Neotropical mustelids: fecal metabolome diversity and its potential for taxonomic discrimination

Chemical proﬁles of non-invasive biological material,such as feces,have great potential to study elusive animals or those with low population ***,we use a metabolomic approach to evaluate Neotropical mustelids as a biological model to describe the diversity of the metabolites present in fecal samples,as well as to evaluate the potential of chemical proﬁles for taxonomic *** collected fecal samples from captive individuals of 5 species of mustelids occurring in Brazil and analyzed them by liquid chromatography coupled to high-resolution mass *** 200 compounds have been annotated;“bile acids,alcohols and derivatives”was the most expressive class in the metabolome of all the *** successfully discriminated 3 taxonomic groups:1—tayra(Eira barbara);2—otters(Lontra longicaudis and Pteronura brasiliensis;1);and 3—grisons(Galictis vittata and Galictis cuja).Several compounds seemed to be associated with food intake and the digestive process,while others were found for theﬁrst time in Neotropical *** concluded that mustelids show high metabolome diversity and that species-speciﬁc identiﬁcation through metabolomic proﬁles is possible,thus contributing to the development and implementation of additional non-invasive approaches in the study of mustelids.

Autoimmune enteropathy(AIE)is a rare immune mediated disorder primarily affecting children,characterized by chronic diarrhea,malabsorption,vomiting,weight loss and villous *** has also been observed in adults presenting diagnostic and treatment challenges due to its overlap with other gastrointestinal disorders such as celiac *** diagnostic criteria for AIE include small bowel villous atrophy,lack of response to dietary restrictions,presence of anti-enterocyte antibodies,and predisposition to autoimmunity without severe *** criteria emphasize characteristic histological findings and exclusion of other causes of villous *** is associated with various autoimmune disorders and can present with overlapping features with Celiac disease,including villous atrophy but without significant intraepithelial *** primarily involves immunosuppression using corticosteroids,calcineurin inhibitors,and anti-tumor necrosis factor therapy,alongside nutri-tional *** the challenges,understanding AIE’s diverse manifest-ations and improving diagnostic criteria are essential for effective management and improved patient *** research is needed to elucidate the pathogenesis,disease progression and long-term outcomes of AIE.

A functional tacrolimus-releasing nerve wrap for enhancing nerve regeneration following surgical nerve repair

Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FDA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.

Noninvasive methods for prediction of esophageal varices in pediatric patients with portal hypertension

AIM: To evaluate clinical and laboratory parameters for prediction of bleeding from esophageal varices (EV) in children with portal hypertension. METHODS: Retrospective study of 103 children (mean age: 10.1 ± 7.7 years), 95.1% with intrahepatic portal hypertension. All patients had no history of bleeding and underwent esophagogastroduodenoscopy for EV screening. We recorded variceal size (F1, F2 and F3), red-color signs and portal gastropathy, according to the Japanese Research Society for Portal Hypertension classification. Patients were classified into two groups: with and without EV. Seven noninvasive markers were evaluated as potential predictors of EV: (1) platelet count; (2) spleen size z score, expressed as a standard deviation score relative to normal values for age; (3)platelet count to spleen size z score ratio; (4) platelets count to spleen size (cm) ratio; (5) the clinical prediction rule (CPR); (6) the aspartate aminotransferase to platelet ratio index (APRI); and (7) the risk score. RESULTS: Seventy-one children had EV on first endoscopy. On univariate analysis, spleen size, platelets, CPR, risk score, APRI, and platelet count to spleen size z score ratio showed significant associations. The best noninvasive predictors of EV were platelet count [area under the receiver operating characteristic curve (AUROC) 0.82; 95%CI: 0.73-0.91], platelet: spleen size z score (AUROC 0.78; 95%CI: 0.67-0.88), CPR (AUROC 0.77; 95%CI: 0.64-0.89), and risk score (AUROC 0.77; 95%CI: 0.66-0.88). A logistic regression model was applied with EV as the dependent variable and corrected by albumin, bilirubin and spleen size z score. Children with a CPR 114. A risk score > -1.2 increased the likelihood of EV (odds ratio 7.47; 95%CI: 2.06-26.99). In this work we axe concerned with the universal associative envelope of a finite-dimensional simple symplectic anti-Jordan triple system （AJTS）. We prove that if T is a triple system as above, then there exists an associative algebra U（T） and an injective homomorphism ε ： T→ U（T）, where U（T） is an AJTS under the triple product defined by （a, b, c） = abc- cba. Moreover, U（T） is a universal object with respect to such homomorphisms. We explicitly determine the PBW-basis of U（T）, the center Z（U（T）） and the Gelfand-Kirillov dimension of U（T）.

High-resolution Fourier spectroscopy as a tool for studying quality of rare-earth-doped crystals

Three examples were considered of the use of high resolution Fourier-transform optical spectroscopy for studying quality of rare-earth-doped crystals. The first example was connected with defects present in crystals grown by flux techniques. The second example dealt with detection of stresses and deformations in rare-earth-containing crystals, by registering splitting of spectral lines. The third example showed that a very small amount （at the level ofppm） of different RE ions present in a crystal could be determined using high-resolution spectroscopy. This work was carried out by the author＇s group in collaboration with several institutions in Rus- sia and abroad.

Myelinosome organelles in pathological retinas: ubiquitous presence and dual role in ocular proteostasis maintenance

The timely and efficient elimination of aberrant proteins and damaged organelles, formed in response to various genetic and environmental stressors, is a vital need for all cells of the body. Recent lines of evidence point out several non-classical strategies employed by ocular tissues to cope with aberrant constituents generated in the retina and in the retinal pigmented epithelium cells exposed to various stressors. Along with conventional strategies relying upon the intracellular degradation of aberrant constituents through ubiquitin-proteasome and/or lysosome-dependent autophagy proteolysis, two non-conventional mechanisms also contribute to proteostasis maintenance in ocular tissues. An exosome-mediated clearing and a myelinosome-driven secretion mechanism do not require intracellular degradation but provide the export of aberrant constituents and “waste proteins” outside of the cells. The current review is centered on the non-degradative myelinosome-driven secretion mechanism, which operates in the retina of transgenic Huntington’s disease R6/1 model mice. Myelinosome-driven secretion is supported by rare organelles myelinosomes that are detected not only in degenerative Huntington’s disease R6/1 retina but also in various pathological states of the retina and of the retinal pigmented epithelium. The intra-retinal traffic and inter-cellular exchange of myelinosomes was discussed in the context of a dual role of the myelinosome-driven secretion mechanism for proteostasis maintenance in different ocular compartments. Special focus was made on the interplay between degradative and non-degradative strategies in ocular pathophysiology, to delineate potential therapeutic approaches to counteract several vision diseases.

The birth hour of mammals:insights from intra-specific variation in wild blue monkeys

While most mammals show birth hour peaks at times of the 24-h cycle when they are less active,there are exceptions to this general *** exceptions have been little explored,but may clarify evolutionary reasons for the diel timing of *** investigated intraspecific variation in birth hour in wild blue monkeys Cercopithecus mitis stuhlmanni,a diurnal primate,to identify factors that differentiated daytime versus nighttime *** and life history data from 14 groups over 14 years revealed that 4%of 484 births occurred during the *** of daytime birth varied with mother’s age,peaking at 15.7 *** whose annual timing deviated most from the population’s peak birth months were 5 times more likely to occur during daytime than those that deviated *** was no evidence that mother’s rank or infant sex influenced birth hour,and mixed evidence that daytime births were more probable in larger *** did not differ significantly for infants born during the day versus ***-aged mothers may be able to handle the consequences of an unusual birth hour more successfully than mothers with less experience or those weakened by *** birth may be more advantageous in the off-season because nights are colder at that time of *** findings are consistent with hypotheses relating birth hour to the risk of losing social protection in group-living animals,but are not consistent with those emphasizing risk of conspecific *** of within-species variation can help in evaluating evolutionary hypotheses for non-random birth hour.
