Alternative relay regulates the adenosine triphosphatase activity of Locusta migratoria striated muscle myosin

Locust(Locusta migratoria)has a single striated muscle myosin heavy chain(Mhc)gene,which contains 5 clusters of alternative exclusive exons and 1 differently included penultimate exon.The alternative exons of Mhc gene encode 4 distinct regions in the myosin motor domain,that is,the N-terminal SH3-like domain,one lip of the nucleotide-binding pocket,the relay,and the converter.Here,we investigated the role of the alternative regions on the motor function of locust muscle myosin.Using Sf9-baculovirus protein expression system,we expressed and purified 5 isoforms of the locust muscle myosin heavy meromyosin(HMM),including the major isoform in the thorax dorsal longitudinal flight muscle(FL1)and 4 isoforms expressed in the abdominal intersegmental muscle(AB1 to AB4).Among these 5 HMMs,FL1-HMM displayed the highest level of actin-activated adenosine triphosphatase(ATPase)activity(hereafter referred as ATPase activity).To identify the alternative region(s)responsible for the elevated ATPase activity of FL1-HMM,we produced a number of chimeras of FL1-HMM and AB4-HMM.Substitution with the relay of AB4-HMM(encoded by exon-14c)substantially decreased the ATPase activity of FL1-HMM,and conversely,the relay of FL1-HMM(encoded by exon-14a)enhanced the ATPase activity of AB4-HMM.Mutagenesis showed that the exon-14a-encoded residues Gly474 and Asn509 are responsible for the elevated ATPase activity of FL1-HMM.Those results indicate that the alternative relay encoded by exon-14a/c play a key role in regulating the ATPase activity of FL1-HMM and AB4-HMM.

Electrochemical urea oxidation reaction(UOR) is a promising alternative to oxygen evolution reaction(OER) for realizing energy-saving hydrogen production. Developing efficient electrocatalysts for UOR becomes a central challenge. Recently, amorphous materials have been extensively used as UOR catalysts because of their numerous defective sites and flexible electronic properties. In this review, recent advancements in the development of amorphous UOR electrocatalysts are analyzed. The UOR mechanism is discussed, and the design of amorphous catalysts is then analyzed. The main catalyst design strategies are illustrated, including nanostructure control,heteroatom doping, composition regulation, and heterostructure construction. Also, electrocatalysts' structureperformance correlation is interpreted. Perspectives in this field are proposed for guiding future studies on the development of high-performance amorphous catalysts towards energy sustainability.

Characterization and spatial analysis of coseismic landslides triggered by the Luding Ms 6.8 earthquake in the Xianshuihe fault zone, Southwest China

On September 5, 2022, a magnitude Ms 6.8 earthquake occurred along the Moxi fault in the southern part of the Xianshuihe fault zone located in the southeastern margin of the Tibetan Plateau,resulting in severe damage and substantial economic loss. In this study, we established a coseismic landslide database triggered by Luding Ms 6.8 earthquake, which includes 4794 landslides with a total area of 46.79 km^(2). The coseismic landslides primarily consisted of medium and small-sized landslides, characterized by shallow surface sliding. Some exhibited characteristics of high-position initiation resulted in the obstruction or partial obstruction of rivers, leading to the formation of dammed lakes. Our research found that the coseismic landslides were predominantly observed on slopes ranging from 30° to 50°, occurring at between 1000 m and 2500 m, with slope aspects varying from 90° to 180°. Landslides were also highly developed in granitic bodies that had experienced structural fracturing and strong-tomoderate weathering. Coseismic landslides concentrated within a 6 km range on both sides of the Xianshuihe and Daduhe fault zones. The area and number of coseismic landslides exhibited a negative correlation with the distance to fault lines, road networks, and river systems, as they were influenced by fault activity, road excavation, and river erosion. The coseismic landslides were mainly distributed in the southeastern region of the epicenter, exhibiting relatively concentrated patterns within the IX-degree zones such as Moxi Town, Wandong River basin, Detuo Town to Wanggangping Township. Our research findings provide important data on the coseismic landslides triggered by the Luding Ms 6.8 earthquake and reveal the spatial distribution patterns of these landslides. These findings can serve as important references for risk mitigation, reconstruction planning, and regional earthquake disaster research in the earthquake-affected area.

Label-free quantitative proteomics analysis models in vivo and in vitro reveal key proteins and potential roles in sciatic nerve injury

Background:The underlying mechanism of sciatic nerve injury(Sni)is a common motor functional disorder,necessitates further research.Methods:A rat model of Sni was established,with the injury group subjected to compressive injury of the right sciatic nerve exposed at the midpoint of the thigh and the sham surgery group undergoing the same surgical procedure.An oxygen-glucose deprivation model was employed to simulate in vitro Sni in PC12 cells.Following data acquisition and quality control,differentially expressed proteins(DEPs)in each model were identified through differential analysis,and enrichment analysis was used to explore the potential functions and pathways of the DEPs.Venn diagrams were drawn,and DEPs from both in vivo and in vitro Sni models were imported into the STRING database to construct a protein-protein interaction network and screen for hub proteins.Results:After the peptide segments obtained from rat nerve blockade and PC12 cells met quality requirements,258 DEPs were identified in rat nerve samples,and 119 DEPs were screened in PC12 cells.Enrichment analysis revealed that DEPs in the rat model were predominantly concentrated in biological functions such as myogenic cell proliferation and signaling related to lipid and energy metabolism.DEPs in the in vitro model were mainly enriched in biological processes such as phagocytosis and were associated with lipid transport and metabolism.Two hub proteins,amyloid precursor protein(APP)and fibronectin 1(FN1),were identified through MCC,MCODE,and Degree scoring.Both PC12 cells and external validation sets showed relatively higher expression of APP and FN1 in injured samples.Results of gene set enrichment analysis indicated that these two proteins were associated with metabolic pathways,such as biosynthesis of glycosaminoglycan chondroitin sulfate and biosynthesis of unsaturated fatty acids.Conclusion:APP and FN1 are potential key molecules involved in Sni and are associated with various metabolic pathways in nerve repair.These findings provide a theore

Even-odd layer-dependent multiferroic in freestanding rare-earth orthorhombic perovskite

Freestanding oxide perovskites possess strong interlayer coupling between adjacent atomic layers,thus exerting a determinative effect on the magnetism and ferroelectricity of these atomic-scale materials.Here,we propose an effective strategy to manipulate magnetism and ferroelectricity in freestanding rare-earth orthorhombic perovskite via modulation of layer thickness.By performing first-principles calculations,an even-odd oscillation is demonstrated in few-layer GdAlO_(3)perovskite(GAP).Specifically,odd-layer systems with charged atomic layers are ferromagnetic polar metals,while even-layer systems are antiferromagnetic ferroelectric semiconductors.This thickness-dependent magnetic phase transition originates from carrier doping,as rationalized by the Stoner criterion.Furthermore,we demonstrate the promotion of in-plane ferroelectricity via the concurrent application of two distinct antiferrodistortive displacements,each driven by formation and breaking of bonds.Analogous multiferroic phases may emerge in other transition metal oxide perovskites supporting multiple valence states,e.g.,few-layer Gd MO_(3)(M=V,Cr,Mn,and ni).This work puts forward a strategy for layer thickness engineering of magnetism and ferroelectricity in 2D oxide perovskite multiferroic materials.

Development and validation of a circulating tumor DNA-based optimization-prediction model for short-term postoperative recurrence of endometrial cancer

BACKGROUND Endometrial cancer(EC)is a common gynecological malignancy that typically requires prompt surgical intervention;however,the advantage of surgical management is limited by the high postoperative recurrence rates and adverse outcomes.Previous studies have highlighted the prognostic potential of circulating tumor DNA(ctDNA)monitoring for minimal residual disease in patients with EC.AIM To develop and validate an optimized ctDNA-based model for predicting shortterm postoperative EC recurrence.METHODS We retrospectively analyzed 294 EC patients treated surgically from 2015-2019 to devise a short-term recurrence prediction model,which was validated on 143 EC patients operated between 2020 and 2021.Prognostic factors were identified using univariate Cox,Lasso,and multivariate Cox regressions.A nomogram was created to predict the 1,1.5,and 2-year recurrence-free survival(RFS).Model performance was assessed via receiver operating characteristic(ROC),calibration,and decision curve analyses(DCA),leading to a recurrence risk stratification system.RESULTS Based on the regression analysis and the nomogram created,patients with postoperative ctDNA-negativity,postoperative carcinoembryonic antigen 125(CA125)levels ofnical utility.Furthermore,using the nomogram,the patients were successfully classified into three risk subgroups.CONCLUSION The nomogram accurately predicted RFS after EC surgery at 1,1.5,and 2 years.This model will help clinicians personalize treatments,stratify risks,and enhance clinical outcomes for patients with EC.

BACKGROUND Thrombocytopenia 2,an autosomal dominant inherited disease characterized by moderate thrombocytopenia,predisposition to myeloid malignancies and normal platelet size and function,can be caused by 5’-untranslated region(UTR)point mutations in ankyrin repeat domain containing 26(ANKRD26).Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1)have been identified as negative regulators of ANKRD26.However,the positive regulators of ANKRD26 are still unknown.AIM To prove the positive regulatory effect of GATA binding protein 2(GATA2)on ANKRD26 transcription.METHODS Human induced pluripotent stem cells derived from bone marrow(hiPSC-BM)INTRODUCTION Ankyrin repeat domain containing protein 26(ANKRD26)acts as a regulator of adipogenesis and is involved in the regulation of feeding behavior[1-3].The ANKRD26 gene is located on chromosome 10 and shares regions of homology with the primate-specific gene family POTE.According to the Human Protein Atlas database,the ANKRD26 protein is localized to the Golgi apparatus and vesicles,and its expression can be detected in nearly all human tissues[4].Moreover,UniProt annotation revealed that ANKRD26 is localized in the centrosome and contains coiled-coil domains formed by spectrin helices and ankyrin repeats[5,6].The most common disease related to ANKRD26 is thrombocytopenia 2(THC2),which is a rare autosomal dominant inherited disease characterized by lifelong mild-to-moderate thrombocytopenia and mild bleeding[7-9].Caused by the variants in the 5’-untranslated region(UTR)of ANKRD26,THC2 is defined by a decrease in the number of platelets in circulating blood and results in increased bleeding and decreased clotting ability[8,10].Due to the point mutations that occur in the 5’-UTR of ANKRD26,its negative transcription factors(TFs),Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1),lose their repression effect[11].The persistent expression of ANKRD26 increases the activity of the mitogen activated protein kinase and e

基于第一性原理计算方法,在考虑磁效应的基础上,对不同温度下合金Co_(2)(HfTa)中的稳定结构、电子性质和热力学性质展开了研究.通过第一性原理计算形成能、弹性性质及声子色散关系,结果表明合金Co_(2)(Hf_(x)Ta_(1-x))在x≤0.75时基态是C36,当x>0.75时基态是C14.这一发现提示了合金成分对其基态结构的影响,并为合金的稳定性提供了重要线索.值得注意的是,考虑了振动熵和电子熵的贡献后,Co_(2)Hf相对稳定的结构变为C36,Co_(2)Ta变为C14,这表明温度会对合金结构稳定性的影响.Co_(2)(Hf_(x)Ta_(1-x))合金在弹性方面呈现出本征脆性.热力学分析表明其在高温下热容趋于稳定,德拜温度近似为常数,振动熵随温度递增且与压强负相关.电子态密度分析揭示了Co_(2)(HfTa)合金三个结构的相稳定性.
