Background: Bone marrow lesions (BMLs) are associated with osteoarthritis (OA). We assessed the performance of two commonly used MRI sequences, IW-TSE and DESS, for reliability in the detection of BMLs and sensitivity to estimate change over time. We suggested that the IW-TSE would demonstrate higher sensitivity to change than DESS in the assessment of BML prevalence and change over time. This study was performed using a subset of the Osteoarthritis Initiative (OAI) cohort. Methods: A sub-group of 144 patients was selected from the OAI progression cohort who all had IW-TSE and DESS MRI acquisitions at baseline and 24 months. BMLs were assessed using a semi-quantitative scale in the global knee, medial and lateral compartments, and subregions. Intra-reader reliability was assessed on a subset of 51 patients. Results: Intra-reader reliability was substantial for the global knee ≥ 0.64, medial ≥ 0.70, and lateral ≥ 0.63 compartments for IW-TSE and DESS. The prevalence of BML detected at baseline was only slightly greater for IW-TSE compared to DESS. The mean BML score at baseline was significantly higher (p ≤ 0.006) for the IW-TSE than the DESS. However, mean change at 24 months was similar for both sequences for all regions except the medial compartment (p = 0.034) and medial femur (p = 0.015) where they were significantly higher for DESS than IW-TSE. Moreover, the prevalence of BML change at 24 months was similar in all regions except the global knee (p = 0.047) and the lateral tibial plateau (p = 0.031). Conclusion: This study does not suggest superior sensitivity to change of one sequence over the other for almost all the regions. The only difference is a higher BML mean change over time detected by the DESS sequence in the medial compartment and femur. These data bring into perspective that both sequences seem equivalent regarding their use for the assessment of BML in clinical trials.

Although the general concept of nanotechnology relies on exploitation of size-dependent properties of nanoscaled materials,the relation between the size/morphology of nanoparticles with their biological activity remains not well ***,we aimed at investigating the biological activity of Se nanoparticles,one of the most promising candidates of nanomaterials for biomedicine,possessing the same crystal structure,but differing in morphology(nanorods *** particles)and aspect ratios(AR,11.5 vs.22.3 vs.1.0)in human cells and BALB/c ***,we report that in case of nanorod-shaped Se nanomaterials,AR is a critical factor describing their cytotoxicity and ***,spherical nanoparticles(AR 1.0)do not fit this statement and exhibit markedly higher cytotoxicity than lower-AR Se *** of cytotoxicity,we also show that morphology and size substantially affect the uptake and intracellular fate of Se *** line with in vitro data,in vivo *** of Se nanomaterials revealed the highest toxicity for higher-AR nanorods followed by spherical nanoparticles and lower-AR ***,we revealed that Se nanomaterials are able to alter intracellular redox homeostasis,and affect the acidic intracellular vesicles and cytoskeletal architecture in a size-and morphology-dependent *** the tested nanoparticles were produced from the similar sources,their behavior differs markedly,since each type is promising for several various application scenarios,and the presented testing protocol could serve as a concept standardizing the biological relevance of the size and morphology of the various types of nanomaterials and nanoparticles.

AIM To assess the occurrence of autoimmune pancreatitis(AIP) in pancreatic resections performed for focal pancreatic *** We performed a retrospective analysis of medical records of all patients who underwent pancreatic resection for a focal pancreatic enlargement at our tertiary center from January 2000 to July 2013. The indication for surgery was suspicion of a tumor based on clinical presentation, imaging findings and laboratory evaluations. The diagnosis of AIP was based on histology findings. An experienced pathologist specialized in pancreatic disease reviewed all the cases and confirmed the diagnosis in pancreatic resection specimens suggestive of AIP. The histological diagnosis of AIP was set according to the international consensus diagnostic *** Two hundred ninety-five pancreatic resections were performed in 201 men and 94 women. AIP was diagnosed in 15 patients(5.1%, 12 men and 3 women) based on histology of the resected specimen. Six of them had AIP type 1, nine were diagnosed with AIP type 2. Pancreatic adenocarcinoma(PC) was also present in six patients with AIP(40%), all six were men. Patients with AIP + PC were significantly older(60.5 vs 49 years of age, P = 0.045), more likely to have been recently diagnosed with diabetes(67% vs 11%, P = 0.09), and had experienced greater weight loss(15.5 kg vs 8.5 kg, P = 0.03) than AIP patients without PC. AIP was not diagnosed in any patients prior to surgery; however, the diagnostic algorithm was not fully completed in every case. CONCLUSION The possible co-occurrence of PC and AIP suggests that preoperative diagnosis of AIP does not rule out simultaneous presence of PC.

The main application of hydrocyclones in most industry sectors is the separation of particles from *** thickening,however,the separation properties of the cyclone should be adjustable,which requires a different operating ***,we investigated an add-on thickening module,consisting of a conical plug and a secondary vortex finder in the underflow of the *** scale experiments are carried out to investigate the function of the concept of annular gap modification by the use of the thickening *** the second step,we transferred this concept to industrial scale for field tests at a municipal wastewater treatment *** the field tests,geometrical properties(annular gap modification)and operational parameters(volume flow rate,sludge type)were *** results show that thickening in terms of increasing the total solids content of sludge is possible with the modified *** the field tests,a thickening factor(total solids content of thickened sludge related to feed sludge)of 1.31(average)was reached for digested ***,a hydrocyclone could be a promising addition in wastewater treatment plants for dewatering of sludge.

Dear Editor,Virus-induced gene silencing （VIGS） is a fast and powerful method to study gene function in plants （Burch-Smith et al., 2004）. It is based on plant defense mechanisms against viral gene replication and allows high-throughput silencing of genes of inter- est （SenthiI-Kumar and Mysore, 2014）. The molecular mechanisms involved in post-transcriptional gene silencing （PTGS） have been studied intensively, and its steps are well known. The silencing pro- cess begins with the recognition through Dicer-like ribonucleases （DCL） of double-stranded RNA （dsRNA） that is generated during viral replication. Upon recognition, the dsRNA is processed into 21-24 nucleotide fragments, termed small interfering RNA （siRNA）. Based on sequence homology, both viral siRNA and plant mRNA associate with an RNA-induced silencing complex （RISC） and are targeted for degradation （Llave, 2010）.

Diagnostic accuracy of MRI and PET/CT for neck staging prior to salvage total laryngectomy

Aim:Lymph node(LN)metastases are associated with poor outcomes in patients with recurrent larynx squamous cell carcinoma(LSCC).Neck dissection(ND)is therefore commonly performed along with salvage total laryngectomy(STL).Here,we assess the rate of occult LN metastases and the diagnostic value of MRI and PET/CT for detecting them in recurrent ***:This retrospective study included patients with recurrent LSCC after primary(chemo)radiotherapy[(C)RT]who were re-staged by MRI and/or PET/CT and treated with STL and ND between 2004 and *** histopathology of ND samples was used as the reference ***:Forty-one patients were *** prevalence of occult metastases in MRI-negative and PET/CT-negative neck nodes was between 3.2%and 6.1%.Negative predictive values of neck node re-staging were 93.9%for MRI,96.8%for PET/CT,and 96.2%for MRI and PET/CT ***:Both MRI and PET/CT afforded good negative predictive values for nodal staging in patients with recurrent LSCC after(C)RT prior to *** selected patients,these radiological modalities,particularly PET/CT,could help to avoid unnecessary surgery to the neck and its associated morbidity.

Distinct gut microbiota profiles in patients with primary sclerosing cholangitis and ulcerative colitis

AIM To characterize the gut bacterial microbiota of patients with primary sclerosing cholangitis(PSC) and ulcerative colitis(UC).METHODS Stool samples were collected and relevant clinical data obtained from 106 study participants, 43 PSC patients with(n = 32) or without(n = 11) concomitant inflammatory bowel disease, 32 UC patients, and 31 healthy controls. The V3 and V4 regions of the 16 S ribosomal RNA gene were sequenced on Illumina Mi Seq platform to cover low taxonomic levels. Data were further processed in QIIME employing Ma As Lin and LEf Se tools for analysis of the output *** Microbial profiles in both PSC and UC were characterized by low bacterial diversity and significant change in global microbial composition. Rothia, Enterococcus, Streptococcus, Veillonella, and three other genera were markedly overrepresented in PSC regardless of concomitant inflammatory bowel disease(IBD). Rothia, Veillonella and Streptococcus were tracked to the species level to identify Rothia mucilaginosa, Streptococcus infantus, S. alactolyticus, and S. equi along with Veillonella parvula and V. dispar. PSC was further characterized by decreased abundance of Adlercreutzia equolifaciens and Prevotella copri. Decrease in genus Phascolarctobacterium was linked to presence of colonic inflammation regardless of IBD phenotype. Akkermansia muciniphila, Butyricicoccus pullicaecorum and Clostridium colinum were decreased in UC along with genus Roseburia. Low levels of serum albumin were significantly correlated with enrichment of order *** PSC is associated with specific gut microbes independently of concomitant IBD and several bacterial taxa clearly distinguish IBD phenotypes(PSC-IBD and UC).
