Enhancement of endogenous midbrain neurogenesis by microneurotrophin BNN-20 after neural progenitor grafting in a mouse model of nigral degeneration

We have previously shown the neuroprotective and pro-neurogenic activity of microneurotrophin BNN-20 in the substantia nigra of the “weaver” mouse, a model of progressive nigrostriatal degeneration. Here, we extended our investigation in two clinically-relevant ways. First, we assessed the effects of BNN-20 on human induced pluripotent stem cell-derived neural progenitor cells and neurons derived from healthy and parkinsonian donors. Second, we assessed if BNN-20 can boost the outcome of mouse neural progenitor cell intranigral transplantations in weaver mice, at late stages of degeneration. We found that BNN-20 has limited direct effects on cultured human induced pluripotent stem cell-derived neural progenitor cells, marginally enhancing their differentiation towards neurons and partially reversing the pathological phenotype of dopaminergic neurons generated from parkinsonian donors. In agreement, we found no effects of BNN-20 on the mouse neural progenitor cells grafted in the substantia nigra of weaver mice. However, the graft strongly induced an endogenous neurogenic response throughout the midbrain, which was significantly enhanced by the administration of microneurotrophin BNN-20. Our results provide straightforward evidence of the existence of an endogenous midbrain neurogenic system that can be specifically strengthened by BNN-20. Interestingly, the lack of major similar activity on cultured human induced pluripotent stem cell-derived neural progenitors and their progeny reveals the in vivo specificity of the aforementioned pro-neurogenic effect.

Rescue of autoimmune hepatitis by soluble MHC class II molecules in an altered concanavalin A-induced experimental model

Background:Soluble major histocompatibility complex class II(sMHCII)molecules have been described to maintain tolerance through the suppression of autoreactive T lymphocytes.In order to evaluate their ability to rescue autoimmune hepatitis(AIH)symptoms,the present work attempted to administer sMHCII molecules to an in vitro as well as in vivo concanavalin A(ConA)-induced AIH model.Methods:The in vitro AIH model consisted of splenocyte stimulation with ConA in the presence or absence of serum-isolated sMHCII molecules.An in vivo ConAmodified model with or without sMHCII treatment was developed.The cytokine profile in culture supernatants and serum was tested by ELISA.Cell markers were evaluated by immunofluorescence,while cell proliferation by tritiated thymidine uptake.AIH symptoms were assessed by daily observations for the establishment of a disease severity scoring system and liver histology was evaluated using a biomolecular imager.Results:The presence of sMHCII molecules in the ConA-stimulated cell cultures leads to a significant reduction of cell proliferation.The administration of sMHCII molecules to the ConA-treated animals showed a significant reduction in the levels of IL-2,IL-4,and IL-10,as well as a decrease in the number of spleen CD4+and CD8+cells.Upon development of a scoring system,it was shown that the sMHCII treatment was accompanied by a slower progression of the disease,while rescuing fibrotic liver morphology.Conclusion:The results presented in this study confirm the ability of sMHCII proteins to alleviate autoimmune hepatitis,possibly highlighting new therapeutic approaches for autoimmune diseases.

The SeqCode,formally called the Code of Nomenclature of Prokaryotes Described from Sequence Data,is a new code of nomenclature in which genome sequences are the nomenclatural types for the names of prokaryotic species.While similar to the International Code of Nomenclature of Prokaryotes(ICNP)in structure and rules of priority,it does not require the deposition of type strains in international culture collections.Thus,it allows for the formation of permanent names for uncultured prokaryotes whose nearly complete genome sequences have been obtained directly from environmental DNA as well as other prokaryotes that cannot be deposited in culture collections.Because the diversity of uncultured prokaryotes greatly exceeds that of readily culturable prokaryotes,the SeqCode is the only code suitable for naming the majority of prokaryotic species.The start date of the SeqCode was January 1,2022,and the online Registry(https://seqco.del)was created to ensure valid publication of names.The SeqCode recognizes all names validly published under the ICNP before 2022.After that date,names validly published under the SeqCode compete with ICNP names for priority.As a result,species can have only one name,either from the SeqCode or ICNP,enabling effective communication and the creation of unified taxonomies of uncultured and cultured prokaryotes.The SeqCode is administered by the SeqCode Committee,which is comprised of the SeqCode Community and elected administrative components.Anyone with an interest in the systematics of prokaryotes is encouraged to join the SeqCode Community and participate in the development of this resource.

Growth responses of seedlings produced by parent seeds from specifi caltitudes

Restoration activities in semi-arid Mediterranean areas like Greece face many obstacles,such as summer droughts that are becoming more intense with climate change,that pose limitations to transplanting success.Seedlings for restoration must be of high quality;a vigorous root system is critical to enable seedlings to tolerate adverse conditions.Here we investigated the eff ects of altitude and source parent for seeds on the growth of subsequent seedlings to determine the best seed sources for obtaining highest-quality,most-tolerant seedlings for restoration eff orts.Seeds of Quercus coccifera L.were collected on an altitudinal gradient of 50 m(200,250 and 300 m a.s.l.)and from specifi c parents at each altitude.Subsequent seedlings were grown for 3 months in a greenhouse with controlled irrigation.The results indicated a strong altitudinal and parental seed eff ect on seedling characteristics.As altitude decreased,biomass of the seedlings increased,and they developed more vigorous roots and more photosynthetic leaf tissue.Thus,altitude and parent are critical factors to consider when producing seed-derived seedling.By collecting seeds from a specifi c altitude and parent,more vigorous and stresstolerant seedlings can be obtained to enhance transplanting success.

Developing an enhanced recovery after surgery program for oncology patients who undergo hip or knee reconstruction surgery

Enhanced recovery after surgery(ERAS)protocols are applied in orthopedic surgery and are intended to reduce perioperative stress by implementing combined evidence-based practices with the cooperation of various health professionals as an interdisciplinary team.ERAS pathways include pre-operative patient counselling,regional anesthesia and analgesia techniques,post-operative pain management,early mobilization and early feeding.Studies have shown improvement in the recovery of patients who followed an ERAS program after hip or knee arthroplasty,compared with those who followed a traditional care approach.ERAS protocols reduce post-operative stress,contribute to rapid recovery,shorten length of stay(LOS)without increasing the complications or readmissions,improve patient satisfaction and decrease the hospital costs.We suggest that the ERAS pathway could reduce the LOS in hospital for patients undergoing total hip replacement or total knee replacement.These programs require good organization and handling by the multidisciplinary team.ERAS programs increase patient's satisfaction due to their active participation which they experience as personalized treatment.The aim of the study was to develop an ERAS protocol for oncology patients who undergo bone reconstruction surgeries using massive endoprosthesis,with a view to improving the surgical outcomes.

Effortful control (EC) is a temperamental self-regulatory capacity, defined as the efficiency of executive attention [1], which is related to individual differences in self-regulation. Although effortful control covers some dispositional self-regulatory abilities important to cope with social demands of successful adaptation to school, such as attention regulation, individual differences in EC have recently been associated with school functioning through academic achievement including the efficient use of learning-related behaviors, which have been found to be a necessary precursor of learning and they refer to a set of children’s behaviors that involve organizational skills and appropriate habits of study. Therefore, the aim of this study is to review the literature on EC’s relationship to academic achievement via learning-related behaviors, which reflect the use of metacognitive control processes in kindergarten and elementary school students. The findings indicate that EC affects academic achievement through the facilitation of the efficient use of metacognitive control processes.

Arachis retusa Krapov. et W. C. Gregory et Valls is endemic in the West-central region of Brazil, occurring In areas endangered by human actions. The establishment of in vitro preservation methods for wild species of Arachis is an alternative to seed banks for germplasm storage, multiplication and distribution. The risk of genetic changes Induced by tissue culture and the monitoring of the genetic stability of the biological material before, during and after storage must be considered In the context of conservation. Random amplified polymorphlc ONA （RAPO） and amplified fragment length polymorphism （AFLP） fingerprinting were used to evaluate the genetic stability of in vitro plants originated from cotyledons and embryo axes of A. retusa. Cotyledons originated shoots through direct organogenesls and embryo axes displayed muItishoot formation Induced by 110 mmol/L and 8.8 mmol/L BAP, respectively. Ninety genomlc regions （loci） generated from RAPO and 372 from AFLP analyses were evaluated. All amplified fragments detected by both techniques in plants derived from the two explant types were monomorphic. The results Indicate that the recovered shoots are genetically stable at the assessed genomic regions.

Effective desalination and anti-biofouling performance via surface immobilized MWCNTs on RO membrane

Desalination is considered a viable method to overcome the issue of water scarcity either from waste water or seawater. For this purpose, this study employed a facile approach to develop surface immobilized oxidized-MWCNTs(o-MWCNTs) onto crosslinked polyvinyl alcohol(PVA) membrane. Firstly, modified polysulphone substrate was synthesized on to which crosslinked PVA layer was spread onto it. PVA layer act as active layer for surface immobilization of o-MWCNTs in varying concentration. The functional group analysis, morphology and roughness of membranes surface was conducted out using FTIR, SEM and AFM respectively. The results showed that modified membranes, immobilized o-MWCNTs enhanced the salt rejection(Na_(2)SO_(4)) upto 99.8%. After contacting with Escherichia coli and Staphylococcus aureus for 2.5 h the bacteria mortalities of the fabricated membrane could reach 96.9%. Furthermore, the antibiofouling tests showed that OP-MWCNTs(1-5) modified membranes have higher anti-biofouling property than the control membrane.
