Signal Transduction Pathways Mediated by Secreted and Non-Secreted Forms of Intact Insulin-Like Growth Factor Binding Protein-3 (IGFBP-3) and Its 1-97 N-Terminal Fragment in PC-3 Human Prostate Cancer Cells

Our previous results indicated that both the secreted and the intracellular form of full length and 1-97 N-terminal fragment of IGFBP-3 induce apoptosis in PC-3 human prostate cancer cells in an IGF-dependent and independent manner. This study was undertaken to delineate possible down-stream signaling pathways that are involved in this process. Intact IGFBP-3 and its N-terminal 1-97 fragments with or without a signal propeptide were fused to YFP and expressed in PC-3 human prostate cancer cells. In some cases, the putative IGF-binding site was presented in full length IGFBP-3 and its N-terminal fragment was also mutated. Extent of apoptosis was quantified using FACS. Up-regulation of total Stat-1 and activation of phospho-Stat-1 were shown by western blot. TGF-β signal was measured by luciferase reporter assay. Results from inhibitor studies indicated that both the Caspase 8 and caspase 9 pathways are involved in IGFBP-3 (non-secreted form) which induced apoptosis in PC-3 cells. Exogenous addition of IGFBP-3 to PC-3 cells increased Stat-1 protein expression/tyrosine phosphorylation. Interestingly, results also showed that knockdown of Stat-1 by siRNA potentiated the IGFBP-3 induced apoptosis in PC-3 cells. In addition, both full-length IGFBP-3 and its 1-97 Nterminal fragments inhibited TGF-β signaling in these cells. This is the first report that compares the signal transduction pathways involved in apoptotic pathways mediated by IGFBP-3 in PC-3 human prostate cancer cells. Non-secreted form of full length IGFBP-3 and its N-terminal fragments induced apoptosis in PC-3 cells via activation of caspase 8 and caspase 9. Although, only non-secreted form of IGFBP-3 is involved in inducing apoptosis in PC-3 cells via caspase 8 and caspase 9 activation pathways but both secreted and non-secreted forms of IGFBP-3 are involved in modulating Stat-1 and TGF-β pathways to induce apoptotic actions in PC-3 cells. Background: Coronavirus disease 2019(COVID-19)is a new viral disease that has caused a pandemic in the *** to the lack of vaccines and definitive treatment,preventive behaviors are the only way to overcome the ***,the present study aimed to determine the preventive behaviors from the disease based on constructs of the health belief ***:: In the present cross-sectional study during March 11–16,2020,750 individuals in Golestan Province of Iran were included in the study using the convenience sampling and they completed the questionnaires through *** scores were calculated using the confirmatory factor *** effects of different factors were separately investigated using the univariate analyses,including students sample t-test,ANOVA,and simple linear ***,the effective factors were examined by the multiple regression analysis at a significant level of 0.05 and through Mplus 7 and SPSS ***: The participants’mean age was 33.9±9.45 years;and 57.1%of them had associate and bachelor's *** regression indicated that the mean score of preventive behavior from COVID-19 was higher in females than males,and greater in urban dwellers than rural ***,one unit increase in the standard deviation of factor scores of self-efficacy and perceived benefits increased the scores of preventive behavior from COVID-19 by 0.22 and 0.17 units *** the contrary,one unit increase in the standard deviation of factor score of perceived barriers and fatalistic beliefs decreased the scores of the preventive behavior from COVID-19 by 0.36 and 0.19 units ***: Results: of the present study indicated that female gender,perceived barriers,perceived self-efficacy,fatalistic beliefs,perceived interests,and living in city had the greatest preventive behaviors from COVID-19 *** interventions were necessary among males and villagers.

Simulation of Low TDS and Biological Units of Fajr Industrial Wastewater Treatment Plant Using Artificial Neural Network and Principal Component Analysis Hybrid Method

Being familiar with characteristics of industrial town effluents from various wastewater treatment units, which have high qualitative and quantitative variations and more uncertainties compared to urban wastewaters, plays very effective role in governing them. With regard to environmental issues, proper operation of wastewater treatment plants is of par- ticular importance that in the case of inappropriate utilization, they will cause serious problems. Processes that exist in environmental systems mostly have two major characteristics: they are dependent on many variables;and there are complex relationships between its components which make them very difficult to analyze. In order to achieve a better and efficient control over the operation of an industrial wastewater treatment plant (WWTP), powerful mathematical tool can be used that is based on recorded data from some basic parameters of wastewater during a period of treatment plant operation. In this study, the treatment plant was divided into two main subsystems including: Low TDS (Total Dissolved Solids) treatment unit and Biological unit (extended aeration). The multilayer perceptron feed forward neural network with a hidden layer and stop training method was used to predict quality parameters of the industrial effluent. Data of this study are related to the Fajr Industrial Wastewater Treatment Plant, located in Mahshahr—Iran that qualita- tive and quantitative characteristics of its units were used for training, calibration and validation of the neural model. Also, Principal Component Analysis (PCA) technique was applied to improve performance of generated models of neural networks. The results of L-TDS unit showed good accuracy of the models in estimating qualitative profile of wastewater but results of biological unit did not have sufficient accuracy to being used. Use of ultrasonic transient elastography (Fibroscan) in the assessment of hepatic focal lesion stiffness

Background and Aim: Hepatic focal lesions differ in their tissue composition and in the degree of stiffness, so our aim was to evaluate the role of Ultrasonic Transient Elastography (Fibroscan) in the measurement of hepatic focal lesions stiffness in order to differentiate hepatocellular carcinoma (HCC) focal lesions from other non HCC focal lesions. Methods: The study was conducted on 34 patients with hepatic focal lesion(s) in the right lobe located near the liver surface and more than or equal4 cmin diameter, detected by imaging studies and diagnosed by CT and histopathology. Stiffness over the focal lesions was measured by the fibroscan. Results: The median value of stiffness was 72.5 kPa over HCC focal lesions, 17.2 kPa over lymphoma focal lesions, 6.5 kPa over metastatic focal lesions and 10.5 kPa over the sarcoidosis focal lesion. Conclusions: Hepatocellular carcinoma focal lesions are much stiffer than lymphoma, metastasis or sarcoidosis focal lesions. Fibroscan may be a useful non-invasive method in the prediction of hepatocellular carcinoma in the future.

Preliminary Study on the Distribution of Irrawaddy Dolphin, <i>Orcaella brevirostris</i>, in Banten Bay

Irrawaddy dolphins (Orcaella brevirostris) are aquatic mammals with critically endangered status able to live in different water salinities habitats, like estuary, rivers and lakes. Banten Bay features estuarine area where rapid economic development, contributed from industry, ports, mining and fisheries, takes place, and thus the presence of Irrawaddy dolphins in the area faces various environmental threats. This research aimed to study the distribution of Irrawaddy dolphins in Banten Bay and explore environmental factors threatening its life. Dolphins observation was conducted from January 2013-February 2014, using direct visual effort by team of observer and indirect effort by local fishermen. Direct visual effort was performed on boat by three observers, with maximum vessel speed 15 km/hour and total length of transect was 404 Km. Indirect visual effort comprised of regular assessment by 32 local fishermen when they conducted capture fishing at two different areas (South and North of Banten Bay). Sightings data from direct and indirect visual observation compiled with respondents opinion were mapped using Arc View®?GIS 3.3, resulting in distribution map of Irrawaddy dolphin in Banten Bay. Irrawaddy dolphin was most frequently observed in the northeast, south and central areas of the bay. Environmental threats alarming the lives of Irrawaddy dolphin in the Banten Bay may include noise pollution, boat accident and entanglement.

Cloud computing is a kind of computing that depends on shared figuring assets instead of having nearby servers or individual gadgets to deal with applications. Technology is moving to the cloud more and more. It’s not just a trend, the shift away from ancient package models to package as service has steadily gained momentum over the last ten years. Looking forward, the following decade of cloud computing guarantees significantly more approaches to work from anyplace, utilizing cell phones. Cloud computing focused on better performances, better scalability and resource consumption but it also has some security issue with the data stored in it. The proposed algorithm intents to come with some solutions that will reduce the security threats and ensure far better security to the data stored in cloud.

A Cross-Sectional Study on Assessment of Oxidative Stress in Coronary Heart Disease Patients in Bangladesh

Oxidative stress is considered a central factor in endothelial dysfunction and plaque formation in coronary heart disease (CHD) patients. In this cross-?sectional study, the stress-induced protein and lipid oxidation and production of signaling molecule, nitric oxide has been investigated in CHD patients in Bangladesh. A total of 140 participants were enrolled of which 70 were CHD patients, and the remaining 70 were control subjects having no cardiovascular disease. The diagnosis of CHD was made by expert physicians through characteristic electrocardiogram and troponin changes. The protein carbonyls and thiobarbituric acid reactive substances (TBARS) levels in plasma of both groups were measured by spectrophotometric methods. The plasma nitric oxide levels were measured by reaction with Griess reagent. Statistical non-parametric tests and t-tests were used to analyze the results between the CHD patients and controls. In the patients, the mean protein carbonyls and TBARS, two important markers of oxidative stress, were 2.87 ± 0.97 nmol/mg and 6.92 ± 3.56 nmol/mL, respectively, which were significantly higher than in the control subjects, 1.71 ± 0.82 nmol/mg and 3.15 ± 1.42 nmol/mL, respectively. Assessment of the cardiovascular signaling molecule nitric oxide showed significantly lower value in the CHD patients (6.29 ± 4.78 μM) compared to the controls (10.75 ± 6.03 μM). There was a significant negative correlation between plasma TBARS and total plasma protein, and a weak negative correlation between TBARS and nitric oxide levels in the CHD patients. Evaluation of the smoking habit strongly suggested that smoking was a risk factor for the development of CHD among the Bangladeshi population. These results provide an overview of oxidative?stress-mediated damages on proteins, lipids and effects on signaling molecule nitric oxide in CHD patients.

Mineral Chemistry and Geothermometry of Amphibole and Plagioclase in the Metabasites, Located at the Tanbour Metamorphic Complex in Southern Iran

Metabasite refers to metamorphosed basalts and other mafic igneous rocks (rich in iron and magnesium). When a mafic igneous rock is subjected to new pressure and temperature conditions during metamorphism, these chemical components will rearrange themselves to form new minerals. Metabasites can be found in many metamorphic belts including Sanandaj-Sirjan metamorphic belt of Iran. The study area is a Tanbour metamorphic complex in Eastern of Sirjan city, which is geologically located at the Sanandaj-Sirjan metamorphic belt in Southern Iran. Metabasite in this complex consists of greenschist, epidote amphibolite and amphibolite. Amphibole and plagioclase are the main minerals in the greenschist and amphibolite, and the a secondary mineral in some micaschist seen in the study area. The electron microprobe analysis was done on this mineralization in greenschist, epidote amphibolite and amphibolite, which showed that the amphiboles in greenschist was a member of the calcic group and Actinolite type, and the amphiboles in epidote amphibolite was a member of the calcic group and these amphiboles were tschermakite up to Ferro-Tschermakite + Ferro-Hornblende type. The amphibole in amphibolite is a member of the calcic group and this amphibole is Magnesio-Hornblende type. The plagioclases in the greenschist is pure albite (An 3.29 - 3.6), and in the epidote amphibolite is oligoclase (An 19.5 - 24.2), while in the amphibolites is oligoclase (An 16.9 - 26.6). The estimated P–T conditions are in favor of their metamorphism under epidote amphibolite (550°C and 8 kbar) and amphibolite (611°C - 652° Cand 10.5 kbar) facies.
