Efficacy and long-term evaluation of intramyocardial injection of autologous CD34-enriched PBMSC in old myocardial infarction

Aims: We have shown that autologous transplant of CD34+-enriched peripheral-blood mononuclear cells (PBMSC) could restore depressed myocardial function, and sustain adequate myocardial function 12 months after surgery in patients with old (>one year-old) myocardial infarction. Our aim is to report the long-term morbidity and mortality efficacy of this procedure. Methods and results: Seventy patients with an old anteroseptal myocardial infarction were followed for 2 to 7 years, 35 had a revascularization procedure and received an intra-myocardial injection of autologous CD34+-enriched PBMSC (8 × 108 mononuclear cells/ml including 3 × 107 CD34+ cells/ml)(Group A). Group B patients only had the revascularization. Abnormal pre-surgical values of LVEF (33.2% ± 4.8%), LVDV (178 ± 13.7 ml), LVSV (120 ± 16 m), LVDD (58.9 ± 3.84 mm), E and A waves without contractility in infarction area in group A patients improved to approximate normal values (50% ± 3% for LVEF;90 ± 9.3 ml for LVDV;80 ± 9.9 ml for LVSV;55.3 ± 3 mm for LVDD;5.2 ± 0.5 cm/s for E wave and 4.18 ± 0.3 cm/s for A wave) 1 year after the procedure and have remained unaltered for all the follow-up period. All the patients remain alive. Only seven patients have been readmitted to the hospital for non-myocardial related events. Group B only 11 patients continued alive to 5 years after surgery and LEVF never increased more than 6%, all of them with many hospitalizations (n ≥ 10) by heart failure events. Conclusion: Intramyocardial injection of CD34+ highly enriched PBSC represent an encouraging alternative for patients with severely scarred and dysfunctional myocardium.

Cell therapy has shown beneficial effects on ventricular function and tissue regeneration in patients with acute and chronic myocardial infarction, although with diverse grades of variability in the results, possibly by proportion, subtype and cell cycle status. Objective: Identify and phenotypically characterize, via CXCR4 and SDF-1 expression, the bone marrow cell subpopulations that are mobilized into the bloodstream in patients with Acute Myocardial Infarction (AMI) and Acute Ischemia (AI) such as acute angina and Chronic Ischemia (CI) such as chronic stable angina, and also determine the cell cycle status of these cells. Method: Patients with AMI and AI were recruited in the ICCU, and patients with CI in the departments of cardiology and cardiovascular surgery. The quantification of cellular subpopulations was made by cytofluorometry with a FACS caliburcyto fluorometry (Becton Dickinson) with specific FITC-labeled anti human monoclonal antibodies against CD34, CD133, CD117, CD48, CXCR4, SDF-1 and Ki67 (Becton Dickinson). Serum concentration of IL-6 and IL-8 were determined by a sequential solid phase chemiluminescent assay performed in a SIEMENS IMMULITE 1000 Analyzer. Statistical analysis was made with the SPSS version 20.0 for Windows. A p value 3/ml) than that in AI (9.2 ± 1.3 × 103/ml) and CI (6.6 ± 1.1 × 103/ml) patients (p p = 0.22 to 0.39), but interestingly in AMI and AI patients, cells were CXCR4+ in almost half of these mobilized cells, although the proportion was significantly higher in AMI patients (46.8% ± 7.1% to 55.7% ± 6.3% vs 23% ± 1.6% to 28.4% ± 2.1%, p = 0.03 to 0.05). A similar behavior was observed with the Ki67 antibody (29.9% ± 2.1% to 36.1% ± 6.3% vs 10% ± 1.2% to 24% ± 1.1%, p = 0.001 to 0.05). Bivariate analysis of the results showed a significant correlation of the cell proportion in AMI but not in AI and CI patients (p = 0.001 to 0.05;0.12 to 0.87 and 0.17 to 0.92 respectively). The juvenile dispersal of raptors is a crucial stage that stretches from parental independence to the establishment of the first breeding *** 2012 and 2020,44 juvenile red kites Milvus milvus from the Spanish breeding population were tagged using GPS telemetry to study their *** left the parental breeding area at the end of their first summer and performed wandering movements throughout the Iberian Peninsula,returning to the parental breeding area the following year,repeating the same pattern until they settled in their first breeding *** analyzed the mean distance from the nest,the maximum reached distances,and the traveled distances(daily and hourly)during the first 2 years of dispersal and compared *** the high individual variability,variables describing the dispersal movements of juveniles showed a decreasing trend during the second dispersal year:80% of individuals reached a shorter maximum distance in the second year,70% decreased their mean distance to the nest,65% decreased their hourly traveled distances,and 50% decreased their daily traveled *** the other hand,the red kites usually combined wandering movements with the establishment of temporary settlement areas(TSA).The average duration of settlement in the TSAs was 75±40 days(up to 182 days)and was located at 182±168 km from the *** those areas,juveniles used 781.0±1895.0 km^(2)(KDE 95%).Some of the TSAs were used by several individuals,which suggests that these areas might be good targets for conservation in future management plans.

Digestate, the product obtained after anaerobic digestion of organic waste for biogas production, is rich in plant nutrients and might be used to fertilize crops. Wheat(Triticum spp. L.) was fertilized with digestate, urea, or left unfertilized and cultivated in the greenhouse for 120 d. Emissions of greenhouse gasses(carbon dioxide(CO_2), methane(CH_4), and nitrous oxide(N_2O)) were monitored and plant growth characteristics were determined at harvest. The digestate was characterized for heavy metals, pathogens, and C and N mineralization potential in an aerobic incubation experiment. No Salmonella spp., Shigella spp., or viable eggs of helminths were detected in the digested pig slurry, but the number of faecal coliforms was as high as 3.6 × 10~4colony-forming units(CFU) g-^(1)dry digestate. The concentrations of heavy metals did not surpass the upper limits established by US Environmental Protection Agency(EPA). After 28 d, 17% of the organic C(436 g kg^(-1)dry digestate) and 8% of the organic N(6.92 g kg^(-1)dry digestate)were mineralized. Emissions of CO_2 and CH_4 were not significantly affected by fertilization in the wheat-cultivated soil, but digestate significantly increased the cumulative N_2O emission by 5 times compared to the urea-amended soil and 63 times compared to the uncultivated unfertilized soil. It could be concluded that digestate was nutrient rich and low in heavy metals and pathogens, and did not affect emissions of CH_4 and CO_2 when applied to a soil cultivated with wheat, but increased emission of N_2O.

Unraveling the factors that determine variation of diversity in tropical mountain systems is a topic for debate in plant *** is especially true in areas where topography is complex due to volcano elevational gradients and where forests are vulnerable to human *** this study we used a set of climatic(temperature,rainfall,and radiation solar),topographic(elevation,slope aspect,and slope orientation)and human disturbance variables to determine their effect on diversity and composition patterns of a tree community,considering three slope aspects of a tropical volcano in southeastern *** sampled trees in seventy 0.1-ha plots distributed on three slope aspects of the Tacanávolcano along an elevational gradient of 1500 to 2500 *** determined diversity patterns(general tree richness,exponential of Shannon index,and pioneer species richness)with linear regression models,and for beta diversity,we used a dissimilarity index(within and between elevational bands 100 m wide).The effect of a set of environmental and human disturbance variables on tree diversity and community composition was analyzed with general linear models and multivariate analyses,*** registered 2,949 individual trees belonging to 176 species and *** average species richness and alpha diversity per plot were 13(standard deviation±6)and 9(±5),*** tree richness and alpha diversity increased in the middle part(unimodal patterns)of the elevational gradient,but pioneer species richness decreased linearly with *** variance explained by general linear models was greater in richness(32%)than in alpha diversity(25.3%).The most important predictor variables were temperature(elevational gradient),which explained the unimodal pattern(richness and alpha diversity increase at intermediate levels of temperature),and slope orientation,which explained the increase in richness and alpha diversity toward the geographic *** temperature had a signific

Plastidial ω-3 desaturase FAD7 is a major contributor to trienoic fatty acid biosynthesis in the leaves of Arabidopsis plants. However, the precise contribution of the other plastidial ω-3 desaturase, FAD8, is poorly understood. Fatty acid and lipid analysis of several ω-3 desaturase mutants, including two insertion lines of AtFAD7 and AtFAD8, showed that FAD8 partially compensated the disruption of the AtFAD7 gene at 22℃, indicating that FAD8 was active at this growth temperature, contrasting to previous observations that circumscribed the FAD8 activity at low temperatures. Our data revealed that FAD8 had a higher selectivity for 18：2 acyl-lipid substrates and a higher preference for lipids other than galactolipids, particularly phosphatidylglycerol, at any of the temperatures studied. Differences in the mechanism controlling AtFAD7 and AtFAD8 gene expression at different temperatures were also detected. Confocal microscopy and biochemical analysis of FAD8-YFP over-expressing lines confirmed the chloroplast envelope localization of FAD8. Co-localization experiments suggested that FAD8 and FAD7 might be located in close vicinity in the envelope membrane. FAD8-YFP over-expressing lines showed a specific increase in 18：3 fatty acids at 22℃. Together, these results indicate that the function of both plastidial ω-3 desaturases is coordinated in a non-redundant manner.
