Macrodomains constitute a protein motif family present in all kingdoms of life1. Emerging evidence indicates that specific macrodomain proteins interact with mono-ADP-ribosylation（MARylation） on target proteins to ‘read’ and ‘erase’ this modification2. Therefore, macrodomains are implicated in many MARylation-dependent cellular activities that include DNA repair processes, gene transcription, and intracellular signaling. Very recently, we found that terminal ADP-ribose glycohydrolase 1（TARG1）, a macrodomain-containing protein is also capable to form complexes with poly-ADP-ribose（PAR） chains in vitro and in cell3. However, the structural basis of the binding of TARG1 to PAR and its biological relevant cross-talk with MARylation are still unknown. Here, an integrated in silico strategy combining docking and molecular dynamics（MD） simulation was applied to predict the structural models of TARG1 bound to both branched and linear forms of pentameric PAR chains. Our 1000 ns-long MD simulations showed that each PAR chain may stably bind at the lip region of the enzymatic site of TARG1 with one terminal ADP-ribose unit inserted into the ADP-ribose binding pocket. Specifically, the binding mode of the former well resembled the one of ADP-ribose alone with TARG1 as observed in NMR structure4. Additionally, the key residues selectively engaged with branched and linear PAR chains were identified. The mutagenesis assays on these residues are ongoing to validate the complex models predicted here. Our work will substantially promote and accelerate the investigations of the role of TARG1 in the cross-talk between mono-and poly-ADP-ribosylations for cell physiology and disease-related processes.

Genotyping-by-sequencing of pear (Pyrus spp.) accessions unravels novel patterns of genetic diversity and selection footprints

Understanding of genetic diversity and marker-trait relationships in pears(Pyrus spp.)forms an important part of gene conservation and cultivar *** of Asian and European pear species,and interspecific hybrids were planted in a common garden ***-by-sequencing(GBS)was used to genotype 214 accessions,which were also phenotyped for fruit quality traits.A combination of selection scans and association analyses were used to identify signatures of *** of genetic diversity,population structure and introgression were also *** 15000 high-quality SNP markers were identified from the GBS data,of which 25%and 11%harboured private alleles for European and Asian species,*** clustering analysis suggested negligible gene flow,resulting in highly significant population differentiation(Fst=0.45)between Asian and European *** hybrids displayed an average of 55%and 45%introgression from their Asian and European ancestors,***(firmness,acidity,shape and so on)variation between accessions was significantly associated with genetic *** frequencies at large-effect SNP loci were significantly different between genetic groups,suggesting footprints of directional *** scan analyses identified over 20 outlier SNP loci with substantial statistical support,likely to be subject to directional selection or closely linked to loci under selection.

Wafer-level mass production of photonic integrated circuits(PIC)has become a technological mainstay in the field of optics and photonics,enabling many novel and disrupting a wide range of existing ***,scalable photonic packaging and system assembly still represents a major challenge that often hinders commercial adoption of PIC-based ***,chip-to-chip and fiber-to-chip connections often rely on so-called active alignment techniques,where the coupling efficiency is continuously measured and optimized during the assembly *** unavoidably leads to technically complex assembly processes and high cost,thereby eliminating most of the inherent scalability advantages of PIC-based *** this paper,we demonstrate that 3D-printed facet-attached microlenses(FaML)can overcome this problem by opening an attractive path towards highly scalable photonic system assembly,relying entirely on passive assembly techniques based on industry-standard machine vision and/or simple mechanical *** can be printed with high precision to the facets of optical components using multi-photon lithography,thereby offering the possibility to shape the emitted beams by freely designed refractive or reflective ***,the emitted beams can be collimated to a comparatively large diameter that is independent of the device-specific mode fields,thereby relaxing both axial and lateral alignment ***,the FaML concept allows to insert discrete optical elements such as optical isolators into the free-space beam paths between PIC *** show the viability and the versatility of the scheme in a series of selected experiments of high technical relevance,comprising pluggable fiber-chip interfaces,the combination of PIC with discrete micro-optical elements such as polarization beam splitters,as well as coupling with ultra-low back-reflection based on non-planar beam paths that only comprise tilted optical *** on our re

Background: Neonatal sepsis is a global health problem that mainly affects low- and middle-income countries. We have previously shown that early neonatal mortality is high at the Ho Teaching Hospital (HTH) of Ghana. We sought to determine the prevalence of neonatal sepsis, sepsis-related mortality, and bacterial species patterns in neonatal and young infant sepsis in this hospital. Methods: A hospital-based study was conducted in the hospital’s neonatal intensive care unit (NICU) from March to June 2018. Blood samples from 96 babies clinically diagnosed with or at risk of sepsis were cultured using the BACTEC 9050® machine. Clinical data including gravida, parity and antibiotic medication before delivery of mother and delivery type, gestation, birth weight and antibiotic medication status were collected for analysis. MALDI-TOF MS identified bacterial isolates, and their identities were confirmed via tuf gene sequence typing. The data were analyzed using GraphPad Prism 8.0.2. Results: Blood cultures were positive in 28 of the babies, with 14 and 12 representing early-onset and late-onset neonatal sepsis, respectively, and two cases of unknown sepsis type. The pygmy marmoset,the smallest of the anthropoid primates,has a broad distribution in Western *** studies using molecular and morphological data have identified two distinct species separated by the Napo and Solimoes-Amazonas ***,reconciling this new biological evidence with current taxonomy,i.e.,two subspecies,Cebuella pygmaea pygmaea(Spix,1823)and Cebuella pygmaea niveiventris(Lönnberg,1940),was problematic given the uncertainty as to whether Spix’s pygmy marmoset(Cebuella pygmaea pygmaea)was collected north or south of the Napo and Solimoes-Amazonas rivers,making it unclear to which of the two newly revealed species the name pygmaea would ***,we present the first molecular data from Spix’s type specimen of Cebuella pygmaea,as well as novel mitochondrial genomes from modern pygmy marmosets sampled near the type locality(Tabatinga)on both sides of the *** these data,we can confirm the correct names of the two species identified,i.e.,*** for animals north of the Napo and Solimoes-Amazonas rivers and *** for animals south of these two *** analyses of the novel genetic data placed into the context of cytochrome b gene sequences from across the range of pygmy marmosets further led us to reevaluate the geographical distribution for the two Cebuella *** dated the split of these two species to 2.54 million years *** discuss additional,more recent,subdivisions within each lineage,as well as potential contact zones between the two species in the headwaters of these rivers.

Pseudomonas syringae ***(Psa)biovar 3,a virulent,canker-inducing pathogen is an economic threat to the kiwifruit(Actinidia spp.)industry *** commercially grown diploid(2×)*** *** is more susceptible to Psa than tetraploid and hexaploid *** information on the genetic loci modulating Psa resistance in kiwifruit is not *** we report mapping of quantitative trait loci(QTLs)regulating resistance to Psa in a diploid kiwifruit population,derived from a cross between an elite Psa-susceptible‘Hort16A’and a resistant male breeding parent *** high-density genetic maps and intensive phenotyping,we identified a single QTL for Psa resistance on Linkage Group(LG)27 of‘Hort16A’revealing 16–19%phenotypic variance and candidate alleles for susceptibility and resistance at this *** addition,six minor QTLs were identified in P1 on distinct LGs,exerting 4–9%*** in the F1 population is improved by additive effects from‘Hort16A’and P1 QTLs providing evidence that divergent genetic pathways interact to combat the virulent Psa *** different bioassays further identified new QTLs for tissue-specific responses to *** genetic marker at LG27 QTL was further verified for association with Psa resistance in diploid Actinidia chinensis *** analysis of Psa-resistant and susceptible genotypes in field revealed hallmarks of basal defense and provided candidate RNA-biomarkers for screening for Psa resistance in greenhouse conditions.

三代红外探测器组件——双色（DC），双波段（DB）以及大规模2-DY0阵要求精细的生产技术：诸如分子束外延（MBE）以及新的列阵制备工艺，此类技术可以满足日趋复杂的器件结构及低成本的需求。AIM公司将采用上述三种技术制备高性能器件以扩大其未来的业务。DC／MW／MW探测器基于II-型超晶格锑化物（IAF通过MBE技术制备而成），像元规格为384×288，像元中心距为40μm。AIM公司的DB／MW／LW／FPA（焦平面列阵）选用了MBE技术，通过在CdZnTe衬底上生长MCT，该技术与IAF公司合作完成。由AIM公司制备了像元规格为640×512，像元中心距为15μm的FPA。尽管在不同的衬底上用MBE方法生长MW／MCT薄膜具有挑战性，但对于制备具备百万像素的大规模2维FPA（MW1280×1024，像元中心距为15μm）而言，这种技术是必要的。本文给出了上述三代FPA及集成探测器杜瓦制冷机组件（IDCAs）的发展现状及近期测试结果。
