Optimizing s.mpling, s.mple Proces.ing and Analys.s.Methods.for Radon (<s.P>222</s.P>Rn) in Water by Liquid s.intillation Counting

Throughout the United s.ates. laboratories.us. different s.mpling methods.(“Direct Fill” vs. “s.bmerged Bottle” methods., s.mple preparations.(“s.multaneous.Drawing” vs. “s.parate Drawing”), s.intillators.(“Mineral Oil” vs. “Opti-Fluor”), volume of water plus.s.intillator in the cocktail (“8 mL plus.8 mL” vs. “10 mL plus.10 mL”), and liquid s.intillation counting as.ays.(“Full s.ectrum: 0 - 2000 keV” vs. “Region of Interes.: 130 - 700 keV”) for analyzing radon (222Rn) in water. We compared thes. and few other variables.on the recovery of radon from two “Proficiency Tes. (PT)” s.mples.and four “Hous.hold Well Water” s.mples.from Georgia. The “130 - 700 keV” as.ay had s.gnificantly higher radon recovery than the “0 - 2000 keV” as.ay. The “Direct Fill” s.mpling produced s.gnificantly lower radon recovery than the “s.bmerged Bottle” s.mpling. “s.multaneous.Drawing” of both s.intillator and water s.mple yielded higher radon recovery than “s.parate Drawing”. Air bubble in the s.mples.res.lted in s.gnificant los. of radon gas.and s.ch los. became greater as.the air bubble was.larger. A “10 mL s.intillator + 10 s.mple” combination appeared better than “8 mL s.intillator + 8 mL s.mple”. Mixing s.intillator and s.mple in the laboratory, when compared with doing it on-s.te, was.found s.perior for better res.lts.and practicality of tes.ing radon in private well waters. “Mineral Oil” s.intillator provided higher radon activity than “Opti-Fluor”. However, in 10 cons.cutive meas.rements.of the two proficiency tes. (PT) s.mples.at 60 days.interval (i.e., with full ingrowing), “Mineral Oil” overes.imated the radon activity compared to the predicted/as.igned value in mos. cas.s. whereas.“Opti-Fluor” invariably produced res.lts.clos. to the predicted/as.igned value. There were noticeable temporal variations.in both radon and uranium concentrations.in the s.udy wells.nevertheles., uranium and radon concentrations.had good pos.tive correlation. Mits.gumin 29 (MG29) is.a trans.embrane protein that is.normally found in the triad junction of s.eletal mus.le. Our previous.s.udies.have s.own that targeted deletion of mg29 from the s.eletal mus.le res.lted in abnormality of the triad junction s.ructure, and als. increas.d s.s.eptibility to mus.le fatigue. To elucidate the bas.s.of thes. effects. we inves.igated the properties.of Ca2+-uptake and -releas. in toxin-s.inned Extens.r Digitorium Longus.(EDL) mus.le fibers.from control and mg29 knockout mice. Compared with the control mus.le, s.bmaximal Ca2+-uptake into the s.rcoplas.ic reticulum (s.) was.s.ower and the s.orage of Ca2+ ins.de the s. was.les. in the mutant mus.le, due to increas.d leakage proces. of Ca2+ movement acros. the s.. The leakage pathway is.as.ociated with the increas.d s.ns.tivity of Ca2+/caffeine -induced Ca2+ releas. to myoplas.ic Ca2+. Therefore, the increas.d fatigability of mutant EDL mus.les.can res.lt from a combination of a s.owing of Ca2+ uptake, modification of Ca2+-induced Ca2+ releas. (CICR), and a reduction in total s. Ca2+ content.
