Various recent.report. on t.berculosis have alarmed an increase in t.e pat.ent.class and subsequent.deat. rat.s across t.e globe. Over and above t.e spread of more dangerous and fat.l forms of t.berculosis like MDR-t. i.e. mult.ple-drug resist.nce t.berculosis, XDR-t. i.e. ext.nsively-drug resist.nce t.berculosis & t.R-t. i.e. t.t.l-drug resist.nce t.berculosis has forwarded an urgent.need t. discover novel ant.t.bercular agent.. t.e current.work is aimed at.combining t.o previously well-known pharmacophores (pyrazoline and benzoxazole nucleus) in order t. design and synt.esize a series of novel benzoxazole-based pyrazoline derivat.ves. t.e synt.esized t.rget.compounds were st.uct.rally confirmed by LCMS, 1H-NMR and 13C-NMR analysis. t.e t.rget.compounds were In vit.o evaluat.d against.M. t.berculosis H37Rv st.ain, mult.drug-resist.nt.t. (MDR-t.) and ext.nsively drug-resist.nt.t. (XDR-t.) st.ains. t.e In vit.o screening result. depict.d t.at.majorit. of t.e t.rget.compounds displayed pot.nt.act.vit. wit. MIC in a range of ~0.8 t. 6.25 μg/mL. Many compounds were found t. be more pot.nt.t.an isoniazid against.MDR-t. wit. MIC value 3.12 μg/mL and XDR-t. wit. MIC value 12.5 μg/mL. Cyt.t.xicit. assay of t.ese act.ve compounds on VERO cell lines also displayed good select.vit. index.

A series of pyrazoline-based new het.rocycles have recent.y been synt.esized from our group where some of t.e compounds display pot.nt.ant.-t.bercular act.vit. against.Mycobact.rium t.berculosis H37Rv. In order t. furt.er explore t.e pot.ncy of t.e compounds, quant.t.t.ve st.uct.re act.vit. relat.onship st.dy is carried out.using genet.c funct.on approximat.on. St.t.st.cally significant.(r2 = 0.85) and predict.ve (r2pred=0.89 and r2m=0.74)?QSAR models are developed. It.is evident.from t.e QSAR st.dy t.at.majorit. of t.e ant.-t.bercular act.vit. is found t. be driven by lipophilicit.. Also, molecular solubilit., Jurs and shadow descript.rs influence t.e biological act.vit. significant.y. Also, posit.ve cont.ibut.on of molecular shadow descript.rs suggest. t.at.molecules wit. bulkier subst.t.ent. are more likely t. enhance ant.-t.bercular act.vit.. Since t.e developed QSAR models are found t. be st.t.st.cally significant.and predict.ve, t.ey pot.nt.ally can be applied for predict.ng ant.-t.bercular act.vit. of new molecules for priorit.zat.on of molecules for synt.esis.

In-plane shear t.st.ng has emerged as an import.nt.met.od t. overcome t.e challenges associat.d wit. t.nsion and compression t.st. and can provide addit.onal informat.on about.deformat.on behaviors under large plast.c st.ains. Mat.rials such as Mg alloys wit. poor levels of bot. duct.lit. and formabilit. cannot.accommodat. large plast.c st.ains. t.erefore, t.nsion and compression t.st. have limit.t.ons in explaining t.e mat.rial behaviors t.at.occur during sheet.met.l forming where large plast.c st.ains are int.oduced. Many st.dies have been conduct.d t. explain t.e deformat.on behaviors of Mg alloys under shear deformat.on t.chniques. t.ese include severe plast.c deformat.on(SPD), especially t.e equal channel angular pressing(ECAP)and equal channel angular ext.usion, rolling combined wit. shear deformat.on i.e. different.al speed rolling(DSR), and also in-plane shear for sheet.met.ls, part.cularly under large levels of plast.c st.ain. t.ese in-plane shear t.chnique involves t.e Miyauchi shear t.st.
