Stripper gas and terminal potential play a key role for the charge state distribution in a tandem pelletron accelerator. The knowledge of this distribution is important for experiments performed on tandem accelerators. The charge state distribution of B, C, Si, Ni, Cu and Au beams is measured by using Ar as stripper gas, and terminal potential is varied from 0.3 to 3.0 MV on 5UDH-2 tandem pelletron accelerator installed at the National Centre for Physics, Islamabad. The individual charge state is measured after the switching magnet at 15° in high-energy portion. It is observed that the higher charge states are stable in the range of lower and middle atomic masses of periodic table, whereas higher atomic mass(Au) shows beam current instability in higher charge states. For carbon,the charge distribution at 1.7 MV terminal potential by varying stripper gas pressure is also studied, which resulted in decreased overall transmission with good current value for higher charge states.

Background: Cytoplasmic male sterility in flowering plants is a convenient way to use heterosis via hybrid breeding and may be restored by nuclear restorer-of-fertility(Rf) genes. In most cases, Rf genes encoded pentatricopeptide repeat(PPR) proteins and several Rf genes are present in clusters of similar Rf-PPR-like(RFL) genes. However, the Rf genes in cotton were not fully characterized until ***: In total, 35 RFL genes were identified in G. hirsutum, 16 in G. arboreum, and 24 in G. raimondii. Additionally,four RFL-rich regions were identified; the RFL-rich region in Gh05 is the probable location of Rf-PPR genes in cotton and will be studied further in the future. Furthermore, an insertion sequence was identified in the promoter sequence of Gh05 G3392 gene in the restorer line, as compared with the CMS-D2 line and maintainer lines. An InDel-R marker was then developed and could be used to distinguish the restorer line carrying Rfl from other genotypes without the Rf1 ***: In this study, genome-wide identification and analysis of RFL genes have identified the candidate Rf-PPR genes for CMS in Gossypium. The identification and analysis of RFL genes and sequence variation analysis will be useful for cloning Rf genes in the future and also for three-line hybrid breeding in cotton.

The growing need for sustainable energy solutions,driven by rising energy shortages,environmental concerns,and the depletion of conventional energy sources,has led to a significant focus on renewable *** energy,among the various renewable sources,is particularly appealing due to its abundant ***,the efficiency of commercial solar photovoltaic(PV)modules is hindered by several factors,notably their conversion efficiency,which averages around 19%.This efficiency can further decline to 10%–16%due to temperature increases during peak sunlight *** study investigates the cooling of PV modules by applying water to their front surface through Computational fluid dynamics(CFD).The study aimed to determine the optimal conditions for cooling the PV module by analyzing the interplay between water film thickness,Reynolds number,and their effects on temperature reduction and heat *** CFD analysis revealed that the most effective cooling condition occurred with a 5 mm thick water film and a Reynolds number of *** specific parameters were found to maximize the heat transfer and temperature reduction *** finding is crucial for the development of practical and efficient cooling systems for PV modules,potentially leading to improved performance and longevity of solar *** cooling fluids or advanced cooling techniques that might offer even better efficiency or practical benefits.

近年来,随着人工制种成本的逐年提升,细胞质雄性不育(Cytoplasmic male sterility,CMS)已经逐渐成为棉花杂种优势利用领域研究的热点。利用哈克尼西棉(Gossypium harknessii)细胞质雄性不育系(CMS-D2)的"三系(不育系,恢复系和保持系)"配套制种不仅可以省去人工去雄,大面积制种时省工省时、操作简便,而且可有效提高杂交制种纯度。然而,棉花CMS-D2恢复系育性恢复能力易受高温胁迫影响,从而限制了棉花"三系"杂交种的推广应用。DNA甲基化(DNA methylation)是1种主要的表观遗传修饰形式,是调节基因功能的重要手段之一。DNA甲基化在真核生物中主要以5-甲基胞嘧啶的形式存在。近年来的研究结果表明,逆境如热胁迫、盐胁迫、干旱影响DNA的甲基化水平,且许多受甲基化变化诱导的基因同胁迫反应有关,说明DNA甲基化参与了环境胁迫下的基因表达调控过程。本研究所用材料为课题组多年构建的棉花CMS-D2高代恢复基因近等基因系材料(保持系ZB,耐高温材料;近等恢复系ZBR,温敏感材料),分别在适温和高温条件下进行花药取样。为了更好地解析二者花药发育过程中响应高温胁迫的表观修饰差异,我们利用全基因组甲基化测序(Whole genome bisulfite sequencing,WGBS)技术,绘制了棉花花药单碱基分辨率甲基化图谱,同时分析了棉花花药发育过程中响应高温胁迫的表观基因组修饰。以适温条件下的保持系ZB为例,研究表明棉花花药基因组在CG、CHG和CHH(H=A或T或C)序列背景下分别表现出68.7%、61.8%和21.8%的甲基化水平。我们还进一步统计分析了mCG、mCHG和mCHH占总的mC的比例,并且发现高温胁迫会引起CG和CHG甲基化水平升高,相反CHH甲基化水平则会降低;胞嘧啶DNA甲基化还具有序列偏好性。此外,为了试图将高温胁迫下的DNA甲基化与基因表达变化联系起来,我们对相同的材料进行转录组测序,首先对比分析了不同表达水平基因的基因体(Gene b o dy)以及上下游2 kb区域在C G、CHG和CHH(H=A或T或C)序列环境下的甲基化水平。基于此,我们还分别依据启动子和基因体区域的甲基化水平对基因进行分类,深入评估DNA甲基化与基因表达变化二者之间的调控关系,结果表明启动子区不发生甲基化的基因比启动子区甲基化的基因显示出更高的表达水平,然而基因体区域甲基化程度似乎与基因表达水平呈现正相关。进一步对差异甲基化相关基因进行KEGG(Kyoto encyclopedia of genes and genomes)富集分析我们发现,高度甲基化基因主要涉及代谢途径、氧化磷酸化、次生代谢生物合成、三羧酸循环等代谢过程;而去甲基化基因主要参与次生代谢生物合成、二萜类生物合成等代谢通路,这说明棉花花药发育过程中响应高温胁迫发生了广泛的DNA甲基化改变。本研究提供了棉花花药基因组的甲基化图谱,较为全面地分析了棉花花药发育过程中响应高温胁迫的DNA甲基化与基因表达变化之间的潜在联系,理论上可加深对棉花CMS-D2及育性恢复表观调控机制的认识,同时为生产上解决恢复系育种瓶颈和培育高产、优质、高抗"三系"杂交棉品种提供理论指导。

The particle structure of a complex system has been explored through a unique Evans' s homogenous nonequilibrium molecular dynamics(HNEMD) simulation technique. The crystalline order–disorder structures(OD-structures) and the corresponding energies of three-dimensional(3 D) nonideal complex systems(NICSs) have been measured over a wide range of plasma states(■, κ) for a body-centered cubic(BCC) structure. The projected technique provides accurate ODstructures with fast convergence and applicable to very small size effect for different temperatures(≡ 1/■) and constant force field(F~*) values. The OD-structure obtained through HNEMD approach is found to be reasonable agreement and more reliable than those earlier identified by simulation approaches and experimental data of NICSs. New simulations of OD-structures show that dusty plasma remains in crystalline(strongly coupled) state at lower temperature and constant F*values, for the whole simulation runs. Our investigations show that the crystalline structure is changed and the particle structure switches from intermediate to disorder(nonideal gaseous) state with an increase of the system's temperature. It has been shown that the long range order shifts toward lower temperature with increasing κ. The presented technique exhibits that the potential energy has a maximum value when the dusty plasma remains in crystalline states(low temperatures),which confirms earlier 3 D simulation results.
