Risk Assessment Capacity Building Program in Zaporizhzhia Ukraine: Emissions Inventory Construction, Ambient Modeling, and Hazard Results

Historically, Ukraine has been a major source of industrial production for the former Soviet Union and the source of pollution associated with an aging industrial infrastructure. The US Environmental Protection Agency (US EPA) and the Ukrainian Ministry of Environment and Natural Resources (MENR) entered into partnership to develop Ukrainian expertise and capacity in risk assessment so that Ukraine could more effectively use its National and Regional Environmental Protection Funds and set priorities for cleanup and regulation. Ukrainian scientists, local officials, and EPA consultants conducted a pilot study in the heavily industrialized Zaporizhzhia Oblast so that the process, analytical tools, and approach for a risk assessment could be developed for and tailored to Ukrainian needs. As a first step, site-specific information was obtained from multiple sources of air pollution and an emissions inventory of air pollution developed. Efforts by local officials were critical for emissions inventory construction. After refinements were made to the inventory, Ukrainian scientists then performed exposure modeling using this information so that ambient concentrations of pollutants could be estimated. 11 industry types (i.e., enterprises) were identified as a major emission source. Results of the modeling effort demonstrated that emissions estimates of particulate matter (as measured by particles of less than 10 micron diameter or “PM10”) and a number of carcinogens were consistent with those from other cities with high concentrations of metallurgical industries in former Soviet Union countries, and were above safety standards. Hazard information was gathered from international databases for each of the estimated pollutants. In a recent study published in Cancer Cell,Bagaev et *** a pan-cancer tumor microenvironment(TME)classification that can be predictive of response to immune checkpoint inhibitor(ICI)therapy.1 The authors presented a detailed visualization that can be generated for each tumor,which combines the 29 functional gene expression signatures(Fges)used to subtype the TME with targetable genomic *** the compartments and pathways within the TME could aid decision-making in patient-specific treatment of cancer,especially in the context of immunotherapies.2 The increasing use of ICIs have shown promising clinical outcomes in several cancer ***,response to treatment is heterogenous and often results in immune-related adverse events.3 Therefore,efforts are focused on the development of prognostic tools to predict response to *** analysis of 468 melanoma patients resulted in grouping of the TME into four major subsets based on manually curated Fges.

The Tissue Reactions and Changes of a Surface of Various Metal Implants after Their Introduction in a Bone Tissue in Experiment

Screw metal implants (3S, Israel) with rough or smooth polished surface were introduced in a tibial proximal condyle of not purebred rabbits. The condition of surrounding tissues in 2 and 6 months after implantation was compared by light microscopy and X-ray methods. Within 6 months after operation the considerable distinctions of radiological and morphological data were revealed not. 2 months later after introduction of implants with a rough surface the effort enclosed for its twisting is, much more, than for removal of the polished product. However, stability of fixing of implants was practically made even at 6 months. On remote rough implants there is a set of tissue scraps whereas on products with a smooth surface the tissue remains were much less. Surrounding tissues strongly join a rough surface, grow into cavities, and during removal of such products there is a considerable trauma of tissues round an implantation place. Smooth implants have the smaller area of contact with organism tissues, they are fixed due to bicortical implantation, during removal easily get out and don’t break off surrounding tissues. The signs of inflammation and formation of merged multinuclear macrophages were not found at all cases, which give evidence to the inertness of material of the mentioned articles for living organism. In some observations however and by implantation of the rough article and by introduction of polished implants, metal particles were found, but after use of the foreign body with grit-blasted treatment of surface metal was found more frequently, and its fragments had larger volume.

Novel Simulation Workshop Improves Performance in Vacuum Delivery

Background: Operative vaginal delivery is a common obstetric intervention, with the potential to cause harm to mother and baby. Training in operative delivery traditionally comprised junior trainees learning the skills under supervision, then practicing and refining them independently. Recently this model has come under scrutiny, with the advent of simulation-based training suggesting a method by which the skills of safe delivery may be taught without risk. Multiple training courses exist, but few have been subjected to evaluation. Aims: The primary aim was to investigate if a simulation-based training workshop, where the development of a successful technique for vacuum delivery occurs using a collaborative, problem-solving approach, improves the rate of correct vacuum cup placement. The secondary aim was to determine if the workshop leads to improvement in theoretical knowledge of vacuum delivery. Methods: Participants on three workshops were assessed for their performance in vacuum delivery prior to and following a multimodal training program. Participants included general practitioner obstetricians, obstetric trainees and resident medical officers. Evaluation occurred using a standardised Likert-scaled rating sheet and utilising a diagrammatic representation of cup placement. Results: The participants demonstrated significant improvement (median post-pre score 1, p in the accuracy of cup placement, in a variety of practical core skills and in theoretical knowledge of vacuum delivery. Discussion: Participation in a simulation-based vacuum delivery workshop improves the performance of obstetric trainees and GP obstetricians. Further work is required to evaluate the performance of such training modalities on clinically relevant outcomes.

H-FABP in Pre Capillary Pulmonary Hypertension: Comparison to Other Surrogate Parameters for Prediction of Severity and Outcome

Background: Heart-type fatty acid-binding protein (H-FABP) is a promising novel biomarker for risk stratification of patients with chronic thromboembolic pulmonary hypertension (CTEPH). Whether disease severity in a group of patients with pre capillary pulmonary hypertension (PH) can be predicted by determination of plasma H-FABP levels remains unknown. Methods: 41 consecutive patients with a mean (±SD) age of 60.6 ± 2.1 years and severe PH were studied with a mean follow up to 541 days (95% CI: 420;661 days). H-FABP, but also the biomarkers NT-Pro-BNP and big-endothelin (Big-ET1), were correlated to parameters derived from right sided cardiac catheterization and cardio-pulmonary exercise test. Results: At baseline H-FABP levels ranged from 620 to 15200 pg/ml (3648 ± 502 pg/ml) and were weakly but significantly correlated to VO2AT (r = –0.37, p = 0.04) and VO2 peak (r =–0.38, p = 0.03). However, invasively measured hemodynamic parameters of PH and right ventricular dysfunction did not correlate with H-FABP levels. In- terestingly, moderate to high correlations between H-FABP and NT-Pro-BNP (r = 0.51, p = 0.01) and Big-ET1 (r = 0.65, p = 0.01) and between Big-ET1 and NT-Pro-BNP (r = 0.5, p = 0.01) could be observed. In contrast to H-FABP, NT-Pro-BNP, and even more Big-ET1, showed significant correlations to different invasively measured hemodynamic parameters of the disease indicating severity and consequently prognosis of severe pre-capillary pulmonary hypertension. However, in a multivariate Cox regression analysis, PVR, mixed venous oxygen saturation and the VE/VCO2 slope (>60) as well as the heart rate recovery within one minute (HRR) but none of the biomarkers were identified as independent factors of poor prognosis. In con- trast, peak workload and mixed venous saturation were identified as risk markers in the idiopathic pulmonary arterial hypertension subgroup. From Droplets and Particles to Hierarchical Spatial Organization： Nanotechnology Challenges for Microfluidics

The compartimentation of fluids in the microliter, nanoliter and picoliter range leads recently to many applications of microfluidics in material development, diagnostics and biological screenings. Droplet-based microfluidics allows the improvement of nanoparticle homogeneity and the tuning of particle properties. It supports combinatorial synthesis of inorganic as well as organic substances and can be applied for the cultivation and screening of bacteria, eucaryotic cells and fish embryos. The well-ordered handling and the addressing of microfluid segments improves the information transfer between chemical, biological and electronic systems. Despite this remarkable technical progress, there is a particular importance of microfluidics for future nanotechnological solutions. The hierarchical spatial organization of liquids, particles and gels in microfluidics represents a fundamental biomimetic principle which overcomes the limits of planar technology and opens the gate for realizing complex structured threedimensional nanoarchitectures. Recent applications of microstructured fluids in chemistry and biology and concepts for future developments will be discussed.

Vitellogenin expression in the oilseed rape pest Meligethes aeneus

When investigating insecticide resistance of pest insects, for example, the pollen beetle Meligethes aeneus, it is relevant to differentiate toxicological and molecular genetic data between male and female specimens. A molecular sex determination method would allow resistance testing to be run without prior sorting of the samples. A one-step quantitative RT-PCR method for quantification of the yolk protein vitellogenin expression in the pollen beetle was established. The expression level of vitellogenin relative to tubulin was determined. Pollen beetles were tested at different time points during their development to determine if vitellogenin is a reliable molecular marker for detection of sexually mature females. The differentiation between females and males by relative expression of vitellogenin to tubulin is conditional regarding the life cycle. Sexually mature females and males could easily be distinguished, whereas immature specimens could not be seperated. Vitellogenin expression is a successful marker for identification of sexually mature pollen beetles. Females from the spring populations showed vitellogenin expression when the temperature was above 10.2°C. Further, detailed observations of vitellogenin throughout the spring indicated a strong relationship between daily temperatures and vitellogenin expression, which is an indicator of oviposition ability.
