19 pandemic,integrated wildlife,domestic animal and humans disease surveillance-response may contribute to prevent future *** conclusion,the earlier a zoonotic pathogen can be detected in the environment,in wildlife or in domestic animals;and the better human,animal and environmental surveillance communicate with each other to prevent an outbreak,the lower are the cumulative costs.

Smart fluid manipulation with automatically controlled paper valves will enable automated and multi-step immunoassays on paper-based microfluidic *** this work,we present an integrated paper-based microfluidic platform with shape-memory polymer(SMP)-actuated fluid valves capable of automated colorimetric enzyme-linked immunosorbent assays(ELISAs).A single-layer microfluidic paper-based analytical device(μPAD)was designed to store all the reagents on the chip,and sequentially transfer reagents to a paper test zone following a specific ELISA protocol through automatic fluidic flow control by the multiple SMP-actuated *** actuation of a paper valve was based on the thermally responsive,duel-state shape transformation of a SMP sheet attached to the root of a paper cantilever beam for driving a hydrophilic paper bridge to connect and disconnect two paper channels.A portable colorimetric reader was developed to control the on-chip valve operations,quantify the colorimetric signal output,display the assay result,and wirelessly transmit the data to a smart phone for the application of *** operations of the paper valve and the entireμPAD were demonstrated with success rates of 97%and 93%,respectively.A detection mechanism for valve malfunction was designed and confirmed effective to identify any mal-operation of individual valves,thus rendering our platform reliable in real *** device calibration,we conducted direct ELISAs of rabbit IgG in phosphate-buffered saline(PBS),and achieved a low limit of detection(LOD)of 27 pM(comparable to that of standard and paper-based ELISAs).In order to demonstrate the clinical application of our multi-step immunoassay platform,we also conducted sandwich ELISAs to quantify the protein level of an inflammatory cytokine,namely tumor necrosis factor(TNF)-α,in surgically injured laryngeal tissues of *** protein levels of TNF-αwere shown similar between the conventional andμPAD ELISAs.

Fifty years ago, Hans A. Panofsky published a paper entitled Determination of stress from wind and temperature measurements. In his famous paper, he presented a new profile function for the mean horizontal wind speed under the condition of diabatic stratification that includes his integral similarity function. With his integral similarity function, he opened the door for Monin-Obukhov scaling in a wide range of micrometeorological and microclimatological applications. In a historic survey ranging from the sixties of the past century down to the present days, we present integral similarity functions for momentum, sensible heat, and water vapor for both unstable and stable stratification, where on the one hand free convection condition and on the other hand strongly stable stratification are addressed.

AIM:To determine correlations between family history,clinical features and mutational status of genes involved in the progression of colorectal cancer(CRC).METHODS:Histo-pathological features and molecular changes[KRAS,BRAF and CTNNB1 genes mutations,microsatellite instability(MSI)phenotype,expression of mismatch repair(MMR)and mucin(MUC)5AC proteins,mutation and expression analysis of TP53,MLH1promoter hypermethylation analysis]were examined in a series of 51 unselected Tunisian CRC patients,10 of them had a proven or probable hereditary disease,on the track of new tumoral markers for CRC susceptibility in Tunisian ***:As expected,MSI and MMR expression loss were associated to the presence of familial CRC(75%vs 9%,P<0.001).However,no significant associations have been detected between personal or familial cancer history and KRAS(codons 12 and 13)or TP53(exons 4-9)alterations.A significant inverse relationship has been observed between the presence of MSI and TP53 accumulation(10.0%vs 48.8%,P=0.0335)in CRC tumors,suggesting different molecular pathways to CRC that in turn may reflect different environmental ***,MUC5AC expression was significantly associated to the presence of MSI(46.7%vs 8.3%,P=0.0039),MMR expression loss(46.7%vs8.3%,P=0.0039)and the presence of familial CRC(63%vs 23%,P=0.039).CONCLUSION:These findings suggest that MUC5AC expression analysis may be useful in the screening of Tunisian patients with high risk of CRC.

Due to the presence of plastids, eukaryotic photosynthetic cells represent the most highly compartmentalized eukaryotic cells. This high degree of compartmentation requires the transport of solutes across intracellular membrane systems by specific membrane transporters. In this review, we summarize the recent progress on functionally characterized intracellular plant membrane transporters and we link transporter functions to Arabidopsis gene identifiers and to the transporter classification system. In addition, we outline challenges in further elucidating the plant membrane permeome and we provide an outline of novel approaches for the functional characterization of membrane transporters.

CAIPIRINHA-accelerated T1w 3D-FLASH for small-bowel MR imaging in pediatric patients with Crohn's disease:assessment of image quality and diagnostic performance

Background:The“Controlled Aliasing In Parallel Imaging Results In Higher Acceleration”(CAPIRINHA)technique greatly accelerates T1w 3D fast low angle shot(FLASH)scans while maintaining high image *** studied image quality and conspicuity of inflammatory lesions on CAIPIRINHA-accelerated imaging for pediatric small-bowel magnetic resonance imaging(MRI).Methods:Forty-four consecutive patients(mean 14±3 years,18 girls)underwent small-bowel MRI(MR enterography,MRE)at 1.5 T including diffusion-weighted imaging(DWI),contrast-enhanced CAIPIRINHA 3D-FLASH and standard 2D-FLASH ***’s disease(CD)was confirmed in 26 patients,18 patients served as *** blinded readings were performed for grading of image quality and conspicuity of CD lesions on CAIPIRINHA FLASH and standard FLASH images in comparison to a reference standard comprising imaging and endoscopic ***:CAIPIRINHA FLASH yielded significantly higher image quality with good inter-observer agreement(κ=0.68)and showed better visual delineation in 40%of the assessed bowel lesions,as compared to standard *** was full agreement for identification of CD patients between CAIPIRINHA and standard *** FLASH detected two small-bowel lesions that were not seen on standard *** revealed additional inflammatory lesions inconspicuous on contrast-enhanced *** showed an overall diagnostic accuracy of 93%.Conclusion:We present first evidence that CAIPIRINHA greatly accelerates T1w imaging in paediatric MRE without trade-off in image quality or lesion conspicuity and is thus preferable to standard FLASH imaging.

X-ray structure of methyl-CpG binding domain(MBD)of MeCP2,an intrinsically disordered protein(IDP)involved in Rett syndrome,offers a rational basis for defining the spatial distribution for most of the sites where mutations responsible of Rett syndrome,RTT,*** have ascribed pathogenicity for mutations of amino acids bearing positively charged side chains,all located at the protein-DNA interface,as positive charge removal cause reduction of the MeCP2-DNA adduct *** of the frequent proline replacements,outside the DNA contact moiety of MBD,can be attributed to the role of this amino acid for maintaining both unfolded states for unbound MeCP2 and,at the same time,to favor some higher conformational order for stabilizing structural determinants required by protein *** hypotheses can be extended to transcription repressor domain,TRD,the other MeCP2-DNA interaction site and,in general,to all the IDP that interact with nucleic acids.

Background Vector-borne diseases cause morbidity and mortality globally. However, some areas are more impacted than others, especially with climate change. Controlling vectors remains the primary means to prevent these diseases, but new, more effective tools are needed. The World Health Organization (WHO) prioritized evaluating novel control methods, such as sterile insect technique (SIT) for control ofAedes-borne diseases. In response, a multiagency partnership between the U.S. Centers for Disease Control and Prevention (CDC), the Special Programme for Research and Training in Tropical Diseases (TDR), WHO, and the International Atomic Energy Agency (IAEA) supported the operational implementation and evaluation of SIT againstAedes aegypti and arboviral diseases in the Pacific through a consortium of regional partners (PAC-SIT Consortium).Main text A workshop was held from 2 to 6 May 2023, during which PAC-SIT country participants, researchers, and stakeholders in SIT, scientific advisory committee members, and organizational partners came together to review the principles and components of SIT, share experiences, visit field sites and the SIT facility, and officially launch the PAC-SIT project. Working in groups focused on entomology, epidemiology, and community engagement, participants addressed challenges, priorities, and needs for SIT *** The PAC-SIT workshop brought together researchers and stakeholders engaged in evaluating SIT for arboviral diseases in the Pacific region and globally. This training workshop highlighted that many countries are actively engaged in building operational capacities and phased testing of SIT. The workshop identified a key need for robust larger-scale studies tied with epidemiological endpoints to provide evidence for the scalability and impact on mosquito-borne diseases.
