Background Branched-chain amino acids(BCAAs),including L-leucine(L-Leu),L-isoleucine(L-Ile),L-valine(L-Val),and L-arginine(L-Arg),play a crucial role in mammary gland development,secretion of milk and regulation of the catabolic state and immune response of lactating ***,it has recently been suggested that free amino acids(AAs)can also act as microbial *** study aimed at evaluating whether the supplementation of lactating sows with BCAAs(9,4.5 and 9 g/d/sow of L-Val,L-Ile and L-Leu,respectively)and/or L-Arg(22.5 g/d/sow),above the estimated nutritional requirement,could influence the physiological and immunological parameters,microbial profile,colostrum and milk composition and performance of sows and their *** At d 41,piglets born from the sows supplemented with the AAs were heavier(P=0.03).The BCAAs increased glucose and prolactin(P<0.05)in the sows’serum at d 27,tended to increase immunoglobulin A(IgA)and IgM in the colostrum(P=0.06),increased the IgA(P=0.004)in the milk at d 20 and tended to increase lymphocyte%in the sows’blood at d 27(P=0.07).Furthermore,the BCAAs tended to reduce the Chao1 and Shannon microbial indices(P<0.10)in the sows’*** BCAA group was discriminated by Prevotellaceae_UCG-004,Erysipelatoclostridiaceae UCG-004,the Rikenellaceae_RC9_gut_group and *** reduced piglet mortality pre-(d 7,d 14)and post-weaning(d 41)(P<0.05).Furthermore,Arg increased the IgM in the sow serum at d 10(P=0.05),glucose and prolactin(P<0.05)in the sow serum at d 27 and the monocyte percentage in the piglet blood at d 27(P=0.025)and their jejunal expression of NFKB2(P=0.035)while it reduced the expression of GPX-2(P=0.024).The faecal microbiota of the sows in Arg group was discriminated by *** combination of BCAAs and Arg tended to increase spermine at d 27(P=0.099),tended to increase the Igs(IgA and IgG,P<0.10)at d 20 in the milk,favoured the faecal colonisation of Oscillospiraceae UCG

Interaction of major genes predisposing to hepatocellular carcinoma with genes encoding signal transduction pathways influences tumor phenotype and prognosis

Studies on rodents and humans demonstrate an inherited predisposition to hepatocellular carcinoma （HCC）. Analysis of the molecular alterations involved in the acquisition of a phenotype resistant or susceptible to hepatocarcinogenesis showed a deregulation of G1 and S phases in HCC of genetically susceptible F344 rats and a G1-S block in lesions of resistant Brown norway （BN） rats. Unrestrained extracellular signal-regulated kinase （ERK） activity linked to proteasomal degradation of dual-specificity phosphatase 1 （DUSP1）, a specific ERK inhibitor, by the CKS1-SKP2 ubiquitin ligase complex occurs in more aggressive HCC of F344 rats and humans. This mechanism is less active in HCC of BN rats and human HCC with better prognosis. Upregulation of iNos cross-talk with IKK/NF-KB and RAS/ERK pathways occurs in rodent liver lesions at higher levels in the most aggressive models represented by HCC of F344 rats and c-Myc-TGF-α transgenic mice. iNOS, IKK/NF-κB, and RAS/ERK upregulation is highest in human HCC with a poorer prognosis and positively correlates with tumor proliferation, genomic instability and microvascularization, and negatively with apoptosis. Thus, cell cycle regulation and the activity of signal transduction pathways seem to be modulated by HCC modifier genes, and differences in their efficiency influence the susceptibility to hepatocarcinogenesis and probably the prognosis of human HCC.

In this letter to the Editor, we would like to comment on the article by Betcher et al., concerning the possibility of teaching military trainees to obtain accurate optic nerve sheath diameter measurements, using a brief didactic and a hands-on training session. In particular, this letter notes the importance of the measurement of optic nerve sheath diameter in detecting the eventual elevated intracranial pressure following traumatic brain injury, highlights several limitations in the use of B-scan for such a purpose and suggests a more accurate evaluation with the standardized A-scan.

Good and bad dispositions between archaea and bacteria in the human gut:New insights from metagenomic survey and co-occurrence analysis

Archaea are an understudied component of the human *** this study,the gut archaeome and bacteriome of 60 healthy adults from different region were analyzed by whole-genome shotgun *** were ubiquitously found in a wide range of abundances,reaching up to 7.2%.The dominant archaeal phylum was Methanobacteriota,specifically the family Methanobacteriaceae,encompassing more than 50%of Archaea in 50 *** previously underestimated Thermoplasmatota,mostly composed of Methanomassiliicoccaceae,dominated in 10 subjects(>50%)and was present in all others except ***,the sole other archaeal phylum,occurred in negligible concentration,except for two samples(4.6-4.8%).This finding confirmed that the human gut archaeome is primarily composed of methanogenic organisms and among the known methanogenic pathway:i)hydrogenotrophic reduction of CO_(2) is the predominant,being the genus Methanobrevibacter and the species Methanobrevibacter smithii the most abundant in the majority of the samples;ii)the second pathway,that involved Methanomassiliicoccales,was the hydrogenotrophic reduction of methyl-compounds;iii)dismutation of acetate or methyl-compounds seemed to be ***-occurrence analysis allowed to unravel correlations between Archaea and Bacteria that shapes the overall structure of the microbial community,allowing to depict a clearer picture of the human gut archaeome.

An experimental investigation of the crystallographic,Raman and transport properties of the Ce_(1-x)(Nd_(0.74)Tm_(0.26))_xO_(2-x/2)(0.1≤x≤0.6) doped ceria system was performed with the aim of setting out correlations between structural features and ionic conductivity of the *** chosen composition ensures that the average size of the Nd^(3+) and Tm^(3+) doping ions coincides with the one of Sm^(3+);even so,the studied system presents larger cell parameters and a wider compositional extent of the CeO_(2)-based solid solution than Sm-doped ***,the occurrence of two different activation energies to ionic conduction below and above ~750 K determines the existence of two distinct conduction *** described experimental results agree with the formation below the threshold temperature of1 V_O2 Tm'_(Ce) trimers,which promote the incorporation of Nd'_(Ce) isolated defects into the CeO_(2)-based solid *** the high temperature range the dissociation of trimers induces the appearance of a lower activation energy;the extrapolation of its value at infinite dilution provides a result in good accordance with the expected binding energy of 1 V_ORE'_(Ce) dimers,pointing at their stability even in the high temperature conduction regime.

Assay of ghrelin concentration in infant formulas and breast milk

AIM: To test if total ghrelin is present in infant formulas. METHODS: Using a radioimmunoassay, we measured total ghrelin concentrations in 19 samples of commercial infant formulas and in 20 samples of human milk. We also determined ghrelin concentration in the serum of infants and lactating mothers. RESULTS: Ghrelin concentrations were significantly higher in artificial milk (2007.1 ± 1725.36 pg/mL) than in human milk (828.17 ± 323.32 pg/mL) (P = 0.005). The mean ghrelin concentration in infant serum (n = 56) was 1115.86 ± 42.89 pg/mL, and was significantly higher (P = 0.023) in formula-fed infants (1247.93 ± 328.07 pg/mL) than in breast-fed infants (1045.7 ± 263.38 pg/mL). The mean serum ghrelin concentration (mean ± SD) in lactating mothers (n = 20) was 1319.18 ± 140.18 pg/mL. CONCLUSION: This study provides evidence that total ghrelin is present in infant formulas. This findingraises diverse questions regarding the uptake, absorption and metabolic effects of this hormone.

Neuroendocrine tumors of the gastro-entero-pancreatic system

Gastro-entero-pancreatic (GEP) neuroendocrine tumors (NETs) are rare neoplasms, although their prevalence has increased substantially over the past three decades. Moreover, there has been an increased clinical recognition and characterization of these neoplasms. They show extremely variable biological behavior and clinical course. Most NETs have endocrine function and secrete peptides and neuroamines that cause distinct clinical syndromes, including carcinoid syndrome; however, many are clinically silent until late presentation with mass effects. Investigation and management should be individualized for each patient, taking into account the likely natural history of the tumor and general health of the patient. Management strategies include surgery for cure or palliation, and a variety of other cytoreductive techniques, and medical treatment including chemotherapy, and biotherapy to control symptoms due to hormone release and tumor growth, with somatostatin analogues (SSAs) and alphainterferon. New biological agents and somatostatintagged radionuclides are under investigation. Advances in the therapy and development of centers of excellence which coordinate multicenter studies, are needed to improve diagnosis, treatment and therefore survival of patients with GEP NETs.
