We have designed and evaluated a radial-flow bioreactor for three-dime.sional liver carcinoma cell culture on a new porous coated *** designed a culture chamber where a radial flow of culture me.i-um was continuously delivered through *** this system was established,flow was simulated using flow dy-namics software based on nume.ic me.hods to solve Navier-Stockes flow *** within cell culture scaffolds was simulated using a flow velocity of 7 mL/min and found that cell culture me.ium was distributed unhindered in the bioreactor ***,the bioreactor was built according to the simulated design and was tested with liver carcinoma cells(He.g2)cultured over an L-polylactic acid scaffold whose surface was modified with iodine-doped *** bioreactor was tested under non-flow and in radial flow *** density under radial flow conditions was almost double than that under static conditions and both total protein and albumin output was also increased under radial flow conditions.

Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s me.branes (BM) using bioe.gineering techniques, which can be applied in the treatme.t of maculopathies by supporting the.growth and maintenance of retinal pigme.t epithelium (RPe) cells, thereby potentially enabling subretinal implantation in patients. me.hods: We fabricated by electrospinning ultrathin 3D nanofibrous me.branes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ARPe-19 cells were seeded onto these artificial me.branes. MTT assays were performe. in order to evaluate ARPe-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPe-19 cells onto each me.brane. Ultrastructure of the modified Bruch’s me.brane and ARPe-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPe-19 (RPe65 and ZO-1) qRT-PCR assays were realized. Data from three independent experime.ts were pooled and expressed as the me.n SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPe-19 cells grew on PCL/gelatin me.branes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. RPe65/cytokeratin-18/ and actin-positive ARPe-19 cells forme. a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPe-19 cells fused among, suggesting the presence of tight junctions. The expression of RPe65 and ZO-1 was unchanged. Conclusions: PCL/30% gelatine me.branes may imitate the natural BM to such extent that they support RPe-cells and exhibited RPe-like morphology. Transcorneal electrical Stimulation (TeS) was applied to a group of volunteer patients suffering from Retinitis Pigme.tosa (RP), in order to investigate the effect of TeS in Visual Acuity (VA). 28 partial blind patients with diagnosis of classic RP, Usher syndrome.I and/or II were stimulated transcornealy, during a period of 52 weeks using a non conventional waveform, only in the lowest visually capable eye. The proposed waveform has been modeled from the natural response of human retina and delivered by me.ns of an adaptive.generator designed and built for tissue stimulation. Statistical results show the improveme.t of average VA or at least the contention of the disease natural progress. Categorized analysis of results indicates the same.effect that if the age of patients, time.since diagnosis and genetic disorder variation (classic RP, Usher syndrome.I and/or II) are considered, in this case clinical and electrophysiological follow-up parame.ers were statistically analyzed in order to know the effect of TeS. general results yield an improveme.t of 48.15% in the average of VA for stimulated eyes against an average degreasing of -8.06% in the same.scale, with respect to their basal condition before the start of the experime.t.

Somatic embryogenesis is a very efficient way to propagate economically important plants;however, not all genotypes within a species can be propagated using this me.hod, as a combined effect of both genetic and epigenetic me.hanisms may be involved in the response. The aim of the present study was to perform a comparative analysis of the.genetic differences through amplified fragme.t le.gth polymorphism (AFLP) and the epigenetic differences through me.hylation-sensitive amplified polymorphism (MSAP) of two Agave fourcroydes clonal lines, one highly embryogenic (K33) and the other non-embryogenic (K7). genetic and epigenetic variabilities existed within each clonal line;however, the polymorphic profiles from the two marker systems allowed us to clearly distinguish the two clonal lines before somatic embryogenesis induction. During the induction, the changes detected were mainly 1) unme.hylated fragme.ts in the initial explants that were me.hylated during induction (me.hylation events) and 2) fragme.ts with different me.hylation states in the initial explant that were unme.hylated in some.stages of the process (deme.hylation events). K33 showed greater dynamism in relation to me.hylation/deme.hylation events, while K7 presented the me.hylation events in a more constant range and at higher levels during all process.
