Plasma phospholipid transfer protein (PLTP) as an emerging determinant of the adaptive immune response

Plasma phospholipid transfer protein(PLTP)is a multifunctional lipid transporter.In addition to phospholipids,PLTP transports various amphipathic compounds,such as cholesterol,alphatocopherol,diacylglycerides,and lipopolysaccharides.In addition to its impact on lipid metabolism and atherosclerosis development,PLTP has recently been reported to modulate inflammation and immune responses.It modulates the phagocytic activity of macrophages and microglial cells1,2 and increases the production of the pro-inflammatory cytokine interleukin 6(IL-6).3–5 In further support of the pro-inflammatory effect of PLTP,the expression level of IL-6 and the number of infiltrating macrophages in aortic tissue after experimental aneurysm induction were shown to be lower in PLTP-deficient mice than in wild-type mice;6 additionally,recombinant PLTP was shown to exert a direct pro-inflammatory effect on rheumatoid arthritis fibroblast-like synoviocytes,independent of its lipid transfer activity.7 Notably,despite its direct pro-inflammatory actions,PLTP can exert indirect antiinflammatory actions through its ability to transport and detoxify lipopolysaccharides in gram-negative bacteria.8,9 In humans,the majority of PLTP-associated proteins are involved in innate immunity and/or the inflammatory response in plasma,and PLTP activity is significantly increased in patients with bacterial infections and systemic inflammation.In addition to the key roles of PLTP in innate immunity,a recent publication from our group entitled“Plasma phospholipid transfer protein(PLTP)modulates adaptive immune functions through alternation of T-helper-cell polarization”10 demonstrated for the first time that PLTP can also modulate adaptive immune responses.This modulation occurred through a profound effect on CD4+T-cell polarization toward the pro-inflammatory Th1 subtype.This novel property also supports the pro-inflammatory and pro-atherogenic effects of PLTP(Fig.1).

A high-power electromagnetic source for disabling improvised explosive devices

Ultra-wideband(UWB)microwave sources driven by specialised pulsed power generators have experienced a considerable development in the last decade due to their wide domain of new applications such as defence or counter-terrorism activity.The authors present the main findings of a research dedicated to the development of a pulsed power-driven electromagnetic field source for disabling improvised explosive devices(IED).The pulsed power generator driving the source is a 13-stage compact Marx producing voltage pulses reaching an amplitude of 0.5 MV,with a pulse repetition frequency(PRF)of up to 100 Hz.The generator is coupled to a bipolar pulse forming line,providing bipolar pulses with a dV/dt of around 1.6 MV/ns.This pulsed power system feeds an array composed of 16 Koshelev-type UWB antennas through an impedance matching transformer.The resulting electromagnetic source is capable to produce pulsed electric fields(PEFs)having a figure-of-merit(FOM)of 1 MV.First,practical experiments were carried out to study the effects of the PEFs on targets.The targets used in the present study are M2B type flashbulbs,known to have the same susceptibility as the US army M6 detonator.Different configurations of wires(shielded,twisted,etc)with different lengths were used in connecting items inside these targets.The tests were performed by placing the flashbulbs at different distances to determine the essential parameters(i.e.,amplitude,duration,and frequency range)of the PEFs required to trigger them.An overview of the experimental campaign and the main findings are also presented followed by conclusions.

In healthy individuals,the intestinal epithelium forms a tight barrier to prevent gut bacteria from reaching blood circulation.To study the effect of probiotics,dietary compounds and drugs on gut barrier formation and disruption,human gut epithelial and bacterial cells can be cocultured in an in vitro model called the human microbial crosstalk(HuMiX)gut-on-a-chip system.Here,we present the design,fabrication and integration of thin-film electrodes into the HuMiX platform to measure transepithelial electrical resistance(TEER)as a direct readout on barrier tightness in realtime.As various aspects of the HuMiX platform have already been set in their design,such as multiple compressible layers,uneven surfaces and nontransparent materials,a novel fabrication method was developed whereby thin-film metal electrodes were first deposited on flexible substrates and sequentially integrated with the HuMiX system via a transfer-tape approach.Moreover,to measure localized TEER along the cell culture chamber,we integrated multiple electrodes that were connected to an impedance analyzer via a multiplexer.We further developed a dynamic normalization method because the active measurement area depends on the measured TEER levels.The fabrication process and system setup can be applicable to other barrier-on-chip systems.As a proof-of-concept,we measured the barrier formation of a cancerous Caco-2 cell line in real-time,which was mapped at four spatially separated positions along the HuMiX culture area.

Generation,bonding and reactivity of transient zinc-substituted silylenes

The metal-substituted silylenes are of high interest,as the theoretical studies indicated that the silylenes with electropositive substituents have a small ΔE_(S-T)(singlet-triplet energy gap)or even the ground-state triplets.However,such compounds are highly unstable,and only two transient alkali metal-substituted silylenes M(^(t)Bu_(3)Si)Si:(M=Li,K)were generated by photoextrusion of the alkali metal-substituted silacyclopropenes and merely studied by spectroscopic method(EPR)at low temperature(14 to 50 K).Herein,we report the generation of transient zinc-substituted silylenes from zinc silacyclopropanyl complexes under very mild and convenient conditions.The generated transient zinc-substituted silylenes are highly reactive and undergo intermolecular cycloaddition with alkenes for the synthesis of zinc-substituted Si-heterocyclic compounds.If there is no substrate,the zinc-substituted silylenes attack the C-C bonds of the β-diketiminato ligands and break the C-C bonds.DFT studies further highlight the silylene nature of the zinc-substituted silylene and a very small ΔE_(S-T)(4.4 kcal/mol).

The sleep-wake cycle stands as an integrative process essential for sustaining optimal brain function and,either directly or indirectly,overall body health,encompassing metabolic and cardiovascular well-being.Given the heightened metabolic activity of the brain,there exists a considerable demand for nutrients in comparison to other organs.Among these,the branched-chain amino acids,comprising leucine,isoleucine,and valine,display distinctive significance,from their contribution to protein structure to their involvement in overall metabolism,especially in cerebral processes.Among the first amino acids that are released into circulation post-food intake,branched-chain amino acids assume a pivotal role in the regulation of protein synthesis,modulating insulin secretion and the amino acid sensing pathway of target of rapamycin.Branched-chain amino acids are key players in influencing the brain's uptake of monoamine precursors,competing for a shared transporter.Beyond their involvement in protein synthesis,these amino acids contribute to the metabolic cycles ofγ-aminobutyric acid and glutamate,as well as energy metabolism.Notably,they impact GABAergic neurons and the excitation/inhibition balance.The rhythmicity of branchedchain amino acids in plasma concentrations,observed over a 24-hour cycle and conserved in rodent models,is under circadian clock control.The mechanisms underlying those rhythms and the physiological consequences of their disruption are not fully understood.Disturbed sleep,obesity,diabetes,and cardiovascular diseases can elevate branched-chain amino acid concentrations or modify their oscillatory dynamics.The mechanisms driving these effects are currently the focal point of ongoing research efforts,since normalizing branched-chain amino acid levels has the ability to alleviate the severity of these pathologies.In this context,the Drosophila model,though underutilized,holds promise in shedding new light on these mechanisms.Initial findings indicate its potential to introduce novel concepts,particularly

Background Comprehensive quantitative evidence on the risk and protective factors for sudden infant death syndrome(SIDS)effects is lacking.We investigated the risk and protective factors related to SIDS.Methods We conducted an umbrella review of meta-analyses of observational and interventional studies assessing SIDS-related factors.PubMed/MEDLINE,Embase,EBSCO,and Google Scholar were searched from inception until January 18,2023.Data extraction,quality assessment,and certainty of evidence were assessed by using A Measurement Tool Assessment Systematic Reviews 2 following PRISMA guidelines.According to observational evidence,credibility was graded and classified by class and quality of evidence(CE;convincing,highly suggestive,suggestive,weak,or not significant).Our study protocol was registered with PROSPERO(CRD42023458696).The risk and protective factors related to SIDS are presented as equivalent odds ratios(eORs).Results We identified eight original meta-analyses,including 152 original articles,covering 12 unique risk and protective factors for SIDS across 21 countries/regions and five continents.Several risk factors,including prenatal drug exposure[eOR=7.84(95%CI=4.81–12.79),CE=highly suggestive],prenatal opioid exposure[9.55(95%CI=4.87–18.72),CE=suggestive],prenatal methadone exposure[9.52(95%CI=3.34–27.10),CE=weak],prenatal cocaine exposure[4.38(95%CI=1.95–9.86),CE=weak],prenatal maternal smoking[2.25(95%CI=1.95–2.60),CE=highly suggestive],postnatal maternal smoking[1.97(95%CI=1.75–2.22),CE=weak],bed sharing[2.89(95%CI=1.81–4.60),CE=weak],and infants found with heads covered by bedclothes after last sleep[11.01(95%CI=5.40–22.45),CE=suggestive],were identified.On the other hand,three protective factors,namely,breastfeeding[0.57(95%CI=0.39–0.83),CE=non-significant],supine sleeping position[0.48(95%CI=0.37–0.63),CE=suggestive],and pacifier use[0.44(95%CI=0.30–0.65),CE=weak],were also identified.Conclusions Based on the evidence,we propose several risk and protective factors for SIDS.This study suggests the need

Background:Adenosine triphosphatase inhibitory factor 1(IF1)is a key protein involved in energy metabolism.IF1 has been linked to various agerelated diseases,although its relationship with physical activity(PA)remains unclear.Additionally,the apolipoprotein A-I(apoA-I),a PA-modulated lipoprotein,could play a role in this relationship because it shares a binding site with IF1 on the cell-surface ATP synthase.We examined here the associations between chronic PA and plasma IF1 concentrations among older adults,and we investigated whether apoA-I mediated these associations.Methods:In the present work,1096 healthy adults(63.8%females)aged 70 years and over who were involved in the Multidomain Alzheimer Prevention Trial study were included.IF1 plasma concentrations(square root of ng/mL)were measured at the 1-year visit of the Multidomain Alzheimer Prevention Trial,while PA levels(square root of metabolic equivalent task min/week)were assessed using questionnaires administered each year from baseline to the 3-year visit.Multiple linear regressions were performed to investigate the associations between the first-year mean PA levels and IF1 concentrations.Mediation analyses were conducted to examine whether apoA-I mediated these associations.Mixedeffect linear regressions were carried out to investigate whether the 1-year visit IF1 concentrations predicted subsequent changes in PA.Results:Multiple linear regressions indicated that first-year mean PA levels were positively associated with IF1 concentrations(B=0.021;SE=0.010;p=0.043).Mediation analyses revealed that about 37.7%of this relationship was mediated by apoA-I(B_(ab)=0.008;SE=0.004;p=0.023).Longitudinal investigations demonstrated that higher concentrations of IF1 at the 1-year visit predicted a faster decline in PA levels over the subsequent 2 years(time×IF1:B=0.148;SE=0.066;p=0.025).Conclusion:This study demonstrates that regular PA is associated with plasma IF1 concentrations,and it suggests that apoA-I partly mediates this association.Additionally,this study f

Objective： Plasma phospholipid transfer protein （PLTP） is a key determinant of lipoprotein metabolism, and both animal and human studies converge to indicate that PLTP promotes atherogenesis and its thromboembolic complications. Moreover, it has recently been reported that PLTP modulates inflammation and immune responses. Although earlier studies from our group demonstrated that PLTP can modify macrophage activation, the implication of PLTP in the modulation of T-cell-mediated immune responses has never been investigated and was therefore addressed in the present study. Approach and results： In the present study, we demonstrated that PLTP deficiency in mice has a profound effect on CD4＋ ThO cell polarization, with a shift towards the anti-inflammatory Th2 phenotype under both normal and pathological conditions. In a model of contact hypersensitivity, a significantly impaired response to skin sensitization with the hapten-2,4-dinitrofluorobenzene （DNFB） was observed in PLTP-deficient mice compared to wild-type （WT） mice. Interestingly, PLTP deficiency in mice exerted no effect on the counts of total white blood cells, lymphocytes, granulocytes, or monocytes in the peripheral blood. Moreover, PLTP deficiency did not modify the amounts of CD4＋ and CD8＋ T lymphocyte subsets. However, PLTP-deficiency, associated with upregulation of the Th2 phenotype, was accompanied by a significant decrease in the production of the pro-Thl cytokine interleukin 18 by accessory cells. Conclusions： For the first time, this work reports a physiological role for PLTP in the polarization of CD4＋ T cells toward the pro-inflammatory Th I phenotype.

The light curve analyses and orbital period variations for two contact binary stars,LS Del and V997 Cyg,are presented in this work which was conducted in the frame of the Binary Systems of South and North project Ground-based photometric observations were performed at two observatories in France.We used the Transiting Exoplanet Survey Satellite(TESS)data for extracting times of minima and light curve analysis of the targe systems.The O-C diagram for both systems displays a parabolic trend.LS Del and V997 Cyg’s orbital periods are increasing at rates of dP/dt=7.20×10^(-08)days yr^(-1)and dP/dt=2.54×10^(-08)days yr^(-1),respectively Therefore,it can be concluded that mass is being transferred from the less massive star to the more massive component with a rate of dM/dt=-1.96×10^(-7)M_(⊙)yr^(-1)for the LS Del system,and dM/dt=-3.83×10^(-7)M_(⊙)yr-1for V997 Cyg.The parameters of a third possible object in the system are also considered.The PHysics Of Eclipsing BinariEs Python code was used to analyze the light curves.The light curve solutions needed a cold starspot due to the asymmetry in the LS Del system’s light curve maxima.The mass ratio fill-out factor,and star temperature all indicate that both systems are contact binary types in this investigation.Two methods were applied to estimate the absolute parameters of the systems:one method relied on the parallax of Gaia DR3,and the other used a P-M relationship.The positions of the systems are also depicted on the M-L,M-R q-L_(ratio),and logM_(tot)-logJ_(0)diagrams.We recommend that further observations and investigations be done on the existence of a fourth body in this system.
