Capecitabine maintenance therapy for XT chemotherapy-sensitive patients with metastatic triple-negative breast cancer

Objective： To investigate the efficacy and safety of capecitabine maintenance therapy（MT） after initial capecitabine plus docetaxel（XT） chemotherapy in patients with metastatic triple-negative breast cancer（m TNBC）.Methods： Fifty-five m TNBC patients treated with XT chemotherapy between May 2007 and June 2013 were retrospectively analyzed. When initial disease control was achieved by the combination chemotherapy, capecitabine was continued for 32 patients（MT）, while 23 patients remained without any treatment（nonMT）. We compared progression-free survival（PFS） and safety of both groups.Results： The median PFS of 55 patients was 8.1 months, overall median PFS time of 32 patients in the capecitabine MT group and 23 in the non-MT group was 10.1 vs. 6.7 months（P=0.032）, respectively. When compared PFS time of maintenance treatment, single-agent capecitabine prolonged PFS by 7.1 months, for non-MT patients, the PFS without any treatment was 3.1 months, and this between-group difference was statistically significant（P=0.003）. Adverse events, including of hematologic toxicity, gastrointestinal toxicities, hand-foot syndrome and abnormal liver function were not significantly different between two groups.Conclusions： After initial disease control was achieved with the XT combination chemotherapy, capecitabine MT can significantly prolong PFS time with a favorable safety profile in m TNBC patients.

Objective:To compare the efficacy of platinum-and non-platinum-based regimens as first-line treatment for advanced triple-negative breast cancer(TNBC)and analyze the relationship between their efficacy and BRCA gene status.Methods:Retrospectively analyze clinical data of 220 patients diagnosed pathologically with advanced TNBC and treated at the Department of Breast Oncology,Peking University Cancer Hospital from 2013 to 2018 and evaluate the efficacy of chemotherapy.A total of 114 patients had BRCA1/2 gene tested by next generation sequencing(NGS)using peripheral blood,and we analyzed the correlation between their efficacy and BRCA1/2 gene status.Results:Non-platinum-based chemotherapy(NPCT)was administered to 129 and platinum-based chemotherapy(PBCT)to 91 study patients.The clinical benefit rate(CBR)and median progression-free survival(PFS)were not statistically different between NPCT and PBCT groups.The median overall survival(OS)was 30.0 and 22.5 months for PBCT and NPCT group,respectively[P=0.090,hazard ratios(HR)=0.703].BRCA status was assessed in 114 patients,14 of whom had deleterious germline BRCA1/2(g BRCA)mutations(seven in each group).In PBCT group,the CBR was 85.7%and 35.1%for patients with and without deleterious g BRCA mutations,respectively(P=0.039).The median PFS were 14.9 and 5.3 months and median OS were 26.5 and 15.5 months for patients with and without deleterious g BRCA mutations,respectively(P=0.001,P=0.161,respectively).Patients in PBCT group had significantly greater rates of grade 3-4 anemia(5.5%vs.0%)and thrombocytopenia(8.8%vs.0%),whereas palmar-plantar erythrodysesthesia(12.4%vs.0%)and peripheral neuropathy(8.6%vs.1.1%)occurred more frequently in NPCT group.Conclusions:Platinum-based regimens are more effective in patients with deleterious g BRCA mutations,but no difference in patients without BRCA gene mutations,so non-platinum is an option in patients without BRCA gene mutations considering the toxicity and side effect.And we recommend that patients with advanced TNBC should have BR

Dzyaloshinskii-Moriya interaction(DMI)is the key ingredient of chiral spintronic phenomena and the emerging technologies based on such phenomena.A nonzero DMI usually occurs at magnetic interfaces or within non-centrosymmetric single crystals.Here,we report the observation of a strong unexpected DMI within a centrosymmetric polycrystalline ferromagnet that has neither a crystal inversion symmetry breaking nor a composition gradient.This DMI is a bulk effect,increases with the thickness of the magnetic layer,and is insensitive to the symmetry of the interfaces or the neighboring materials.We observe a total DMI strength that is a factor of>2 greater than the highest interfacial DMI in the literature.This DMI most likely arises from the strong spin-orbit coupling,strong orbital hybrization,and a“hidden”long-range asymmetry in the material.Our discovery of the strong unconventional bulk DMI in centrosymmetric,composition-uniform magnetic single layers provides fundamental building blocks for the emerging field of spintronics and will stimulate the exploitation of unconventional spin-orbit phenomena in a wide range of materials.

Ecotoxicity assessment of soil irrigated with domestic wastewater using different extractions

The toxicity of soil irrigated with treated domestic wastewater （site A） and untreated gray wastewater （site B） were investigated. Soil extracts were prepared using distilled water, acid solvent （0.1 mol·L-1 HCl）, and organic solvent （acetone：petroleum ether：cyclohexane= 1：1：1） to understand the type of pollutants responsible for the ecotoxicity associated with wastewater irrigation. The soil toxicity was assessed using a luminescence inhibition assay with Vibrio fischeri for acute toxicity, a micronucleus assay with Vicia faba root tips and a single cell gel electrophoresis assay of mice lymphocytes for genotoxicity. The physicochemical properties and the heavy metal （HM） contents of the irrigated soil were also analyzed. The results indicated that the wastewater irrigation at site A had no effects on the soil properties. With the exception of Pb, Zn, Fe, and Mn, the accumulation of HMs （Cu, Ni, and Cr） occurred. However, the irrigation at site A did not result in obvious acute toxicity or genotoxicity in the soil. The soil properties changed greatly, and HMs （Cu, Ni, and Cr） accumulated in site B. There were significant increases in the acute toxic and genotoxic effects in the soils from site B. The ecotoxicity in site B came primarily from organic-extractable pollutants.

Obesity-induced inflammation in white adipose tissue,characterized by increased macrophage infiltration and associated with macrophage population shift from anti-inflammatory M2 to proinflammatory M1 macrophages,largely contributes to obesity-induced insulin resistance and influences T2 DM pathogenesis.GSK3,a serine/threonine kinase,has been reported to participate in various cellular processes.We sought to examine the potential mechanism by which GSK3,a serine/threonine kinase implaicated in various cellular processes,modulates obesity-induced visceral adipose tissue(VAT) inflammation.Male C57 BL/6 J mice were fed a high-fat diet for 10 weeks while being treated with control or GSK3 inhibitors.RNA-Sequencing results using VAT demonstrated that GSK3 inhibitor treatment reversed obesity-specific expression of genes associated with inflammation.Consistently,GSK3 inhibition reduced M1 macrophages and increased M2 macrophages in VAT,while decreasing inflammatory monocytes.These anti-inflammatory effects of GSK3 inhibition were found to be driven,at least in part,by inhibiting production of apoptosis inhibitor of macrophage(AIM) in macrophages via inactivating STAT3 to reduce free fatty acid and chemokine level produced from VAT to suppress circulatory monocyte chemotaxis.Our findings suggest that GSK3 may act as an important regulator of obesity-induced inflammation and characterize the novel role of GSK3 in shifting macrophage polarization and reinforce its therapeutic potential for obesity-induced inflammation and its associated diabetes.
