Differential Expression of Leptin and Leptin Receptor m.NA in Obese and Non-Obese Wom.n across the m.nstrual Cycle Title

OBJECTIVE: The aim.of this study was to investigate the m.ssenger ribonucleic acid expression of leptin (L-m.NA) and the leptin receptor (OB-Rm.NA) in subcutaneous fat tissue and to determ.ne their relationship with leptin and testosterone and steroid serum.levels over the course of the m.nstrual cycle (m.), in obese (OBW) and non-obese wom.n (NOW). DESIGN: Observational study investigating m.NA transcript expression of leptin and the leptin receptor from.hum.n subcutaneous fat specim.ns, as well as leptin serum.levels, both in relation to the testosterone and sex horm.nes during the m. in obese and non-obese wom.n. m.THODS: Nineteen wom.n, thirteen with a body m.ss index (Bm.) > 25 Kg/m. and 6 with Bm. 2 were included. RT-PCR was used to detect L-m.NA transcripts of leptin and its receptor in sam.les from.28 fat biopsies taken from.seven volunteers (20 from.OW and 8 from.NOW). The serum.concentrations of leptin and steroid horm.nes were m.asured throughout the m. using radioinm.no analysis. RESULTS: The percentages of wom.n expressing leptin L-m.NA during the phases of the m. were: early follicular (EF), 14.3%;late follicular (LF), 28.6%;early luteal (EL), 14.3%;and late luteal (LL), 42.9%. The percentages of OB-Rm.NA were: EF, 71.4%;LF, 14.3%;EL, 28.6%;and LL, 71.4%. Leptin serum.levels were directly related to Bm. (p m.NA expression and its receptor in subcutaneous hum.n tissue are regulated throughout the m. in a periodic m.nner. The physiological role of testosterone in regulation of expression L-m.NA and OB-Rm.NA during the secretory phase of the m. in obese wom.n requires further investigation.

Lodging occurs when the crop canopy is too heavy for the strength of the stem.and it fallsover onto the ground. This decreases crop yield and quality, and it m.kes harvest difficult.A research experim.nt was set up in a spearm.nt field on a center pivot with m.d elevationspray application (m.SA) overhead sprinklers, where the water was applied from.a “m.delevation” of 2 m.above the ground level (AGL), and low elevation precision application(LEPA) sprinklers, where the water was em.tted directly onto the soil surface through draghoses without wetting the crop canopy. Every-other span of this full-size center pivot wasconfigured with m.SA and LEPA sprinklers alternatively. In 2018, im.gery was collectedwith an unm.nned aerial vehicle (UAV) from.a cross section of this field. In 2019, a crosssection was again collected, but in addition UAV im.gery was collected from.m.rked lodgedand un-lodged areas of the field to validate the lodging detection m.thod. These UAV-basedim.gery data were captured with a ground sam.le distance (GSD) of 0.03 m. This researchintroduces using the texture feature, which is based on im.ge entropy, was used to evaluate the degree of lodging. The results from.2018 showed that the average entropy of thegrayscale im.ge from.LEPA (5.5 (m.an) ± 0.27 (standard deviation)) was significantly(P m.SA. Also, the entropy valueextracted from.the im.ges in 2019 from.the m.rked un-lodged locations were significantlyhigher com.ared to that of the lodged areas. Overall, the LEPA irrigation treatm.nt was significantly less lodged com.ared to m.SA. m.reover, the entropy value, or texture feature, isa viable m.thod for estim.ting lodging using low altitude RGB im.gery.

Callus cultures of Annona m.ricata and Annona purpurea were induced in m.rashige and Skoog(m.)m.dium.supplem.nted with different concentrations of 1-naphthylacetic acid(NAA),6-benzyladenine(BA)and 2,4-dichlorophenoxyacetic acid(2,4-D)utilized hypocotyls with *** highest percentage of callus form.tion was the treatm.nt supplem.nted with 3 m. L-1 NAA for ***(100%)while for *** in lower percentage(75%).BA stim.lated the form.tion of shoots in all the evaluated concentrations,being the concentration of 2 m. L-1 the one that induced the greater form.tion of shoots for ***(23 shoots/explant)and ***(28 shoots/explant).The content of total phenols,flavonoids and antioxidant activity was m.asured in the callus obtained from.both *** results showed that a higher content of total phenols was quantified in callus of ***(27.8 m. g-1 dw)com.ared to ***(23.2 m. g-1 dw).The highest content of total flavonoids was observed in the callus of ***(8.0μg g-1 dw).Antioxidant activity was determ.ned by the 2,2-diphenyl-1-picrylhydracil radical *** concentration required for 50%inhibition(IC50)of the 2,2-diphenyl-1-picrylhydracil radicals were 4.22μg m.-1 in m.thanolic extracts of callus of ***,while in extracts of callus of *** was 2.86μg m.-1,in both cases was greater than that found for *** culture of the species studied in this work represents an alternative for the production of natural antioxidants.

Identification and Quantification of m.nosaccharides in Aloe vera Gel by Acid and Enzym.tic Hydrolysis and Wet Heat Treatm.nt

Aloe vera (Aloe barbadensis m.ller) possesses curative and therapeutic properties attributed to the polysaccharides found in its tissue. This work aim.d to study hydrolyzed (chem.cal and enzym.tical) Aloe vera gel extracts using ultrafiltration m.m.ranes with a pore size of 0.22 m.cron (μm.. Nine chem.cal treatm.nts were achieved with H2SO4 and three wet heat treatm.nts were carried out at a pressure of 1.2 lbf/in2 for 15 m.nutes;in both cases, three different concentrations of Aloe vera gel juice (AGJ) were used: 1.5%, 2.5% and 3.5% w/w. The concentrations of H2SO4 were 0.10%, 0.25% and 0.40% w/w. Chem.cal experim.nts are perform.d over a factorial 32 design and results were analyzed using SPSS software (version 17, SPSS Inc.), finding the one labeled T7 (15 m. of AGJ 3.5% with 15 m. of H2SO4 0.10% added) the best of them. as it leaded to 0.0446 m./m. of liberated glucose. Am.ng the three wet heat treatm.nts, the one labeled TC3 (15 m. of AGJ 3.5% with 15 m. of H2O added) was the best-perform.ng one, as it leaded to 0.292 m./m. of liberated glucose. Furtherm.re, an enzym.tic hydrolysis was carried out using Novozym.s’ Pectinex? AR and Viscozym.?. Hydrolisis with both enzym.s yield to better results than acid hydrolysis: in the treatm.nt with Pectinex? AR, 3.282 m./m. of liberated glucose were obtained and 3.302 m./m. in the treatm.nt with Viscozym.?. The hydrolyzed substances obtained by acid and enzym.tic hydrolysis, as well as by wet heat treatm.nt, were subsequently analyzed by thin layer chrom.tography (TLC), using glucose, galactose and arabinose 1000 ppm.solutions as reference patterns. Am.ng the treatm.nts by H2SO4, the one labeled T4 obtained an Rf value of 50, the sam. as on the galactose reference pattern.

We studied the Chrom.um.VI) rem.val capacity in aqueous solution by the lem.n shell, using the diphenylcarbazide m.thod to evaluate the m.tal concentration. So, the highest biosorption of the m.tal (50 m./L) occurs within 100 m.nutes, at pH of 1.0, and 28°C. According to tem.erature, the highest rem.val was observed at 60°C, in 11 m.nutes, when the m.tal (1 g/L) is com.letely adsorbed. At the analyzed concentrations of Cr(VI), lem.n shell, showed excellent rem.val capacity, besides it rem.ves efficiently the m.tal in situ (97.2% rem.val, 7 days of incubation, 5 g of biom.ss). After 1 hour of incubation the studied biom.ss reduces 1.0 g of Cr(VI) with the sim.ltaneous production of Cr(III);so it can be used to elim.nate it from.industrial wastewater.

Lizards have been reported as im.ortant pollinators on several oceanic *** we evaluate the potential role of Galápagos lava lizards(m.crolophus spp.)as pollinators across their *** 3 years,we sam.pled pollen transport by 9 lava lizard species on the 10 islands where they are present,including 7 single-island ***,only 25 of 296 individuals sam.led(8.4%)transported pollen of 10 plant species,the m.st com.on being Prosopis juliflora,Exodeconus m.ersii,Sesuvium.*** Cordia *** least 8 of these plant species were native,and none were confirm.d as introduced to the *** the low overall proportion of individuals carrying pollen,this was observed in 7 of the nine lizard species,and on 8 of the ten m.in islands(Española,Fernandina,Floreana,Isabela,m.rchena,Pinta,Santa Cruz and Santiago),suggesting that this is a widespread *** results reported here support the potential role of lava lizards as pollinators across their radiation,although they m.y represent a relatively m.dest contribution when com.pared with birds and ***,we cannot discard that lizards m.y be ecologically significant for particular plant species and ecosystem. given the specific clim.tic condition and functional diversity of each island.
