Effects of soil salinity on rhizosphere soil microbes in transgenic Bt cotton fields

With increased cultivation of transgenic Bacillus thuringiensis （Bt） cotton in the saline alkaline soil of{2., assessments of transgenic crop biosafety have focused on the effects of soil salinity on rhizosphere microbes and Bt protein residues. In 2.13 and 2.14, investigations were conducted on the rhizosphere microbial biomass, soil enzyme activities and Bt protein contents of the soil under transgenic Bt cotton （variety GK19） and its parental non-transgenic cotton （Simian 3） cultivated at various salinity levels （1.15, 6.00 and 11.46 dS m-1）. Under soil salinity stress, trace amounts of Bt proteins were ob- served in the Bt cotton GK19 rhizosphere soil, although the protein content increased with cotton growth and increased soil salinity levels. The populations of slight halophilic bacteria, phosphate solubilizing bacteria, ammonifying bacteria, nitrifying bacteria and denitrifying bacteria decreased with increased soil salinity in the Bt and non-Bt cotton rhizosphere soil, and the microbial biomass carbon, microbial respiration and soil catalase, urease and alkaline phosphatase activity also decreased. Correlation analyses showed that the increased Bt protein content in the Bt cotton rhizosphere soil may have been caused by the slower decomposition of soil microorganisms, which suggests that salinity was the main factor influencing the relevant activities of the soil microorganisms and indicates that Bt proteins had no clear adverse effects on the soil microorganisms. The results of this study may provide a theoretical basis for risk assessments of genetically modified cotton in saline alkaline soil.

Aphis gossypfi is a globally distributed species and therefore has a highly variable life cycle. Populations of A. gossypii in northern{1. exhibit greater genotypic diversity and a broader host range, yet the details of life cycles of different biotypes is still unclear. In this study, the Cytb and 1.S gene regions of A. gossypfi collected from 5 common summer hosts and 4 primary hosts were analyzed. A total of 57 haplotypes were obtained from 1.046 individual A. gossyp# sequences. The sequence included 44 variable sites, 2. of which were parsimony informative sites and 1. of which were singleton variable sites. The most frequent 3 haplotypes were found in 896 individuals, representing a total of 85.7% of all individuals and 36 haplotypes were found in 1.individual. A neighbor-joining tree was constructed using 2.1}haplotypes that were found in more than 2.individuals. Considering the individual host plant, 5 biotypes were identified. Type 1.corresponded exactly to the cucurbit host-race and the other 4 biotypes were found as cotton host-races. Type 3 was the most abundant biotype in cotton fields in northern{1..

BACKGROUND： Differentiation of liver progenitor cells（LPCs） to functional hepatocytes holds great potential to develop new strategies for hepatocyte transplantation and the screening of drug-induced cytotoxicity. However, reports on the efficient and convenient hepatic differentiation of LPCs to hepatocytes are few. The present study aims to investigate the possibility of generating functional hepatocytes from LPCs in an indirect co-culture ***： Mouse LPCs were co-cultured in Transwell plates with an immortalized human hepatic stellate cell line（HSCli） we previously established. The morphology, expression of hepatic markers, and functions of mouse LPC-derived cells were monitored and compared with those of conventionally cultured LPCs. RESULTS： Co-culturing with HSC-li cells induced differentiation of mouse LPCs into functional hepatocyte-like cells. The differentiated cells were morphologically transformed into hepatocyte-like cells 3 days after co-culture initiation. In addition, the differentiated cells expressed liver-specific genes and possessed hepatic functions, including glycogen storage, lowdensity lipoprotein uptake, albu{12. secretion, urea synthesis, and cytochrome P450 1A2.enzymatic ***： Our method, which employs indirect co-culture with HSC-li cells, can efficiently induce the differentiation of LPCs into functional hepatocytes. This finding suggests that this co-culture system can be a useful method for the efficient generation of functional hepatocytes from LPCs.

背景与目的:Peutz-Jeghers综合征(Peutz-Jegherssyndrome,PJS)是一种常染色体显性遗传性疾病。脆性组氨酸三连体(fragilehistidinetriad,FHIT)基因是定位于3p1..2.性位点区域的重要抑癌基因。我们以往的研究结果认为PJS患者在3p1..2.域可能存在易感基因。本文旨在明确PJS患者FHIT基因突变与癌变的关系和FHIT基因的改变及规律。方法:应用变性高效液相色谱(DHPLC)分析、聚合酶链反应-单链构象多肽分析技术(PCR-SSCP)和DNA测序方法,对6个PJS家系中的1.个患者及其2.例正常家族成员的FHIT基因进行了研究。结果:在6个家系中,有1.患者的FHIT基因第1.9位核苷酸由G→T,相应的第54位密码子编码的氨基酸由谷氨酸变成终止密码子;在第62.密码子处存在碱基G插入,使读码框架发生移位,在第1.1.密码子处提前出现终止密码子;在2.癌变患者的息肉标本DNA和癌标本DNA中检测到FHIT基因第8外显子的纯合性缺失;在3个散发的PJS患者中发现,患者与其母亲在第8外显子,有相同的SSCP带型和DHPLC峰型,DNA测序结果显示,在第98位密码子处发生同义突变,由CAT→CAC,相应编码的氨基酸未发生改变,均为苏氨酸。另外,有7例患者和2.正常人在FHIT基因的第6内含子5'端第42.核苷酸位点发生由A→G的点突变。

Gonadotropin-releasing hormone （GnRH） is secreted from neurons within the hypothalamus and is necessary for reproductive function in all vertebrates. GnRH is also found in organs outside of the brain and plays an important role in Leydig cell steroidogenesis in the testis. However, the signalling pathways mediating this function remain largely unknown. In this study, we investigated whether components of the mitogen-activated protein kinase （MAPK） pathways are involved in GnRH agonist （GnRHa）-induced testis steroidogenesis in rat Leydig cells. Primary cultures of rat Leydig cells were established. The expression of 3β-hydroxysteroid dehydrogenase （3β-HSD） and the production of testosterone in response to GnRHa were exa{2.ed at different doses and for different durations by RT-PCR, Western blot analysis and radioimmunoassay （RIA）. The effects of GnRHa on ERK1.2. JNK and p38 kinase activation were also investigated in the presence or absence of the MAPK inhibitor PD-98059 by Western blot analysis. GnRHa induced testosterone production and upregulated 3β-HSD expression at both the mRNA and protein levels; it also activated ERK1.2. but not JNK and p38 kinase. Although the maximum effects of GnRHa were observed at a concentration of 1.0 nmnol L-1.after 2. h, activation of ERKI/2.by GnRHa reached peak at 5 {2. and it returned to the basal level within 60 {2.. PD-98059 completely blocked the activation of ERKI/2. the upregulation of 3β-HSD and testosterone production. Our data show that GnRH positively regulates steroidogenesis via ERK signalling in rat Leydig cells. ERK1.2.activation by GnRH may be responsible for the induction of 3β-HSDgene expression and enzyme production, which may ultimately modulate steroidogenesis in rat Leydig cells.
