A novel approach based on metabolomics coupled with network pharmacology to explain the effect mechanisms of Danggui Buxue Tang in anaemia

Danggui Buxue Tang(DBT) is a famous Chinese medicinal decoction. Mechanism of DBT action is wide ranging and unclear. Exploring new ways of treatment with DBT is useful. Sprague–Dawley(SD) rats were randomly divided into 3 groups including control(NC, Saline), the DBT(at a dose of 8.10 g?kg–1), and blood deficiency(BD)(Cyclophosphamide(APH)-and Cyclophosphamide(CTX)-induced anaemia). A metabolomics approach using Liquid Chromatography-Quadrupole-Time-of-Flight/Mass Spectrometry(LC/Q-TOFMS) was developed to perform the plasma metabolic profiling analysis and differential metabolites were screened according to the multivariate statistical analysis comparing the NC and BD groups, and the hub metabolites were outliers with high scores of the centrality indices. Anaemia disease-related protein target and compound of DBT databases were constructed. The TCMSP, Chem Mapper and STITCH databases were used to predict the protein targets of DBT. Using the Cytoscape 3.2.1 to establish a phytochemical component–target protein interaction network and establish a component, protein and hub metabolite protein–protein interaction(PPI) network and merging the three PPI networks basing on BisoGenet. The gene enrichment analysis was used to analyse the relationship between proteins based on the relevant genetic similarity by ClueGO. The results shown DBT effectively treated anaemia in vivo. 11 metabolic pathways are involved in the therapeutic effect of DBT in vivo; S-adenosyl-L-methionine, glycine, L-cysteine, arachidonic acid(AA) and phosphatidylcholine(PC) were screened as hub metabolites in APH-and CTX-induced anaemia. A total of 288 targets were identified as major candidates for anaemia progression. The gene-set enrichment analysis revealed that the targets are involved in iron ion binding, haemopoiesis, reactive oxygen species production, inflammation and apoptosis. No association exists between E-cadherin gene polymorphism and tumor recurrence in patients with hepatocellular carcinoma after transplantation

BACKGROUND: E-cadherin is an epithelial cell adhesion molecule, and decreased E-cadherin expression in liver cancer is associated with poor prognosis. A -160 C -> A polymorphism in the promoter region of E-cadherin has been reported to decrease gene transcription. This allelic variation may be a potential genetic marker for identifying those individuals at higher risk for invasive/metastatic disease. METHODS: The effect of E-cadherin gene polymorphism on risk of tumor recurrence was studied in 93 patients with hepatocellular carcinoma (HCC) after liver transplantation, and determined whether this polymorphism is a biomarker for the risk of tumor recurrence. RESULTS: The genotype frequencies in the patients with recurrence were C/C: 0.667, C/A: 0.311, and A/A: 0.022, and in-the patients without recurrence C/C: 0.604, C/A: 0.271 and A/A: 0.125. No significant difference was found between the two groups (P = 0.171). Between -160 C -> A polymorphism and the clinicopathological data, there were no statistically significant differences in the distribution of the parameters as to age, gender, portal vein tumor thrombi, preoperative alpha-fetoprotein level, tumor size, or histopathological grading (P > 0.05). CONCLUSION: The results of this study show no association exists between the E-cadherin genotype and the risk of tumor recurrence in Chinese patients with HCC.

Low dosage kinetic hydrate inhibitors(KHIs)are a kind of alternative chemical additives to high dosage thermodynamic inhibitors for preventing gas hydrate formation in oil&gas production wells and transportation *** this paper,a new KHI,poly(N-vinyl caprolactam)-co-tert-butyl acrylate(PVCapco-TBA),was successfully synthesized with N-vinyl caprolactam(NVCap)and tert-butyl *** kinetic inhibition performances of PVCap-co-TBA on the formations of both structureⅠmethane hydrate and structureⅡnatural gas hydrate were investigated by measuring the onset times of hydrate formation under different conditions and compared with commercial KHIs such as PVP,PVCap and inhibex *** results indicated that PVCap-co-TBA outperformed these widely applied inhibitors for both structureⅠand structureⅡ*** the same dosage of KHI,the maximum tolerable degree of subcooling under which the onset time of hydrate formation exceeded 24 hours for structureⅠhydrate was much lower than that for structureⅡ*** inhibition strength increased with the increasing dosage of PVCap-co-TBA;The maximum tolerable degree of subcooling for the natural gas hydrate is more than10 K when the dosage was higher than 0.5%(mass)while it achieved 12 K when that dosage rose to0.75%(mass).Additionally,we found polypropylene glycol could be used as synergist at the dosage of 1.0%(mass)or so,under which the kinetic inhibition performance of PVCap-co-TBA could be improved *** evaluation results demonstrated that PVCap-co-TBA was a very promising KHI and a competitive alternative to the existing commercial KHIs.

Cost-Effectiveness Analysis of Combined Chemotherapy Regimen Containing Bedaquiline in the Treatment of Multidrug-Resistant Tuberculosis in China

Objective This study aims to estimate the cost-effectiveness of the combined chemotherapy regimen containing Bedaquiline(BR)and the conventional treatment regimen(CR,not containing Bedaquiline)for the treatment of adults with multidrug-resistant tuberculosis(MDR-TB)in *** A combination of a decision tree and a Markov model was developed to estimate the cost and effects of MDR patients in BR and CR within ten *** model parameter data were synthesized from the literature,the national TB surveillance information system,and consultation with *** incremental cost-effectiveness ratio(ICER)of BR *** was *** BR(***)had a higher sputum culture conversion rate and cure rate and prevented many premature deaths(decreased by 12.8%),thereby obtaining more quality-adjusted life years(QALYs)(increased by 2.31 years).The per capita cost in BR was as high as 138,000 yuan,roughly double that of *** ICER for BR was 33,700 yuan/QALY,which was lower than China's 1×per capita Gross Domestic Product(GDP)in 2020(72,400 yuan).Conclusion BR is shown to be cost *** the unit price of Bedaquiline reaches or falls below57.21 yuan per unit,BR is expected to be the dominant strategy in China over CR.

The high-amplitude δ Scuti variable CY Aqr is probably a triple system

Data representing 864 times of light maxima of the high-amplitude δ Scuti star CY Aqr were col- lected from the literature, based on which, long-term period changes of the variable star were investigated. A revised period and new ephemerides were given for the pulsating star. Remarkable cyclic variations were found in the O - C residuals which can be attributed to the light-time effects due to probable unseen com- ponents of the object. By using Kopal＇s method, the orbital parameters of the supposed component stars were derived. The solution suggests that CY Aqr is very probably in a triple system orbited eccentrically by two low-mass companions with periods of 54.2 and 47.3 yr. The lower limits on masses were estimated as 0.04 Me and 0.02 Me, respectively, for the two hidden companions.

MnO_(x)-CeO_(2) catalysts are developed by hydrolysis driving redox method using acetate precursor(3 Mn1 Ce-Ac) and nitrate precursor(3 Mn1 Ce-N) for the selective catalytic reduction(SCR) of NO_(x) by NH_(3).A counterpart sample(Cop-3 Mn1 Ce) was prepared by the NH_(3)·H_(2) O co-precipitation method for comparison *** the results of physicochemical properties characterization and performance test,we find that the 3 Mn1 Ce-Ac catalyst with some nanorod structures is highly active for the deNOx *** SCR activity of the 3 Mn1 Ce-Ac catalyst is more admirable than the 3 Mn1 Ce-N and the Cop-3 Mn1 Ce catalysts due to plentiful Lewis acid sites,excellent low-temperature reducibility,and superior surface area resulted from O_(2) generation during the pre paration *** 3 Mn1 Ce-Ac still exhibits the greatest performance for the deNO_(x )process when gaseous acetone is in the SCR feed *** NOx conversion and N2 selectivity over the 3 Mn1 Ce-Ac are both improved by gaseous acetone above150℃ due to the inhibition of SCR undesired side reactions(NSCR & C-O reactions) and "slow-SCR" process.
