Developmental environment contributes to rapid trait shifts among newly colonized subterranean habitats

Recent colonization of extreme environments provides unique opportunities to study the early steps of adaptation and the potential for rapid convergent ***,phenotypic shifts during recent colonization may also be due to plasticity in response to changes in the rearing ***,we analyzed a suite of morphological and behavioral traits in paired surface,subterranean,and facultatively subterranean Mexican tetras(Astyanax mexicanus)from recent introductions in two separate watersheds outside of their native *** found a variety of phenotypic and behavioral shifts between subterranean and surface populations that are similar to those observed in relatively ancient populations in *** this rapid morphological divergence,we found that most of these trait differences were due to plasticity in response to rearing *** most trait assays in common-garden,lab-raised fish indicated that phenotypic shifts in wild fish were the result of plasticity,we also found evidence of genetic control in several traits present in subterranean ***,wall-following behavior,an important subterranean foraging behavior,was greater in lab-born subterranean fish than in lab-born surface fish,suggesting rapid divergence of this trait between subterranean and surface ***,this study sheds light on the early steps of subterranean evolution,identifies potential rapid behavioral evolution,and suggests that plasticity in traits involving exploratory behavior may facilitate subterranean colonization.

nvestigating the role of exiting vehicles and turn indicator usage in gap acceptance at single-lane roundabouts

Roundabouts are becoming a preferred form of intersection control type due to their safety and capacity benefits. Yield control at roundabout entries inherently reduces fuel consumption and emissions when compared to other conventional intersection types. However, these benefits can be limited by poor driver behavior and judgment when entering the roundabout. This research addresses the nature of gap distributions and use of turn indicators by exiting vehicles at three single-lane roundabouts in Vermont, New York, and Alaska. Presented here is a comparison of vehicle headways measured at two different locations in each roundabout. Rejected headways are analyzed in the context of priority abstaining events when entering drivers yield to vehicles exiting on the same leg of the roundabout suggesting that ＂true＂ critical gaps are being overestimated. Results indicate that exiting vehicles, particularly those that do not use their turn indicators when departing from the major-stream of traffic have an influence on the entry decision of drivers on the same approach. This behavior is of particular concern for intersection efficiency （i.e., delay） and sustainability （i.e., excess fuel consumption and emissions）. Results prompt the consideration of more consistent guidance on and enforcement of turn indicator use during roundabout negotiations.

Spectral square moments of a resonance sum for Maass forms

Let f be a Maass cusp form for Г0（N） with Fourier coefficients 1 k2. λf（n） and Laplace eigenvalue 1/4 ＋k2 For real α≠0 and β 〉 0, consider the sum Sx（f; α,β） = ∑n λf（n）e（αnβ）φ（n/X）, where φ is a smooth function of compact support. We prove bounds for the second spectral moment of Sx （f;α, β）, with the eigenvalue tending towards infinity. When the eigenvalue is sufficiently large, we obtain an average bound for this sum in terms of X. This implies that if f has its eigenvalue beyond X1/2＋ε, the standard resonance main term for Sx（f; ±2√q 1/2）, q ∈Z＋, cannot appear in general. The method is adopted from proofs of subconvexity bounds for Rankin-Selberg L-functions for GL（2） × GL（2）. It contains in particular a proof of an asymptotic expansion of a well-known oscillatory integral with an enlarged range of Kε≤ L≤ K1-ε. The same bounds can be proved in a similar way for holomorphie cusp forms.

A comparison of cyanoacrylate to sutures for wound closure following acoustic transmitter insertion in rainbow trout

Tag retention,wound healing,wound closure,and tagged fish survival are critically important after surgical implantation of acoustic transmitters in *** study compared the use of sutures to cyanoacrylate adhesives and bandages for post-surgery wound *** rainbow trout Oncorhynchus mykiss were assigned to one of seven treatments:a control group where fish were only handled,two suture groups(tagged with dummy acoustic transmitters or non-tagged),and four cyanoacrylate adhesive groups(non-tagged,tagged with adipose fin applied as a biological bandage,tagged with anal fin applied,and non-tagged with water-proof bandage).Tag retention was significantly improved in adhesive with anal fin(89%)and tagged suture(78%)treatments when compared to only 11%tag retention in the adhesive with adipose fin *** redness severity increased over the 14 weeks of the study in the tagged and sutured fish,and in those receiving the waterproof *** redness severity was also significantly higher in the waterproof bandage and tagged and sutured treatments than both tagged adhesive treatments at the end of the *** gape was significantly more severe in the adhesive with adipose treatment than in the control,suture,tagged suture,and adhesive treatments,but no significant differences were observed between the adhesive with adipose treatment and the adhesive with anal fin or bandage *** gape never completely closed in the adhesive with adipose group,while both the tagged suture treatment and tagged adhesive with anal treatment wounds completely closed between four-and six-weeks *** results of this study indicate that cyanoacrylate adhesive used in conjunction with anal fin tissue is a suitable post-surgical treatment.

Town-scale microbial sewer community and H2S emissions response to common chemical and biological dosing treatments

Controlling hydrogen sulfide(H2S)odors and emissions using a single,effective treatment across a town-scale sewer network is a challenge faced by many water *** of a sewer diversion provided the opportunity to compare the effectiveness of magnesium hydroxide(Mg(OH)2)and two biological dosing compounds(Bioproducts A and B),with different modes of action(MOA),in a field-test across a large sewer ***(OH)2 increases sewer p H allowing suppression of H2S release into the sewer environment while Bioproduct A acts to disrupt microbial communication through quorum sensing(QS),reducing biofilm *** B reduces H2S odors by scouring the sewer of fats,oils and grease(FOGs),which provide adhesion points for the microbial *** revealed that only Mg(OH)2 altered the microbial community structure and reduced H2S emissions in a live sewer system,whilst Bioproducts A and B did not reduce H2S emissions or have an observable effect on the composition of the microbial community at the dosed *** results recommend in situ testing of dosing treatments before implementation across an operational system.

Nigral dopaminergic neuron replenishment in adult mice through Ve-cadherin-expressing neural progenitor cells

The function of dopaminergic neurons in the substantia nigra is of central importance to the coordination of movement by the brain＇s basal ganglia circuitry. This is evidenced by the loss of these neurons, resulting in the cardinal motor deficits associated with Parkinson＇s disease. In order to fully understand the physiology of these key neurons and develop potential therapies for their loss, it is essential to determine if and how dopaminergic neurons are replenished in the adult brain. Recent work has presented evidence for adult neurogenesis of these neurons by Nestin＋/Sox2 neural progenitor cells. We sought to further validate this finding and explore a potential atypical origin for these progenitor cells. Since neural progenitor cells have a proximal association with the vasculature of the brain and subsets of endothelial cells are Nestin＋, we hypothesized that dopaminergic neural progenitors might share a common cell lineage. Therefore, we employed a Ve-cadherin promoter-driven CReeRT2：TIlox/Tlox transgenic mouse line to ablate the tyrosine hydroxylase gene from endothelial cells in adult animals. After 26 weeks, but not 13 weeks, following the genetic blockade of tyrosine hydroxylase expression in Ve-cadherin＋ cells, we observed a significant reduction in tyrosine hydroxylase＋ neurons in the substantia nigra. The results from this genetic lineage tracing study suggest that dopaminergic neurons are replenished in adult mice by a Ve-cadherin＋ progenitor cell population potentially arising from an endothelial lineage.

The multiplexed,point-of-care measurement of specific antibodies could improve the speed with which diseases are diagnosed and their treatment *** this end,we are developing e-DNA scaffold sensors,which consist of a rigid,nucleic acid“scaffold”attached on one end to an electrode and presenting both a redox reporter and an epitope on the *** the absence of antibody,the reporter efficiently transfers electrons when interrogated ***-induced steric hindrance limits movement,reducing electron transfer in a manner that is both easily measured and quantitatively related to target *** we have used monoclonal antibodies to explore the analytical performance of e-DNA sensors,showing that they support the rapid,single-step,quantitative detection of multiple antibodies in small volume ***,in contrast,we employ authentic human samples to better explore the platform’s clinical ***,we developed e-DNA sensors targeting three HIV-specific antibodies and then compared the analytical and clinical performance of these against those of gold standard serological *** so we find that,although the multistep amplification of an eLISA leads to a lower detection limits,the clinical sensitivity of eLISAs,e-DNA sensors and lateral-flow dipsticks are indistinguishable across our test *** thus appears that,by merging the quantitation and multiplexing of eLISAs with the convenience and speed of dipsticks,e-DNA scaffold sensors could significantly improve on current serological practice.
