Plastins regulate ectoplasmic specialization via its actin bundling activity on microfilaments in the rat testis

In this paper, technology computer aided design （TCAD） was applied to optimize the fabrication process to reduce the parasitic capacitance of PMOS transistor at the source/drian （S/D） junction （Cj） in developing the 65nm low leakage （65nmLL） technology. It was found that Cj can be effectively reduced by combining relative high-energy well implant and proper threshold voltage （Vt） implant method. Through measured data and TCAD simulation, this paper also demonstrates a "doping compensation effect" by tuning Vt and Halo implants with proper species, energy, and dosage, to achieve the Cj reduction.

Plastins are a family of actin binding proteins （ABPs） known to cross-link actin microfilaments in mammalian cells, creating actin microfilament bundles necessary to confer cell polarity and cell shape. Plastins also support cell movement in response to changes in environment, involved in cell/tissue growth and development. They also confer plasticity to cells and tissues in response to infection or other pathological conditions （e.g., inflammation）. In the testis, the cell-cell anchoring junction unique to the testis that is found at the Sertoli cell-cell interface at the blood-testis barrier （BTB） and at the Sertoli-spermatid （e.g., 8-19 spermatids in the rat testis） is the basal and the apical ectoplasmic specialization （ES）, respectively. The ES is an F-actin-rich anchoring junction constituted most notably by actin microfilament bundles. A recent report using RNAi that specifically knocks down plastin 3 has yielded some insightful information regarding the mechanism by which plastin 3 regulates the status of actin microfilament bundles at the ES via its intrinsic actin filament bundling activity. Herein, we provide a brief review on the role of plastins in the testis in light of this report, which together with recent findings in the field, we propose a likely model by which plastins regulate ES function during the epithelial cycle of sDermatogenesis via their intrinsic activity on actin microfilament organization in the rat testis.

The aging population is an important issue around the world especially in developed *** med-ical advances have substantially extended life span,the same cannot be said for the duration of health *** are seeing in-creasing numbers of elderly people who are frail and/or have multiple chronic conditions;all of these can affect the quality of life of the elderly population as well as increase the burden on the healthcare *** is mechanistically related to common medical conditions such as diabetes mellitus,ischemic heart disease,cognitive decline,and frailty.A recently accepted concept termed‘Accelerated Biological Aging’can be diagnosed when a person’s biological age—as measured by biomarkers of DNA methylation—is older than their corresponding chronological ***,a conditionally essential amino acid,has received much attention in the past few years.A substantial number of animal studies have provided a strong scientific foundation sug-gesting that this amino acid can improve cellular and metabolic health,including blood glucose control,so much that it has been labelled one of the‘longevity amino acids’.In this review article,we propose the rationale that an adequately powered random-ized-controlled-trial(RCT)is needed to confirm whether taurine can meaningfully improve metabolic and microbiome health,and biological *** trial should incorporate certain elements in order to provide the much-needed evidence to guide doctors,and also the community at large,to determine whether this promising and inexpensive amino acid is useful in improving human metabolic health.

Thoracic surgery is known to cause severe pain that may not subside during the course of hospitalization. Early pain control is important in these patients because it improves ventilation and promotes mobility. Lung transplant recipients can experience post-operative complications that lead to extended hospital stays. This increases the time a patient is sedentary, which further deconditions patients;therefore, rehabilitation should be initiated early post-operatively. We present a unique case of a critically ill double lung transplant recipient whose rehabilitation post-operatively was hindered by severe vasopressor-induced ischemic pain. Due to debilitating pain, he was unable to regain mobility. His pain management course was challenging due to sensitivity to opioids, renal failure, and anticoagulation.

Perpendicular magnetic clusters with configurable domain structures via dipole-dipole interactions

Magnetic single-domain islands based on in-plane anisotropy （usually, shape anisotropy） and their dipole-dipole interactions have been investigated extensively in recent years. This has been driven by potential applications in magnetic recording, spintronics, magneto-biology, etc. Here, we propose a concept of out- of-plane magnetic dusters with configurable domain structures （multi-flux states） via dipole-dipole interactions. Their flux stages can be switched through an external magnetic field. The concept has been successfully demonstrated by patterned [Co/Pd] islands. A [Co/Pd] multilayer exhibits a large perpendicular anisotropy, a strong physical separation, and uniform intrinsic properties after being patterned into individual islands by electron beam lithography. A three- island cluster with six stable flux states has been realized by optimizing island size, thickness, gap, anisotropy, saturation magnetization, etc. Using [Co/Pd] multilayers, we have optimized the island structure by tuning magnetic properties （saturation magnetization and perpendicular anisotropy） using Landau-Liftshitz- Gilbert （LLG） simulation/calculation. Potential applications have been proposed, including a flexi-programmable logic device with AND, OR, NAND, and NOR functionalities and a magnetic domino, which can propagate magnetic current as far as 1 μm down from the surface via vertical dipole-dipole interactions.

探讨原发性高血压患者的遗传性,评估线粒体D环控制区基因变异在高血压发病中的作用。提取原发性高血压患者和正常血压人群DNA各20例,用3对交叉重叠引物扩增全部线粒体控制区D环基因,直接基因测序并重建邻接关系树,分析原发性高血压的基因变异特点。结果发现部分高血压病患者具有明显群聚倾向,与正常血压人群和其他无群聚倾向的高血压患者比较,存在高频率、高密度的D环控制区基因变化(PC、np182C >T、np189A >G、np247G >A、np16187C >T、np16189T >C、np16264C >T、np16270C >T和np16311T >C等多态性变化显著,并因此造成np16184～16193微卫星区域多聚C长度改变。研究提示部分高血压病患者有群聚现象,基因型np152C、np182T、np247A、np16187T、np16189C、np16264T、np16270T和np16311C可能是此聚类族高血压患者的易感遗传标记。

Effect of music on procedure time and sedation during colonoscopy:A meta-analysis

AIM:To integrate results from different studies in examining the effectiveness of music in reducing the procedure time and the amount of sedation used during colonoscopic procedure. METHODS: An electronic search in various databases was performed to identify related articles. Study quality was evaluated by the Jadad’s scale. The random effect model was used to pool the effect from individual trials and the Cohen Q-statistic was used to determine heterogeneity. Egger’s regression was used to detect publication bias. RESULTS: Eight studies with 722 subjects were included in this meta-analysis. The combined mean difference for the time taken for the colonoscopy procedure between the music and control groups was -2.84 with 95% CI (-5.61 to -0.08), implying a short time for the music group. The combined mean difference for the use of sedation was -0.46 with 95%CI (-0.91 to -0.01), showing a significant reduction in the use of sedation in the music group. Heterogeneity was observed in both analyses but no publication bias was detected. CONCLUSION: Listening to music is effective in reducing procedure time and amount of sedation during colonoscopy and should be promoted.

AIM:To establish the roles of lipopolysaccharide (LPS)/CD14/toll-like receptor 4 (TLR4)-mediated inflammation in a rat model of human necrotizing enterocolitis (NEC).METHODS: Six pairs of intestinal samples from human NEC were collected before and after recovery for histological and molecular analysis of inflammatory cytokines and signaling components. In the rat NEC model, we isolated 10-cm jejunum segments and divided them into six groups (n=6) for sham operation, treatment with LPS, bowel distension, combined bowel distension and LPS stimulation, and two therapeutic groups. The potential eff icacy of a recombinant CD18 peptide and a monoclonal CD14 antibody was evaluated in the latter two groups. The serum and tissue levels of several inflammatory mediators were quantified by real-time polymerase chain reaction, ELISA and ***: Human acute phase NEC tissues displayed significant increases (P<0.05) in levels of TLR4, CD14, myeloid differentiation protein (MD)-2, tumor necrosis factor (TNF)-α and nuclear factor-κB when compared to those after recovery. The histological and inflammatory picture of human NEC was reproduced in rats that were treated with combined bowel distension and LPS, but not in the sham-operated and other control rats. Serum levels of interleukin-6 and TNF-α were also elevated. The NEC pathology was attenuated by treating the NEC rats with a monoclonal CD14 antibody or an LPS-neutralizing ***:LPS and distension are required to produce the histological and inflammatory features of NEC. A potential treatment option is blocking LPS activation and leukocyte infi ltration.
